两个黄瓜(Cucumis sativus L.)叶色突变体的比较分析研究   总被引:1,自引:1,他引:0  
通过对黄瓜黄绿叶突变体9110Gt和NCG-042植株表型观测、遗传分析和分子标记验证,证明9110Gt是区别于NCG-042的新叶色突变体。这两个黄绿叶突变体在表型上存在一定的区别:9110Gt在苗期表现叶色黄化,而NCG-042的心叶在整个生育期都表现黄化。遗传分析证明,两个突变体的叶色突变性状分别由两个不同的等位基因控制,且两基因间存在互补作用。分子标记的检测结果也进一步证实了这一结论。  相似文献

Permethrin resistance in 16 field populations of Colorado potato beetle was established by bioassay. Permethrin-resistant genotypes were detected using two genotyping techniques, bidirectional polymerase chain reaction amplification of specific alleles (bi-PASA) and single stranded conformational polymorphism (SSCP). The bi-PASA technique was 86% and SSCP was 95% reliable. In terms of accuracy, bi-PASA determined the correct genotype compared to the actual sequencing data 98% and SSCP 93% of the time. There was marked correlation between increasing survivorship (increasing LD50values) and increasing proportions of resistant genotypes (increasing percentage of resistant allele). These techniques allowed the detection of a resistant allele 98% of the time when sampling only 384 beetles from an overwintering adult population (4995 beetles/ha) that possessed a 1% resistant allele frequency at a cost of $500/ha, substantiating the cost effectiveness of the genotyping approaches to monitoring.  相似文献

为了解江苏淮北地区小麦品种资源的籽粒硬度概况及硬度基因型分布规律,以74份近年来江苏淮北地区所育品种(系)和38份来自黄淮其他麦区的常用亲本为材料,采用单籽粒谷物硬度测试仪、KASP标记检测技术和基因扩增及测序技术对其SKCS硬度值及硬度基因型进行鉴定。硬度检测结果表明,供试小麦品种(系)硬度变化范围较大,但硬质麦的比例最大,为70.5%。与常用亲本相比,江苏淮北地区育成品种中软质麦比例较高,为34.3%,但在高代品系中软质麦比例下降到20.5%。基因型检测结果表明,在Puroindoline-D1位点,供试品种(系)中共检测到4种基因型,即野生型(Pina-D1a/Pinb-D1a)、Pina-D1b、Pinb-D1b和Pinb-D1p,其频率依次为25.0%、2.7%、67.9%和4.5%。其中,野生型和Pinb-D1p主要分布在江苏淮北地区。不同硬度基因型的硬度值也存在差异,其中以Pina-D1b基因型的硬度值最高,野生型(Pina-D1a/Pinb-D1a)硬度值最低,Pinb-D1b和Pinb-D1p两硬质类型的籽粒硬度没有显著性差异。在Pinb-2位点,供试品种(系)中共检测到25份材料为Pinb-B2b基因型,包含21份硬质麦、2份混合麦和2份软质麦,其平均硬度值为63.8。  相似文献

Starch is composed of two glucose polymers: amylose, formed by long linear chains, and amylopectin, which is larger and highly branched. The ratio between the two polymers (usually 22–35% amylose, 68–75% amylopectin) affects the starch properties, thereby determining different aspects of wheat quality. Waxy protein, which is the sole enzyme responsible for amylose synthesis, showed polymorphism in a collection of one hundred and three Mexican landraces, including null alleles at Wx-A1 and Wx-B1 loci, respectively. Molecular characterization of these alleles showed that the null Wx-A1 allele presented in only one accession was novel (Wx-A1o) and involved a deletion spanning the three and a half last exons of the gene. Some of the accessions lacking the Wx-B1 protein contained the common allele Wx-B1b (12.6%) (deletion of approximately 67 kb), while four others (3.8%) possessed the novel Wx-B1l, characterized by the deletion of one cytosine in the second exon of the gene and leading to a change in the ORF. This novel allele is particularly interesting because the absence of the Wx-B1 protein in this case was not associated with the lack of the other genes included in the approximately 67 kb region lost with the common null allele Wx-B1b.  相似文献

Cysteine‐rich secretory protein‐3 (CRISP‐3) and some of its nonsynonymous polymorphism have been related to the fertility and freezability of stallion semen; however, the role of the CRISP‐3 gene and its seminal plasma protein in the raw semen quality is still unknown. The aim of this study was to evaluate the relationship of CRISP‐3 with semen quality in stallions. DNA was obtained from blood samples of 100 stallions, from which 30 stallions were randomly selected to obtain 60 ejaculates. Through PCR amplification and sequencing, the variation of four nonsynonymous SNPs from CRISP‐3 was identified and haplotypes were derived. Semen quality was assessed through the total motility (MOT), sperm vitality (SV), normal morphology (NM), functional integrity of membrane (MI) and a seminal quality index (SQi). CRISP‐3 protein content of seminal plasma (SP) was determined by ELISA. The effect of the genotype, the haplotype and the concentration of the CRISP‐3 protein on the seminal quality were evaluated through generalized linear models and linear regression analyses. Homozygous genotypes for SNP1, SNP2 and SNP3 and the heterozygous genotype for SNP4 showed a positive effect on seminal quality. Different haplotypes with positive effect on MOT, SV, NM, MI and SQi were identified. The allelic substitution analysis resulted in positive regression coefficients for MOT (SNP2) and MI (SNP2 and SNP3). A high level of CRISP‐3 resulted in a higher MOT and SQi. It is concluded that the quality of stallion semen is influenced by the genotype of CRISP‐3 and the concentration of CRISP‐3 protein in SP.  相似文献

