采用聚丙烯酰胺凝胶电泳法对21条藏獒的红细胞乳酸脱氢酶(RBC-LDH)和血清到脱氢酶(S-LDH)同工酶酶谱及其多态性进行研究。结果发现:(1)RBC-LDH同工酶共有3条区带,呈现两种电泳表型,11条藏契为LDH2A型(57.89%),8条藏獒为LDH2a(42.11%),(2)S-LDH共有5条区带,呈现两种电泳表型,2条藏獒为LDH1A(9.52%),19条藏獒为LDH1a(90.48%)  相似文献

AIM: To make hyperglycemia models and observe the effects of hyperglycemia on expressive type transformation of vascular smooth muscle cell (VSMC) in rats. METHODS: Using tissue culture and radioactivity analysis methods.RESULTS:①Hyperglycemia group was lower than control group in NO2- content, nitric oxide synthase activity and cGMP content;②Hyperglycemia group was higher than control group in endothelin, insulin, total chdesterol and [3H]-TdR penetrated rate of VSMC. CONCLUSION: It was possible that hyperglycemia reduces the expressive type of VSMC to change and promotes VSMC proliferation.  相似文献

采用大样本取样野外实地测定方法,对丹东地区雨久花种群生殖株的数量性状和生殖分配规律进行了分析。结果表明,在开花期,雨久花种群单个生殖株植株高度和花序长分别为(91.59±7.96)cm和(11.10±2.46)cm,花数为(68.47±18.52)个;生殖株生物量、根生物量、茎生物量、叶生物量和花序生物量分别为(9.97±4.13)g、(1.21±0.66)g、(6.36±2.91)g、(1.35±0.76)g和(1.05±0.51)g。花期雨久花种群生殖株变异系数以叶生物量最大,为56.41%;其次为根生物量54.14%;株高最小,为4.87%,各数量性状具有较大的表型可塑性。雨久花种群生殖株表型可塑性遵循着规律性,即随着株高的增加,花序长和花数量呈幂函数增加,根生物量和花序生物量呈直线增加;花序生物量与总生物量之间不存在相关的函数关系。  相似文献

[目的]比较不同表现类型平贝母的有效成分总生物碱的含量差别,为育种提供依据。[方法]采用超声法提取法提取平贝母中的总生物碱,并采用比色法测定其含量。[结果]平贝母常见的绿茎绿叶品系的总生物碱含量为0.170 3%,紫茎紫叶和紫茎绿叶分别为0.180 7%和0.183 0%。[结论]不同表现类型平贝母在总生物碱含量上没在明显差异。  相似文献

AIM: To investigate the genetic cause of 2 Chinese families with Marfan syndrome. METHODS: The clinical and laboratory investigations were performed in the 2 unrelated Chinese families. Family 1 had 1 patient with cardiac problem. Family 2 had 2 patients:one died, and the other with respiratory and cardiac problems. Next generation sequencing and Sanger sequencing in the Marfan syndrome causal gene FBN1 were performed in the patient, his unaffected sister and the parents of family 1. Sanger sequencing covering all the exons and intron-exon boundaries were performed in the patient and the parents in family 2. Bioinformatic analysis was engaged in the variations unravelled. Fifty healthy individuals were also investigated in the same manner. RESULTS: Both patients were diagnosed with Marfan syndrome. A novel mutation c.4685G>A(p.Cys1562Tyr) was detected in the patient of family 1 but was absent in his parents and the unaffected sister. This is a previously unreported novel mutation. In the mutation a conserved Cys was substituted by a Tyr in amino acid 1562 affecting a TGF-β binding domain and the secondary structure in the encoded protein. We also detected the mutation c.3706T>C(p.Cys1236Arg) in the patient of family 2. It was absent in the unaffected parents, and therefore was a de novo mutation too. This mutation has been previously reported and known to be associated with neonatal Marfan syndrome. Both mutations were absent in the 50 healthy controls. We also compared the genotype and phenotypes of the 2 families. CONCLUSION: We report 2 de novo mutations in 2 Chinese families with Marfan syndrome. One of the 2 mutations is novel. The phenotype of the mutation c.4685G>A(p.Cys1562Tyr) in family 1 is associated with classical Marfan syndrome, while that of c.3706T>C(p.Cys1236Arg) in family 2 is with neonatal type of Marfan syndrome. De novo mutations may be a cause for a proportion of mutations underlying the disease. The novel mutation also expends the mutational spectrum of the FBN1 gene.  相似文献

