4-香豆酸辅酶A连接酶(4-coumarate:coenzyme A ligase,4CL)催化各种羟基肉桂酸生成相应的硫酯,从而调控木质素和黄酮类物质的生成.4CL的芥子酸催化活性一直是备受关注的一个问题,目前只有拟南芥(Arabidopsis thaliana)的At4CL4和大豆(Glycine max)的Gm4CL1具有芥子酸转化能力.本研究用水稻(Oryza sativa subsp.japonica)苗期的叶片为材料,提取总RNA并以此为模板,用反转录PCR扩增水稻的Os4CL5基因,并将其连接到原核表达载体pET22-b(+)上,构建了原核表达载体pET-4CL5(+Val).通过定点突变的方法删除了水稻Os4CL5的Val337,构建了原核表达载体pET-4CL5(-Val).重组质粒转化大肠杆菌(Escherichia coli)BL21(ED3)并经异丙基-β-d-硫代半乳糖苷(isopropyl-β-d-thiogalactoside,IPTG)诱导表达,十二烷基硫酸钠聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis,SDS-PAGE)检测表明,融合蛋白的分子量为57.0 kD,与预测值一致.His-SpinTrap蛋白纯化系统纯化回收重组蛋白,然后对重组蛋白的酶学性质进行表征.结果表明,野生型Os4CL5对不同底物表现出了不同的催化效率,香豆酸是最适底物,其次是阿魏酸,再次是咖啡酸,对芥子酸没有活性.与野生型Os4CL5相比,突变型Os4CL5对香豆酸、咖啡酸和阿魏酸的催化效率有所提高,并且其底物特异性发生了明显改变,获得了对芥子酸的催化活性.本研究为利用基因工程手段调控木质素的生物合成提供了理论依据.  相似文献

For spatial crop and agro-systems modelling, there is often a discrepancy between the scale of measured driving data and the target resolution. Spatial data aggregation is often necessary, which can introduce additional uncertainty into the simulation results. Previous studies have shown that climate data aggregation has little effect on simulation of phenological stages, but effects on net primary production (NPP) might still be expected through changing the length of the growing season and the period of grain filling. This study investigates the impact of spatial climate data aggregation on NPP simulation results, applying eleven different models for the same study region (∼34,000 km2), situated in Western Germany. To isolate effects of climate, soil data and management were assumed to be constant over the entire study area and over the entire study period of 29 years. Two crops, winter wheat and silage maize, were tested as monocultures. Compared to the impact of climate data aggregation on yield, the effect on NPP is in a similar range, but is slightly lower, with only small impacts on averages over the entire simulation period and study region. Maximum differences between the five scales in the range of 1–100 km grid cells show changes of 0.4–7.8% and 0.0–4.8% for wheat and maize, respectively, whereas the simulated potential NPP averages of the models show a wide range (1.9–4.2 g C m−2 d−1 and 2.7–6.1 g C m−2 d−1 for wheat and maize, respectively). The impact of the spatial aggregation was also tested for shorter time periods, to see if impacts over shorter periods attenuate over longer periods. The results show larger impacts for single years (up to 9.4% for wheat and up to 13.6% for maize). An analysis of extreme weather conditions shows an aggregation effect in vulnerability up to 12.8% and 15.5% between the different resolutions for wheat and maize, respectively. Simulations of NPP averages over larger areas (e.g. regional scale) and longer time periods (several years) are relatively insensitive to climate data aggregation. However, the scale of climate data is more relevant for impacts on annual averages of NPP or if the period is strongly affected or dominated by drought stress. There should be an awareness of the greater uncertainty for the NPP values in these situations if data are not available at high resolution. On the other hand, the results suggest that there is no need to simulate at high resolution for long term regional NPP averages based on the simplified assumptions (soil and management constant in time and space) used in this study.  相似文献

Abstract

In rice plant, carbohydrates accumulated in leaf sheaths before heading are translocated to grain and affect yield formation greatly. To clarify the intrinsic mechanism of carbohydrate metabolism in the leaf sheath, we investigated the temporal and spatial variations of carbohydrate metabolism in the third leaf sheath counted from the top and their varietal differences. The results revealed that the amount of carbohydrate decreased from the base to the tip of the leaf sheath, irrespective of variety and developmental stage. However, the proportion of starch content in the basal one-fifth of the leaf sheath to that in the whole sheath varied from 35% to 60% with the variety. Comparing the activities of enzymes related to starch metabolism at the base, middle and tip of the leaf sheath in IR65598-112-2 (New plant type) with those in Nipponbare, the activities of ADP-glucose pyrophosphorylase, branching enzyme and granule-bound starch synthase (GBSS) showed varietal differences. Particularly, the activity of GBSS may play an important role in the varietal difference in spatial variation of starch content in the leaf sheath. In IR65598-112-2, the sucrose content in the leaf sheath was extremely high, suggesting that sucrose may be one of the carbohydrate reserves in this line.  相似文献

Summary

This study, in die Chilean Magellan Region, undertook the structural and biométrie characterization of Nothofagus betuloides (Mirb.) Blume (coihue de Magallanes) production forests to provide information for sustainable forest management. Six locations were surveyed, five on the continent and one on Tierra del Fuego Island. A total of 6,102 ha were photointerpreted and then checked and described in the field according to their location, environmental characteristic, and vegetation type. The degree of disturbance, stage of development, status of regeneration, and composition and cover of the understory were also described. From the surveyed locations, a total of 3,807 ha were production forests of which a 76% area had Nothofagus pumilio (lenga) and N. betuloides mixed forests. The remaining areas (24%) were pure N. betuloides forests. Nondisturbed, old-growth forests covered 47% of the surveyed area. The remaining area (53%) had some disturbance. In the nondisturbed forests, mean volume stocks were 438 m3 ha-1, and in disturbed forests, mean volume stocks were 316 m3 ha-1.  相似文献