Grain physical characteristics and milling behavior of a durum wheat line in which both wild-type puroindoline genes were translocated and stabilized after backcrossing (Svevo-Pin) were compared with the parent line (Svevo). The only observed differences between grain characteristics were the mechanical resistance and starchy endosperm porosity revealed through vitreosity measurement. A significant increase of flour and a decrease of semolina yield and break milling energy were observed from Svevo-Pin in comparison with the non-recombinant parent line in accordance to the lower grain mechanical resistance and higher porosity measurements. Moreover, the particle size distribution shown for Svevo-Pin flour appeared consistent with a lower adhesion between starch granules and the protein matrix attributed to the presence of wild-type puroindolines. Coarse bran yield was conversely increased. This appeared to be due to a lower starchy endosperm recovery as a higher proportion of grain starch was found in this bran fraction. Flour from the durum parent line was inversely enriched in phytic acid, a cellular marker of the aleurone layer. Starch damage was also lower in Svevo-Pin flours in comparison with Svevo. All of the observed differences between translocation and parent lines were confirmed independent of the culture growth conditions (n = 12).  相似文献

【目的】研究亚洲大豆栽培品种地理群体的遗传多样性、特异性和群体分化。【方法】应用大豆基因组64对SSR分子标记技术,对亚洲216份栽培大豆品种遗传变异进行分析。【结果】亚洲大豆栽培品种遗传多样性丰富,地理群体(中国东北、中国黄淮、中国南方、朝鲜半岛、东南亚、南亚)间存在较多互补等位变异数,最多的在中国黄淮与南亚群体间;各地理群体拥有各自特有或特缺的等位变异。亚洲大豆全群SSR标记遗传距离聚类(聚成6类)与地理群体分类间有极显著相关性,地理分群有其相应的遗传基础。亚洲全群由2类血缘组成,分别占中国国内和国外2大类群的绝大部分;地理群体间2类血缘组成的差异明显。国内与国外各群体间以中国南方与东南亚群体间分化最小;国外群以东南亚与朝鲜半岛群体间分化最小;国内群以中国黄淮与中国南方群体分化最小。【结论】亚洲大豆栽培品种地理群体间具有位点和等位变异的特异性,各群体间可以相互补充的位点及其等位变异甚丰富,利用国外栽培品种可以拓宽中国品种的遗传基础。  相似文献

The closely linked genes puroindoline a (Pina) and puroindoline b (Pinb) control most of the variation in wheat (Triticum aestivum) grain texture. Mutations in either Pina or Pinb result in hard grain with wild type forms of both genes giving soft grain. Asian noodles are prepared from both hard and soft classes of wheat. Our objective was to examine color and texture characteristics of white salted noodles processed from flours of transgenic isolines of Hi-Line hard red spring wheat over expressing Pina-D1a, Pinb-D1a or both and a control giving a range in grain texture from very soft to hard. White salted noodles were prepared and color and texture characteristics were measured. The three softer textured transgenic isolines showed greater change in L* with time than Hi-Line. The noodles were more adhesive (more negative value), firmer, and chewier as the grain texture became successively softer when cooked at 5 min. These texture differences were not as apparent when noodles were cooked for an optimum time. Starch pasting properties did not explain the noodle textural differences. A possible explanation for the noodle texture differences may be related to starch damage which ranged from 2.2% for HGAB to 6.7% for Hi-Line, flour particle size differences and subsequent water absorption differences among the four genotypes. Over expression of puroindolines did not enhance quality of white salted noodles when prepared under these conditions.  相似文献

In this study, we characterized the genetic resistance of the Andean bean cultivars Kaboon and Perry Marrow and their relation to other sources of anthracnose resistance in common bean. Based on the segregation ratio (3R:1S) observed in two F2 populations we demonstrated that Kaboon carries one major dominant gene conferring resistance to races 7 and 73 of Colletotrichum lindemuthianum. This gene in Kaboon is independent from the Co-2 gene and is an allele of the Co-1 gene present in Michigan Dark Red Kidney (MDRK) cultivar. Therefore, we propose the symbol CO-1 2 for the major dominant gene in Kaboon. The Co-1 is the only gene of Andean origin among the Co anthracnose resistance genes characterized in common bean. When inoculated with the less virulent Andean race 5, the segregation ratio in the F2 progeny of Cardinal and Kaboon was 57R:7S (p = 0.38). These data indicate that Kaboon must possess other weaker dominant resistance genes with a complementary mode of action, since Cardinal is not known to possess genes for anthracnose resistance. Perry Marrow, a second Andean cultivar with resistance to a different group of races, was shown to possess another resistant allele at the Co-1 locus and the gene symbol Co-1 3 was assigned. In R × R crosses between Perry Marrow and MDRK or Kaboon, no susceptible F2 plants were found when inoculated with race 73. These findings support the presence of a multiple allelic series at the Andean Co-1 locus, and have major implications in breeding for durable anthracnose resistance in common bean.
