Summary At present research into the inhibition of plant growth by sub-optimal temperatures is concerned with the examination of a large number of factors. No single aspect of the response to temperature has been found to be of paramount importance. As a consequence selection for low temperature tolerance is complicated.Here a new approach is proposed in which inhibition of cell division is regarded as a limiting process in growth under sub-optimal temperatures, and the role of other processes as the supply of substrate for cell division. This approach may be of use in the examination of the response of plant growth to temperature and could form the basis of a method for screening and selection for cold tolerance using cells in culture.Guestworker Project Group Tomato Physiology.  相似文献

In vitro regenerated sunflower (Helianthus annuus L.) plants (R1) were self-fertilized and the R2 generation was evaluated for qualitative traits. A broad range of phenotypic variation was observed and mutation frequencies were calculated. Some in vitro induced variant phenotypes were similar to known spontaneous or induced mutations in sunflower, while others were new. Chlorophyll and carotenoid deficiencies, chimaerical variegation, fasciated stem and capitulum, abnormal shoot development, and other morphological variations, were noted. Substitution of anthers with petaloid structures in a disk-floret mutant indicates a possible homeotic mutation induced by in vitro tissue culture.  相似文献

In vitro culturing of plant cells can cause changes in karyotype. Chromosome variations following long-term propagation in suspension culture of Triticum timopheevii (Zhuk.) Zhuk. were studied by routine staining and C-banding. The culture was highly heterogeneous with respect to the number and structure of chromosomes. The modal class cells had a lower chromosome number than T. timopheevii (2n= 28). This data was confirmed by cytophotometric analysis of nuclear-DNA content. Frequencies of chromosome loss varied for different homoeologous groups. At genome chromosomes tended to be preferentially eliminated in cells of different ploidy levels. Deletions, insertions, translocations, telocentric chromosomes, isochromosomes and dicentrics and their derivatives were observed in cultured cells. Chromosomes of various homoeologous groups differed in the frequencies and spectra of re—arrangements, but most aberrations occurred in the G-genome chromosomes. In vitro chromosome modifications did not correspond to in vivo variation. Presumably, this difference was caused by differences in the mechanisms of adaptation to the environment at the levels of the cell and the whole organism. G-genome chromosomes were more frequently involved in this process, both in vivo and in vitro.  相似文献

Summary Interspecific crosses were made to introduce the disease resistance of Cyclamen graecum into C. persicum cultivars and the abortive hybrid embryos were rescued by ovule culture. Diploid and tetraploid cultivars of C. persicum (CPD, 2n=2x=48; CPT, 2n=4x=96) were the pistillate parents and wild form of C. graecum (CG, 2n=84) were the staminate parents. After pollination, crossed ovaries were periodically collected and examined using paraffin sections. Histological observations suggested that both hybrid ovules of CPD × CG and CPT × CG should be transferred to the culture medium 35 days after pollination. Based upon this observation, crossed ovaries were collected 35 days after pollination, then ovules with placenta were explanted on culture medium and cultured in the dark at 25°C. Plantlets were induced from ovules cultured in MS medium containing 3% sucrose and 10% coconut milk. The hybrids (2n=66) derived from CPD × CG failed to yield viable seeds by self-pollination, although they showed some pollen fertility. In contrast, the hybrids (2n=90) derived from CPT × CG showed high pollen fertility and yielded viable seeds by self-pollination. Genetic control of somatic embryogenesis in cotton petiole callus cultures   总被引:8,自引:0,他引:8  
Summary Three commercial varieties (Acala SJ-5, Coker 312 and Paymaster 303) and three exotic accessions (T1, T25 and T169) of cotton (Gossypium hirsutum L.) were tested for ability to undergo somatic embryogenesis. Sections of split petiole were cultured on 3 media and evaluated for embryogenesis after 180 days. Embryogenic T25 and Coker 312 plants were selected and crossed in a diallel with non-embryogenic Acala SJ-5, Paymaster 303, T1 and T169 plants. F1, F2 and BC1 populations were generated and tested for embryogenesis on a medium of MS salts and vitamins (1962) plus (per liter) 4.0 mg NAA, 1.0 mg Kn, 30 g glucose, 100 mg myo-inositol, 2.0 g Gelrite and 0.75 g MgCl2. Segregation for both occurrence and magnitude of embryogenesis was observed, suggesting the action of more than one gene.  相似文献

Summary The effects of the addition of sulfurous acid into culture solution and of cold treatment of the solution were examined to simplify the culture of detached wheat ears. In the simplified method, detached ears could be cultured at room temperature on the liquid medium containing 100 g/l sucrose and 0.075% sulfurous acid without any sterilization. The immature seeds in detached ears cultured by this method were treated with low temperature or with chemicals known to have vernalizing effect. The chemical treatment did not affect the chilling requirement of immature embryos, although photoperiodic response and narrow-sense earliness were reduced by kinetin and trypsin. The low temperature treatment drastically affected the chilling requirement, and fully vernalized mature seeds having normal germinability were obtained by treating the detached ears in culture with low temperature from 10 days after anthesis.  相似文献

Summary Interspecific hybrids have been obtained in an incompatible cross between Glycine max and G. tomentella through the in vitro culture of hybrid embryos. The percentage of successful pod setting in the crosses averaged 12.8% but there were marked differences depending on the soybean cultivar used as the female parent. Hybrid embryos at globular to heart stages were extracted from the embryo sac 15–25 days after pollination and cultured in vitro. Hybrid plants were successfully obtained by culturing the embryos on B5 medium supplemented with 0.1 mg/l IBA followed by culture on B5 medium supplemented with 0.1 mg/l TBA plus 0.25 mg/l 2-iP. The F1 plants resembled the wild male parent in growth form, but had an intermediate leaf shape between that of the parents.  相似文献

The culture of unpollinated ovules is shown to be a suitable system for the production of haploid sugar beet (Beta vulgaris L.). The yield of haploids depended upon the genotype and varied between 0 and 13 % with a mean of 1.0 %. Haploid plants could be produced from approximately 50 % of all genotypes examined. The majority of the haploids isolated (about 90%) maintained the haploid genome level during the in vitro culture and propagation; 10% of the haploid clones showed a spontaneous doubling to the diploid genome level.  相似文献
