In this study we investigated the arylsulfatase-producing bacterial community (ARS-BC) in the rhizosphere soils of field-grown rape in comparison with that of barley. For this, the rhizosphere soils from both plant species were sampled four times during plant growth. Soil arylsulfatase (ARS) activity and the density and the structure of the cultivable ARS-BC on M9-Xsulf medium were then determined. ARS activity in rape rhizosphere was greater than in barley rhizosphere and evolved along the phenology in the two rhizosphere soils. In parallel, the average density of ARS-BC in the rape rhizosphere was higher than that in the barley rhizosphere. Moreover, ARS activity is correlated with ARS-BC density both in rape and barley rhizosphere soils. The structure of the ARS-BC in the rape rhizosphere was different from that in the barley rhizosphere. In the rape rhizosphere, the ARS-BC was substantially more structured than in the barley rhizosphere. Among the ARS-BC, Actinobacteria and Pseudomonads were significantly present in the both rhizosphere soils. Actinobacteria predominated in the barley rhizosphere while Pseudomonads were mostly represented under rape. It is possible that the differences in ARS activity observed between rape and barley can be attributed to a different ARS-BC size and/or a different ARS-BC structure under these two plant covers. This impact of rape may be connected to a selective effect of rhizodeposits released by rape roots to the functional bacterial community. Our findings suggest that plant species, via their rhizodeposits, may affect the functional bacterial community and thus influence the dynamic of S in soil. 相似文献

To explore long-term impact of organic and inorganic fertilizers on microbial communities, we targeted both the total bacterial community and the autotrophic ammonia oxidizing bacteria (AOB) in soil from six treatments at an experimental field site established in 1956: cattle manure, sewage sludge, Ca(NO3)2, (NH4)2SO4, unfertilized and unfertilized without crops. All plots, except the bare fallows, were cropped with maize. Effects on activity were assessed by measuring the basal respiration and substrate induced respiration (SIR) rates, and the potential activity of the AOB. To determine the bacterial community composition, 16S rRNA genes were used to fingerprint total soil communities by terminal restriction fragment length polymorphism analysis and AOB communities by denaturing gradient gel electrophoresis. The fertilization regimes had clear effects on both activity and composition of the soil communities. Basal respiration and r, which was kinetically derived as the exponentially growing fraction of the SIR-response, correlated well with the soil organic C content (r=0.93 and 0.66, respectively). Soil pH ranged from 3.97 to 6.26 in the treatments and was found to be an important factor influencing all microbial activities. pH correlated negatively with the ratio between basal respiration and SIR (r=0.90), indicating a decreased efficiency of heterotrophic microorganisms to convert organic carbon into microbial biomass in the most acid soils with pH 3.97 and 4.68 ((NH4)2SO4 and sewage sludge fertilized plots, respectively). The lowest SIR and ammonia oxidation rates were also found in these treatments. In addition, these treatments exhibited individually different community fingerprints, showing that pH affected the composition of AOB and total bacterial communities. The manure fertilized plots harbored the most diverse AOB community and the pattern was linked to a high potential ammonia oxidation activity. Thus, the AOB community composition appeared to be more strongly linked to the activity than the total bacterial communities were, likely explained by physiological differences in the populations present. 相似文献

The organic compounds released from roots (rhizodeposits) stimulate the growth of the rhizosphere microbial community. They may be responsible for the differences in the structure of the microbial communities commonly observed between the rhizosphere and the bulk soil. Rhizodeposits consists of a broad range of compounds including root mucilage. The aim of this study was to investigate if additions of maize root mucilage, at a rate of 70 μg C g−1 day−1 for 15 days, to an agricultural soil could affect the structure of the bacterial community. Mucilage additions moderately increased microbial C (+23% increase relative to control), which suggests that the turnover rate of microorganisms consuming this substrate was high. Consistent with this, the number of cultivable bacteria was enhanced by +450%. Catabolic (Biolog® GN2) and 16S-23S intergenic spacer fingerprints exhibited significant differences between control and mucilage treatments. These data indicate that mucilage can affect both the metabolic and genetic structure of the bacterial community as shown by a greater catabolic potential for carbohydrates. We concluded that mucilage is likely to significantly contribute to differences in the structure of the bacterial communities present in the rhizosphere compared to the bulk soil. 相似文献

