In carrot, two codominant sequence-tagged site (STS) markers, flanking in tight linkage the Meloidogyne javanica resistance (Mj-1) locus, were employed to investigate the association between expression of resistance and locus dosage. Phenotypic expression of homozygous resistant (R); heterozygous; and homozygous susceptible (S) individuals in an F2 population of 396 F2 plants from ‘Brasília-1252’ (R) × ‘B6274’ (S) was estimated for three resistance criteria: total egg production per plant (TEP), egg production per gram of fibrous root (EPG) and root gall index (RGI). The homozygous resistant class had average values of 403.9 for TEP; 147.5 for EPG and 0.8 for RGI. The heterozygous class had 1,673; 477.3; and 0.16 whereas the homozygous susceptible class had 68,604; 11,877; and 2.54, respectively. The dominance ratio (d/a) indicated that genomic region(s) derived from the resistant parent encompass genetic factor(s) with almost complete dominance for RGI (d/a = 0.93–0.94) and incomplete dominance for transformed (TEP)0.25 and (EPG)0.25 (d/a = 0.63–0.65). Broad sense heritabilities were high varying from 72.9% for (EPG)0.25 to 86.0% for RGI. Narrow sense heritability values ranged from 55.9% for RGI to 64.3%for (TEP)0.25. Highly significant orthogonal contrasts were observed between homozygous resistant vs. heterozygous for (TEP)0.25 and (EPG)0.25. Marker-assisted selection could greatly facilitate the incorporation of the Mj-1 allele in both male-fertile and male-sterile counterpart lines in order to obtain F1 hybrids with the most effective levels of resistance. In pepper (Capsicum annuum), the major genes (R‐genes) Me1 and Me3 confer resistance against root‐knot nematodes (Meloidogyne spp.). The combination of R‐genes and quantitative resistance factors in the same genotype is considered a good breeding strategy for increasing the durability of R‐genes. To ascertain this hypothesis, five pepper inbred lines, differing in their quantitative resistance level, were combined with Me1 or Me3 genes in F1 hybrids. The resistance of inbred lines and F1 hybrids was evaluated in a greenhouse with soil naturally infected by M. incognita in two successive growing years. In both years, lines carrying Me3 were less infected by the nematode when combined with quantitative resistance. An increase in nematode infection was observed in the second growing year in lines carrying Me1 or Me3, independently of quantitative resistance. The infection level recorded in inbred lines without R‐genes was similar in both years. The effectiveness of quantitative resistance controlling M. incognita is confirmed in greenhouse conditions, although the durability of Me1 and Me3 when combined with quantitative resistance factors was not seen to increase.  相似文献

A germplasm collection of 39 eggplant Solanum melongena genotypes and one accession of S. torvum was evaluated for resistance to the root-knot nematode (Meloidogyne javanica) by using controlled greenhouse tests. Different degrees of resistance to the disease caused by this nematode species were observed among the genotypes tested. Complete resistance (no nematode egg mass formation up to 7 weeks after inoculation) was detected in one accession belonging to domesticated S. melongena species. Good levels of incomplete resistance were also observed in some commercial eggplant genotypes. This is apparently the first report of a source of complete resistance to M. javanica within the S. melongena gene pool and our finding could greatly facilitate the incorporation of this trait into commercial eggplant cultivars. S. torvum, previously described as resistant to several root-knot nematode species (including M. javanica, M. arenaria, and M. incognita) was also confirmed as a source of resistance to this Brazilian population of M. javanica. Therefore, S. torvum can be considered as one of the most promising sources of broad-spectrum resistance genes against root-knot nematodes. Broad-spectrum resistance is of paramount importance for breeding programmes focused on the development of cultivars for tropical and subtropical areas of the world where multiple infestation of soils with distinct Meloidogyne species is a very common feature.  相似文献

研究了白花曼陀罗(D a tura m etel L inn.)叶提取物对南方根结线虫(M eloid ogyne incogn ita Ch it-w ood.)生长发育的影响。结果表明,白花曼陀罗叶提取物对南方根结线虫胚胎发育和胚后发育有抑制作用。1%白花曼陀罗叶提取物处理48 h,仅有19.88%的卵完成胚胎发育,且变异卵增多;胚后发育历程、初侵染、初产卵日分别推迟2、4、6 d,雌虫产卵量降低。  相似文献

