Seasonal distribution of phytoplasmas in Australian grapevines 总被引:1,自引:0,他引:1
The distribution and persistence of phytoplasmas were determined in Australian grapevines. Phytoplasmas could be detected using the polymerase chain reaction (PCR) from shoots, cordons, trunks and roots throughout the year, and phytoplasmas appear to persistently infect Australian grapevines from year to year. Phytoplasmas were not always detected in samples from the same sampling area from one sampling period to the next. Phytoplasma detection by PCR was improved by sampling from shoots, cordons and trunks, especially during October (early spring). The diseases expressed by the 20 grapevines used in the distribution and persistence studies were monitored. Australian grapevine yellows disease (AGY) was expressed by 17/20 grapevines at some time during the study, whilst only 4/20 and 15/20 grapevines expressed restricted growth disease (RG) and late season leaf curl disease (LSLC), respectively. All grapevines with RG and LSLC also had AGY. The three diseases were persistently expressed in some grapevines and remission of disease was observed in others. The results of PCR detection in the same grapevines indicated that phytoplasmas were more frequently detected in AGY-affected grapevines that also expressed RG and LSLC compared with grapevines expressing AGY alone. A monoclonal antibody to an Albanian isolate of plum pox potyvirus (PPV) was obtained (MAbAL), that specifically recognized strain M of this virus. The specificity of MAbAL, assessed by comparative ELISA on 130 PPV isolates of different geographical origin, 22 of which were also tested by comparative IC-PCR, gave consistent and highly reproducible results. MAbAL seems to be elicited by a stable surface determinant that makes it particularly suitable for successful use under a wide range of conditions. Simple diagnosis by ethanol immersion (SDEI) to detect Glomerella cingulata was used to detect three other fungi that also cause latent infection of strawberry plants. Signs on strawberry leaves with asymptomatic latent infection by Colletotrichum acutatum became visible using SDEI. Salmon-pink conidial masses were produced in the acervuli on the treated leaves 5 days after incubation at 28°C. In the case of Dendrophoma obscurans, pycnidia with amber conidial masses formed 5 days after incubation at 28°C. The pycnidia were observed mainly on the ribs, and conidial masses exuded from the ostiole. These macroscopic conidial masses were similar to those of G. cingulata and C. acutatum. When water was dripped onto a lesion caused by D. obscurans, the pycnidia exuded white filamentous conidial masses, making the distinction of D. obscurans from G. cingulata or C. acutatum. On petioles with latent infection by Fusarium oxysporum f. sp. fragariae, white aerial hyphae grew out from the vascular tissues on the cut surface 3 days after incubation at 28°C and were easily observed by eye or with a loupe. Thus, SDEI was also useful for diagnosing latent infection of strawberry plants by C. acutatum, D. obscurans, and F. oxysporum f. sp. fragariae. 相似文献

This paper reports results of a study conducted to investigate the concentrations of seven mineral elements in yak blood, forage and water resources around the Qinghai Lake in Qinghai Province in different seasons. Meanwhile, the nutritional compositions of the forage were also surveyed. The results suggest that the mineral elements and the forage nutrients change in a seasonal pattern. In yak blood,the sodium(Na)concentration varies from 0.291 to 0.034 mg/mL,and this is lower than the normal value. In the forage,the ratio calcium(Ca)to phosphorus(P)is 4.06~7.47:1 and potassium(K)to Na 30~27:1. These results indicate that the nutrition of the yak in the area is deficient in Na but high in K. For the withered forage sampled in February,the protein content is only 31.14% of the total protein in the forage growing at puerile stage in June. The severe loss of protein by 68. 9% and decrease of effective nutrients in the wintered forage are considered to be the reasons resulting in the poor condition of yak in winter and spring seasons. 相似文献

运用统计学和ArcGIS软件的地统计分析模块对海南砖红壤香蕉园59个土壤采样点两个取样土层(0~20 cm和20~40 cm)养分进行空间变异性研究。结果表明:香蕉园土壤各养分变异系数均为中等变异。0~20 cm土层土壤各养分含量大于20~40 cm土层。0~20 cm和20~40 cm土层中的碱解氮、交换性钙和交换性镁及0~20 cm土层的有机质均符合高斯模型,0~20 cm土层速效磷与20~40 cm土层速效钾符合球状模型,而20~40 cm土层速效磷与0~20 cm土层速效钾符合圆形模型,20~40 cm土层有机质符合指数模型。各养分的变程范围为109.217~279.796 m,有机质具有强烈的空间相关性。0~20 cm土层速效钾的块金效应值小于20~40 cm土层,0~20 cm土层碱解氮、速效磷、交换性钙、交换性镁高于20~40 cm土层。有机质、20~40 cm土层速效磷、0~20 cm土层速效钾的块金效应小于25%,其它养分均在25%~75%,具有中等的空间相关性。确定了合理取样数,0~20 cm碱解氮、速效磷、速效钾、交换性钙、交换性镁、有机质合理取样数分别为:5、30、26、29、25、3;20~40 cm碱解氮、速效磷、速效钾、交换性钙、交换性镁、有机质合理取样数分别为:4、44、30、26、25、3,就可以满足变量施肥的要求。 相似文献