Pollution induced community tolerance (PICT) has been suggested as an end-point measurement less affected by confounding environmental factors compared to standard methods of microbial growth, activity and community composition. We evaluated the use of PICT to determine Cu toxicity in vineyard soils polluted with Cu based fungicides (25-1120 mg Cu kg−1). These soils also varied in pH (4.3-7.3), organic C (0.31-6.91%) and texture (14-56% silt). PICT was estimated as bacterial community tolerance to Cu measured by the [3H]leucine incorporation method. Bacterial tolerance to Cu increased 9 times in the most polluted compared to the unpolluted soils. Cu tolerance was also affected to a minor degree by pH, organic C and soil texture. Lower bacterial tolerance was found in soils with high pH and organic C, probably due to Cu becoming less bioavailable in soils with high pH and organic C content. The silt content appeared to increase bacterial tolerance, probably due to fine soil particles decreasing Cu bioavailability during the PICT detection phase. Despite the effects of other environmental factors, the main determinant of increased bacterial community tolerance to Cu was the pollution level. PICT measured with the leucine incorporation technique thus appears to be a sensitive and stable concept to evaluate toxic impacts, unless soils with very different pH, organic C or texture are studied. 相似文献

2003年3月和2004年4月,新疆某养殖场和西安灞桥区某养殖场的白斑狗鱼(Esox lucius Linnaeus)相继爆发“败血症”。从具有明显症状的濒死病鱼体表病灶和内脏组织分离得到12株优势菌株,经人工感染试验表明:其中10株分离菌为致病菌。对分离菌株进行形态、培养特征、生理生化试验鉴定,认为该病原菌为杀鲑气单胞菌史氏亚种(Aeromonas salmonida subsp.smith)。血清学试验表明各分离菌株具有相同的保护性抗原。组织病理学研究表明:患病狗鱼病灶肌纤维明显肿胀断裂,肌纤维束之间出现间隙;肝胰组织严重坏死,细胞核溶解消失,细胞浆崩解;肾小管绝大部分均已坏死,细胞破裂,胞浆呈红色团块状流失于管腔。脾脏细胞大量坏死,髓质内含铁血黄素沉积。代表菌株EL0304-1药敏试验结果为:对供试24种抗菌药物中的磺胺二甲嘧啶钠、磺胺二甲嚯唑钠等不敏感;庆大霉素、卡那霉素等中度敏感;对氧氟沙星、培氟沙星、新霉素等高度敏感。 相似文献

Water stress, with its negative consequences on plant growth and survival, can be mitigated by Azospirillum brasilense inoculation. In tomato, A. brasilense delays wilting caused by a vascular pathogen, Clavibacter michiganensis subsp. michiganensis, by yet unknown mechanisms. We studied morphological, anatomical and physiological changes induced by A. brasilense in tomato that relate to water stress tolerance, which could explain the deferral in symptom expression. For this purpose, tomato seeds were treated or not with A. brasilense BNM65, and 5 weeks later plants were challenged with C. michiganensis subsp. michiganensis or mock inoculated with water. There was a large growth promotion associated to Azospirillum: treated plants had higher total biomass and leaf area. In relation to water stress tolerance, Azospirillum treated plants had larger xylem vessel area, higher stem specific hydraulic conductivity, thicker stems, and lower shoot/root dry matter and specific leaf area. These changes were opposite to those induced by C. michiganensis subsp. michiganensis. We conclude that A. brasilense favoured a better adjustment of plant-water relations by several mechanisms, and thus, transitorily alleviated symptoms expression of a vascular disease. 相似文献