ABSTRACT

A pot experiment was carried out to investigate the tolerance of cucumber plants (Cucumis sativus L.) to root-knot nematode after inoculation with Glomus intraradices. Plants were inoculated with G. intraradices for four weeks and then transplanted in soil treated with Meloidogyne incognita for a further five weeks. The low phosphorus (P) loamy soil was amended with 50 and 100 mg P kg?1 soil. Mycorrhizal colonization increased shoot dry weight, shoot length, leaf numbers, root fresh weight and shoot P concentration, whereas nematode penetration and reproduction were significantly decreased. Similarly, P fertilization usually increased shoot growth and significantly decreased the number of galls and the number of egg masses and eggs per g root. Our results indicate that inoculation with G. intraradices and P fertilizer confer tolerance of cucumber plants to M. incognita by enhancing plant growth and by suppressing reproduction and/or galling of nematodes during the early stages of plant growth.  相似文献

拟禾本科根结线虫是严重影响水稻产量的植物寄生线虫之一,已成为制约中国及亚洲地区水稻产量的重大问题。拟禾本科根结线虫寄主范围广泛,可危害高地、低洼地和深水水稻;通过巨细胞从水稻根中摄取营养物质,从而在根系上形成许多弯钩状根结;其食道腺分泌物可以直接对水稻产生危害,影响水稻细胞代谢和免疫,从而影响水稻产量。目前水稻抗根结线虫病的品种较少,主要通过化学药剂在苗床进行防治。本文对拟禾本科根结线虫的寄主范围、危害特性、侵染特点、致病机制及防治方法等进行了综述,并对将来通过基因编辑等遗传工程技术提高水稻品种抗性的策略进行了展望,以期为我国水稻拟禾本科根结线虫的防治提供理论依据。  相似文献

【目的】研究南方根结线虫(Meloidogyne incognita)Me3毒性变异群体的分子标记,用快速有效的分子方法来鉴定毒性变异的发生。【方法】以南方根结线虫的无毒群体以及抗性基因Me3毒性群体为材料,用根据南方根结线虫基因组设计的100对引物和19对来自文献的引物进行扩增,筛选出在3个种群中扩增的差异条带,设计SCAR引物,并建立多重PCR反应体系。【结果】得到了7对能在3个群体稳定扩增出差异条带的多态性引物,把其中的2个位点开发为SCAR标记,能够区分开3个线虫群体。多重PCR反应体系在无毒群体和Me3毒性群体中分别扩增出1条999 bp和1条629 bp的条带,在混合群体扩增出999和629 bp的2条带。【结论】成功开发了对Me3表现毒性的南方根结线虫变异群体的分子标记,可用多重PCR体系一次性鉴定Me3毒性变异的发生。  相似文献

为明确云南省紫茎泽兰根结线虫病的病原种类,于2019年2月在云南省澜沧县林下三七种植区采集根部带有明显根结的紫茎泽兰根系进行根结线虫分离,通过观察所分离根结线虫的2龄幼虫、雌成虫、会阴花纹特征对其进行形态学鉴定,并利用序列比对、系统发育树分析、序列特异性扩增区段(sequence characterized amplified region,SCAR)对其进行分子生物学鉴定。结果表明,该病原线虫雌成虫会阴花纹呈圆形至卵圆形,背弓中等高或低平,侧区一侧或两侧延伸形成翼状,尾区有刻点,2龄幼虫、雌成虫形态特征及形态测量指标与北方根结线虫Meloidogyne hapla相似;该病原线虫rDNA的ITS序列和mtDNA的COI序列与NCBI数据库中已登录的北方根结线虫相应序列相似度较高,分别达99.35%和98.05%以上;该病原线虫rDNA的ITS序列、mtDNA的COI序列分别以99%、100%的支持率与北方根结线虫聚为同一分支;利用SCAR特异性引物,该病原线虫均能扩增出大小约1 500 bp的基因特异性条带。综合形态学和分子生物学鉴定结果将云南省紫茎泽兰根结线虫病病原种类鉴定为北方根结线虫。  相似文献