AIM: To investigate the effect of cyclopamine on Hedgehog (HH) signaling, phenotypic transformation and matrix accumulation induced by aristolochic acid (AA) in renal tubular epithelial cell NRK-52E. METHODS: NRK-52E cells were randomly divided into control group (treated with solvent only), AA group (treated with AA at concentrations of 1, 5, 10 mg/L) and cyclopamine group (treated with AA at concentration of 10 mg/L plus cyclopamine at concentrations of 1, 5, 10 μmol/L). After cultured for 24 h, the mRNA expression of Ptch1, Smo, α-SMA, E-cadherin, ZO-1, BMP-7, type I collagen and type III collagen was quantified by real-time PCR. The protein levels of Shh and TGF-β1 were detected by ELISA. Immunofluorescence staining was used to evaluate the expression of Ptch1, Smo, α-SMA, E-cadherin and type III collagen in the NRK-52E cells. RESULTS: AA increased the expression of TGF-β1, α-SMA and type III collagen, decreased the expression of E-cadherin and ZO-1 protein, and down-regulated the expression of Ptch1, Shh and Smo mRNA in the NRK-52E cells, indicating that AA activated HH signaling, and phenotypic transformation and matrix accumulation occurred in AA-treated NRK-52E cells. Treatment with cyclopamine inhibited HH signaling by decreasing Smo expression and increasing Ptch1 expression. Moreover, cyclopamine also down-regulated the expression of TGF-β1, α-SMA, type I collagen and III collagen, and up-regulated the expression of BMP-7, ZO-1 and E-cadherin. CONCLUSION: AA induces phenotypic transformation and matrix accumulation in renal tubular epithelial cells, which can be inhibited by cyclopamine treatment. The possible mechanism is that cyclopamine suppresses the activation of HH signaling, resulting in the reduction of epithelial-to-mesenchymal transition and matrix deposition.  相似文献

将不同表型和数目的精子注射到兔卵胞质或卵周隙中,结果表明,精子的表型对精子注射有影响。卵胞质中注射单个已获能死精子的受精率和发育率分别为53.26%和39.13%,注射单个未获能死精子的受精率和发育率为43.21%和24.69%,而注射单个未获能精子头核的受精率和发育率为18.18%和11.04%。前二者之间没有显著的差异。但与后者比较,受精率差异极显著,发育率差异显著。卵周隙中注射1~4个已获能运动精子的受精率和发育率均为23.08%,注射1~4个未获能运动精于的受精率和发育率为29.03%和25.81%,而注射1~4个已获能不运动精子的受精率和发育率均为5%。前二者差异不显著,但与后者比较,差异极显著。  相似文献

对2003年在全省范围内所选的黧蒴优树进行表型分析,着重研究了其叶、果的性状。结果表明:多个角度证明了5株优势木法选黧蒴优树的方法是合理的,大部分数量指标完全超过5株优势木,体现优中选优的特点。方差分析结果表明:黧蒴优树的叶果性状无论从家系间或种源间均呈极显著差异。动态聚类分析显示黧蒴优树叶果有3种类型,第1类为重果圆叶型,有8个家系;第3类为轻果尖叶型,有90个家系;第2类介于第1、3类之间,有69个家系。  相似文献

以陆地棉冀无2031无菌苗下胚轴为外植体,在MS中附加激素0.1mg/LIAA 0.1mg/L2,4-D 0.1mg/LKT的培养基上诱导出愈伤组织。选择疏松愈伤组织继代到附加0.1mg/LZT的培养基中,经2-3个月的继代培养诱导出颗粒状疏松胚性愈伤组织。胚性愈伤组织在无激素、每升加2g谷氨酰胺和0.5g天门冬酰胺的胚性愈伤增殖萌发培养基上培养2个月左右,可陆续形成不同时期的体胚苗。对冀无2031不同时期的体细胞萌发后的体胚苗进行观察,发现了真叶畸形苗、子叶节生根胚、胚轴侧生根胚、腋芽丛生苗和以腋芽发育为主的再生苗5种新类型的体胚苗。  相似文献
