The aims of this study were to analyse the protein phosphatase 1 regulatory subunit 11 (PPP1R11) expression and cellular localization in yak follicles and investigate its effects on cell proliferation, apoptosis and oestrogen secretion in granulosa cells (GCs). Ten healthy and non-pregnant female yaks (4-year-old) were used as experimental animals. The mRNA relative expression level of PPP1R11 in GCs from small (<3.0 mm), medium (3.0–5.9 mm) and large (6.0–9.0 mm) follicles was detected by RT-qPCR, and the cellular localization of PPP1R11 protein was detected by immunohistochemistry staining (IHC). After isolation, culture and identification of yak GCs in vitro, si-PPP1R11 and si-NC (negative control) were transfected into GCs. RT-qPCR and immunofluorescence staining were used to evaluate the interference efficiency, and ELISA was performed to detect oestrogen concentration. Then, EdU staining and TUNEL staining were conducted to analyse cell proliferation and apoptosis. In addition, the oestrogen synthesis, proliferation- and apoptosis-related genes were detected by RT-qPCR after knockdown PPP1R11. The results showed that PPP1R11 is mainly located in ovarian GCs, and the expression levels of PPP1R11 in GCs from large follicles were significantly higher than that from medium and small follicles. Transfection of si-PPP1R11 into GCs could significantly inhibit the expression of PPP1R11. Interestingly, the oestrogen secretion ability and the expression level of oestrogen pathway-related genes (STAR, CYP11A1, CYP19A1 and HSD17B1) were also significantly downregulated. Moreover, the proportion of positive cells was decreased, and cellular proliferation-related genes (PCNA, CCNB1 and CDC25A) were significantly downregulated after knockdown PPP1R11. However, the proportion of apoptotic cells was increased, and apoptosis-related genes (BAX, CASP3 and P53) were significantly upregulated. Taken together, this study was the first revealed the expression and cellular localization of PPP1R11 in yak follicles. Interference PPP1R11 could reduce oestrogen secretion, inhibit proliferation and promote apoptosis in GCs, which provided a basis for further studies on the regulatory mechanism of PPP1R11 in follicle development. 相似文献

【目的】了解江苏地区禽腺病毒(Fowl adenovirus, FAdV)的基因遗传演化情况及致病性,为FAdV的流行病学研究和疫病防控提供参考。【方法】通过SPF鸡胚盲传、PCR鉴定、电镜观察、全基因组测序、序列比对与相似性分析判定病毒血清分型,并将分离株以5×105.33 EID50剂量胸肌注射的方式感染10日龄SPF雏鸡进行动物回归试验。【结果】PCR结果显示,该分离株为Ⅰ群FAdV阳性,在透射电镜下可见球型、无囊膜、具有腺病毒典型的二十面体结构,通过全基因组和Hexon基因进行序列分析表明该分离毒株为FAdV D种血清11型毒株,命名为JSNT-1株。将该毒株感染SPF雏鸡,死亡率为10%(1/10),病死鸡剖检可见肝脏褪色变黄,出血肿胀,边缘钝圆;肾脏肿大出血,苍白等病变。通过实时荧光定量PCR试验可从口腔及泄殖腔中检测到排毒,且病毒在鸡体内多个组织器官均有分布。病理组织学结果显示,死亡鸡的肝细胞变性坏死,出现嗜碱性核内包涵体;肾小球上皮细胞变性坏死;心肌可见大量炎性细胞浸润。【结论】分离株为FAdV血清11型,感染SPF鸡后临床发... 相似文献

糯玉米陕白糯11是西北农林科技大学农学院专用玉米研究室,1998年用自选系Y89作母本,自选系Y90为父本杂交育成。经过2002~2004年陕西省糯玉米区试。2004年省特用玉米生产试验,表现出籽粒容重749g/L,粗蛋白10.91%,粗脂肪5.41%,粗淀粉70.08%,支链淀粉占粗淀粉99.49%,赖氨酸含量0.31%,鲜食穗感官及蒸煮加工品质评分90,达到国家糯玉米一级指标,三年区试16点次11点增产5 点次减产,干籽率产量6238.5kg/hm2较中糯1号增产20.3%,抗倒,农艺性状优良,可作 速冻加工、鲜食上市或专用淀粉加工的原料,2005年3月7日通过陕西省第36次农作物品种审定委员会审定推广。 相似文献

Summary The mutant genotype pg11pg11 pg12pg12-epistasis of double recessive factors-determines greater precocity of flowering. The possibility of using this mutant in the production of hybrid seed as a mean of synchronizing flowering in parent strains of different earliness is studied. The differences between mutant and green control genotypes in the characters of days to pollen shedding, pollen production, and height of plants are evaluated. Four mutant lines are considered together with their respective green near-isolines in two distinct localities and across four successive plantings.The mutant near-isolines presented lower values than those of the green near-isolines for all the characters. The maximum reductions in days to flowring were obtained in early plantings and late maturity strains. The reduction in pollen production was about 50%. The reduced height of the mutants is explained, at least in part, by the correlation of this factor with days to flowering. The mutants did not prove to be more sensitive to the environment than their green near-isolines. Seed production was not studied since the mutant is very deficient for this character and it could therefore only be utilized as the male parent. We concluded that the mutant, could be used to synchronize flowering in the industrial production of hybrid seed. 相似文献

Forty-one lettuce cultivars (butterhead type), carrying race-specific resistance gene Dm11 in combination with some others, were studied for field resistance to Bremia lactucae. Significant differences in the level of field resistance were revealed. Most of the cultivars under study were highly susceptible under field conditions. There are evident relationships between combinations of race-specific Dm genes and field resistance. The cultivars with gene combinations Dm2, Dm3 and Dm11 are susceptible. A very high level of field resistance was detected in the cultivars with combination of genes Dm5/8, Dm6 and Dm16 (cvs. ‘Celia’, ‘Katanga’ and ‘Musette’). 利用化学药剂与生防菌结合,对由尖孢镰刀菌芹菜专化型引起的西芹黄萎病进行综合防治研究。结果表明:苗期使用50 %多菌灵可湿性粉剂(WP)、50 %福美双•多菌灵WP(活根菌灭)或40 %噁霉灵WP(噁霉敌)灌根均能显著消除育苗基质中的病原菌孢子,孢子下降率达99.00 %以上;大田继续使用上述3种化学药剂或改用生防菌芽孢杆菌S11和M3,都能够显著降低西芹黄萎病发病率,防治效果最高为85.98 %;其中持续单独使用50 %多菌灵WP或苗期使用50 %多菌灵WP、大田改用生防菌芽孢杆菌对西芹的生长没有不良影响。 相似文献
