Background

The diaphragm is the major respiratory muscle affected by Duchenne muscular dystrophy (DMD) and is responsible for causing 80% of deaths. The use of mechanical forces that act on the body or intermittent pressure on the airways improves the quality of life of patients but does not prevent the progression of respiratory failure. Thus, diseases that require tissue repair, such as DMD, represent a group of pathologies that have great potential for cell therapy. The application of stem cells directly into the diaphragm instead of systemic application can reduce cell migration to other affected areas and increase the chances of muscle reorganisation. The mdx mouse is a suitable animal model for this research because its diaphragmatic phenotype is similar to human DMD. Therefore, the aim of this study was to assess the potential cell implantation in the diaphragm muscle after the xenotransplantation of stem cells.

Methods

A total of 9 mice, including 3 control BALB/Cmice, 3 5-month-old mdx mice without stem cell injections and 3 mdx mice injected with stem cells, were used. The animals injected with stem cells underwent laparoscopy so that stem cells from GFP-labelled rabbit olfactory epithelium could be locally injected into the diaphragm muscle. After 8 days, all animals were euthanised, and the diaphragm muscle was dissected and subjected to histological and immunohistochemical analyses.

Results

Both the fresh diaphragm tissue and immunohistochemical analyses showed immunopositive GFP labelling of some of the cells and immunonegativity of myoblast bundles. In the histological analysis, we observed a reduction in the inflammatory infiltrate as well as the presence of a few peripheral nuclei and myoblast bundles.

Conclusion

We were able to implant stem cells into the diaphragm via local injection, which promoted moderate muscle reorganisation. The presence of myoblast bundles cannot be attributed to stem cell incorporation because there was no immunopositive labelling in this structure. It is believed that the formation of the bundles may have been stimulated by cellular signalling mechanisms that have not yet been elucidated.  相似文献

Reasons for performing study: Assessing patients' quality of life (QOL) is a core part of clinical decision making. Various methodologies for assessing patients' QOL have been developed in human medicine and small animal veterinary disciplines. In contrast, the lack of aids for QOL assessment in equine veterinary practice leaves practitioners reliant on subjective assessments of QOL, which may be prone to avoidable errors. Objectives: This paper suggests pragmatic ways in which QOL may be enhanced, while remaining appropriate for the time, financial and owner‐based constraints within equine practice. Methods: Through interdisciplinary research, this paper identifies, adapts and applies insights from several areas of research and practical experience in order to develop an overarching approach to making QOL‐based decisions in clinical cases. Results: The paper identifies 6 steps involved in QOL‐based decision making and provides examples of how these steps may be practically applied. These include deciding what each clinician feels is important; deciding how to evaluate it, including taking owners' views into consideration; making decisions about each case and achieving the desired clinical outcomes. Conclusions: Practitioners can draw their own conclusions on how they may improve QOL assessment in practice and may usefully share these with colleagues. Reporting cases and sharing practical examples of QOL tools used on the ground are vital to the development of this field and appropriate methodologies. Potential relevance: Improvements in QOL assessment are relevant to all areas of equine veterinary practice, and several areas of research. 为探索治疗奶牛感染性蹄病的新药物,以患蹄病的绵羊为奶牛感染性蹄病的动物模型进行临床治疗试验。试验药物是针对奶牛感染性蹄病痛原特点研制的5种配方,即配方Ⅰ~配方Ⅴ。选择自然感染蹄病的绵羊50只,随机分为7个组,各组羊只分别为配方Ⅰ组6只、配方Ⅱ组7只、配方Ⅲ组10只、配方Ⅳ组9只、配方Ⅴ组8只、药物对照组5只、空白对照组5只。结果表明,在本试验药物组中,临床疗效依次为配方Ⅴ〉配方Ⅲ〉配方Ⅱ〉配方Ⅳ〉配方Ⅰ,配方Ⅴ的效果最佳,有效率和治愈率分别达100%、88.9%。综合比较,本试验药物的治疗效果要优于传统药物。  相似文献

以福建永安安砂库区生态公益林为研究对象,通过设置径流小区法,分别在坡度25°和32°的林地中,设计不间伐、间伐强度20%、40%、60%4种处理,研究不同间伐强度对安砂库区生态公益林养分流失的影响。结果表明:林地坡度25°情况下,在养分月流失量及流失总量上,间伐强度60%、40%均明显大于不间伐处理,而间伐强度20%与不间伐处理间差异并不明显,且流失量较少。林地坡度32°条件下,养分的月流失量及流失总量总体上随着间伐强度的加大而增加。此外,同一间伐强度下,养分流失总量随着坡度的增加而增加。可见,对库区生态公益林进行抚育间伐时,选择坡度较缓的林地进行低强度间伐(间伐强度20%)较为合理,能够在很好地发挥生态公益林的生态效益的同时获得一定的经济效益,进而实现库区生态公益林的可持续经营。  相似文献
