对东北矮紫杉进行了愈伤组织的诱导,并建立了细胞悬浮培养系.研究表明,茎、叶均可诱导出愈伤组织,愈伤组织诱导培养基以MS+2,4-D 1.0 mg/L+NAA 1.0 mg/L+KT 0.1 mg/L为佳,愈伤组织继代培养基以B5+2,4-D 2.0 mg/L+KT 0.1 mg/L为佳.以嫩枝为外植体,愈伤组织的诱导率较高、状态较好,且较适于悬浮培养.悬浮培养的接种量2.0 g/瓶为最佳密度,细胞生长周期为20 d,对数生长期为10~16 d,细胞收获期为16 d.试验中还发现,聚乙烯吡咯烷酮是东北矮紫杉组培中解除培养物褐化的较好抗氧化剂,使用浓度为1mg/L. 相似文献

Natural spawning, early development and larviculture of the ornate goby Istigobius ornatus in captivity were studied for the first time. I. ornatus spawned 46 times from 31 October 2013 to 31 October 2014. Fecundity ranged from 246 to 10,214 eggs per clutch, with an average hatching rate of 77.8% ± 9.9% (M ± SEM). Fertilized eggs (1.31–1.54 × 0.46–0.50 mm in diameter) were adhesive demersal and oval‐shaped. Embryonic development lasted 84 hr at 27.5 ± 0.5°C. Newly hatched larvae [2.12 ± 0.04 mm in total length (TL)] transformed to the juvenile stage completely when TL was 7.79 mm. Effects of different water temperatures (24, 28 and 32°C) and salinities (10, 15, 20, 25, 30, 35 and 40 g/L) on per cent survivals (%) and survival activity indices (SAIs) were tested. Survival was not significantly different under different temperatures; SAIs was significantly higher at 28°C. Larvae showed the significantly higher survival and SAIs at salinities 10–30 g/L than at 35 and 40 g/L. Effect of different prey densities on survival was significantly higher in 7 days post hatch larvae fed 20 and 30 rotifers/ml. 为探讨厚壳贻贝稚贝对自然微生物膜中海洋细菌的附着行为反应,本论文研究了厚壳贻贝(Mytilus coruscus)稚贝附着与硅烷基化附着基表面、微生物膜密度以及细菌种属系统发育之间的相互关系。研究表明所测海洋细菌均能显著促进厚壳贻贝稚贝的附着;其中Staphylococcus sp. 1和Cobetia sp. 1表现出较低诱导活性,且这两株细菌形成的微生物膜密度与稚贝附着率之间无显著相关性;其他7株海洋细菌均表现出中等程度诱导活性,且所形成的微生物膜密度与稚贝附着率之间呈显著相关性。系统发育分析表明所测海洋细菌对厚壳贻贝稚贝附着的诱导活性与细菌种属无关。因而,硅烷基化表面的海洋附着细菌对厚壳贻贝附着有着显著性促进作用,本研究将为后续开展厚壳贻贝稚贝附着机制奠定了基础。 相似文献

- Although the frequency of occurrence of plastic ingestion in the large-sized dolphinfish and tunas taken by the Hawai'i longline fishery is very low (frequency of occurrence < 5% of sampled individuals), the ingestion of plastic in smaller-sized specimens caught with pole-and-line gear by commercial and recreational fishers has not been investigated.
- This study examined ingestion of >0.25 mm marine plastic debris (MPD) by four predatory fish species caught by commercial fishers around the Main Hawaiian Islands, and documented ingestion in three species: 85.7% of albacore tuna (n = 7), 40.0% of skipjack tuna (n = 10) and 12.5% of dolphinfish (n = 8).
- Yellowfin tuna (n = 10) did not contain any MPD, probably owing to the high proportion of empty stomachs (60%).
- For skipjack tuna, the frequency of occurrence of MPD ingestion was significantly higher for the smaller-sized specimens caught with pole-and-line (40%), compared with the larger-sized specimens caught with longlines (0%).
- For dolphinfish, the frequency of occurrence of MPD ingestion was similar for the similar-sized specimens caught with pole-and-line and with longlines.
- The ingested MPD items were micro–meso plastics, between 1 and 25 mm. While most ingested items were fragments, albacore also ingested line and skipjack also ingested sheets.
- The predatory fishes ingested light MPD items that float in sea water, but there were species-specific differences in their polymer composition: albacore contained more polypropylene and polyethylene, and skipjack contained more elastomers, characterized by a high percentage of ester plasticizers.
- Altogether, these results suggest that albacore and skipjack tunas ingest plastic of different types and polymers. Yet more research is needed to understand how differences in vertical distribution, foraging ecology and diet influence the MPD sampled by these predatory fish species.

Correlations between fine‐scale oceanographic features and aggregations of marine mammals are frequently reported, but the physical forces shaping these relationships are rarely explored. We conducted a series of oceanographic observations and remote sensing surveys of an oceanographic feature near Grand Manan Island known to attract marine mammals on flood tides. We tracked drift drogues from cliff‐top with a theodolite and conducted box‐type surveys with an acoustic Doppler current profiler (ADCP) to assess flow patterns within the oceanographic feature. The feature was also visualized with Synthetic Aperture Radar (SAR) scenes. Drift drogues were advected towards a shear line originating near the northern tip of the island and entrained in one or more eddies downstream. ADCP surveys confirmed the presence of the shear line between rapid easterly flow and slower return flow. As the tide progressed, the shear line extended and manifested a single anti‐cyclonic eddy at its distal end. As the flood tide progressed, northerly flow along the eastern shore of the island intensified and deflected the shear line northwards, shedding the eddy at slack high water. SAR images confirmed the presence of the shearline and eddy system, illustrating the evolution of a wake behind the island on flood tides. Profiles of flow direction and acoustic backscatter revealed secondary flows within the wake consistent with models and observations of other wakes. Oceanographic and remote sensing observations confirm that an island wake is generated by tidal flow past Grand Manan Island and provide an ecological context for the predictable aggregations of odontocete and mysticete cetaceans observed foraging within this region. 相似文献
