Identification of Heat Tolerance Linked Molecular Markers of Chinese Cabbage (Brassica campestris L.ssp. pekinensis) 总被引:1,自引:0,他引:1
Genetically stable population of recombination inbred line (RIL) was derived from a cross between a heat tolerant line 177 and a heat sensitive line 276 of Chinese cabbage (Brassica campestris L. ssp.pekinensis ) by single seed descent. The RILs were analyzed using isozyme, RAPD and AFLP techniques in order to find molecular markers that are linked to heat tolerance quantitative trait loci (QTL). The results of variance analysis of single factor indicated that there were 9 molecular markers closely linked with heat tolerance QTL, including 5 AFLP markers, 3 RAPD markers and 1 PGM isozyme marker. Total genetic contribution of these makers to heat tolerance was 46.7%. Five of the nine markers distributed in one linkage group,the remaining 4 markers were located in separate groups. Thus the 9 heat tolerance linked markers distributed in 5 independent locations in the genome of Chinese cabbage. 相似文献

Heat treatments change the chemical and physical properties of wood and dimensional stability and hygroscopicity are affected
as a result of modifications of wood cell components. This study evaluated the water absorption of wood specimens treated
with boron compounds followed by heat treatment. Sugi (Cryptomeria japonica D. Don) sapwood specimens treated with either boric acid (BA) or disodium octoborate tetrahydrate (DOT) solutions were heat-modified
at either 180° or 220°C for 2 or 4 h. Carbohydrate composition and water absorption of the specimens were then measured and
compared with those of untreated and unheated specimens. Wood carbohydrates were significantly degraded in the specimens after
heat treatment. The heat treatment evidently decreased the water absorption and the heat-modified specimens absorbed less
water than unheated specimens. The higher the treatment temperature and the longer the treatment time, the lower the amount
of absorbed water. The boron-treated and heat-modified specimens, however, showed increased water absorption due to the hygroscopic

AIM:To study the role of ghrelin in cell protection by up-regulating heat shock protein 70 (HSP70) and inhibiting apoptosis induced by oxidative stress through extracellular regulated protein kinases 1/2 (ERK1/2) signaling pathway in the PC12 cells. METHODS:Sodium nitoprusside (SNP) was used to induce oxidative stress injury in the PC12 cells. The cultured PC12 cells were divided into SNP-injured group (incubated with SNP at 0.5 mmol/L for 6, 12, 18 and 24 h), ghrelin pretreatment group (ghrelin at 100 nmol/L was given 30 min before adding SNP); HSP70 inhibitor group (quercetin at 10 μmol/L was added 60 min before ghrelin treatment), ERK inhibitor group (ERK 1/2 inhibitor PD98059 was added 60 min before ghrelin treatment) and control group (added same amount of culture medium only). The apoptotic rate was detected by flow cytometry. The protein expression was determined by Western blot and immunocytochemistry. RESULTS:Compared with control group, the apoptotic rate of PC12 cells in SNP-injured group was significantly increased (P<0.05). Compared with SNP-injured group, ghrelin (100 nmol/L) pretreatment significantly inhibited SNP-induced apoptosis of PC12 cells (P<0.05), and significantly up-regulated the protein expression of HSP70 (P<0.05). Time-effect analysis showed that ghrelin had the most significant effect at 18 h after SNP injury. Quercetin, an inhibitor of HSP 70, significantly reduced the anti-apoptotic effect of ghrelin (P<0.05). Ghrelin pretreatment promoted the phosphorylation of ERK1/2. ERK1/2 inhibitor PD98059 significantly inhibited the effects of ghrelin on up-regulation of HSP70 expression (P<0.05). CONCLUSION:Ghrelin upregulates the expression of HSP70 and inhibits the apoptosis in the PC12 cells induced by oxidative stress by promoting the phosphorylation of ERK1/2. 相似文献

为了探讨微波杀菌对芒果原浆品质的影响,研究不同微波功率、处理时间、样品初始温度对芒果原浆菌落总数及维生素C含量、褐变度(BD)和色泽等品质指标的影响,采用正交试验优化微波杀菌工艺条件,并与巴氏杀菌工艺进行比较,分析杀菌前后芒果原浆菌落总数及感官品质的变化。结果表明,微波处理具有非常好的杀菌效果,且能抑制芒果原浆的褐变度、保护芒果原浆的营养成分和色泽。芒果原浆微波杀菌工艺最佳参数为:微波功率550 W,处理时间60 s,样品初始温度35℃,此工艺下菌落总数为50 CFU/m L,杀菌率达99.9%。与巴氏杀菌处理相比,采用微波杀菌不仅能达到更好的杀菌效果,而且原有品质保持良好。该杀菌工艺的研究为芒果原浆的贮存提供了理论依据。 相似文献

为全面了解苹果基因组中热激转录因子(Hsf)的序列特征及进化,采用生物信息学手段,在苹果全基因组水平上鉴定出50个MdHsf基因,并对其系统发育关系、序列特征、表达情况以及选择压力进行详细分析。系统发育与序列分析显示:与拟南芥和水稻相似,50个MdHsf基因可分为A、B、C 3个亚族;2个或多个MdHsf基因位于同一个末端进化支,说明该基因家族在苹果中发生了物种特异性扩增;尽管MdHsf基因的内含子数目和长度变异较大,但其蛋白的保守基序和功能结构域具有较高的保守性,这可能与功能约束有关。基于EST数目,可推知:除了MdHsfA2a和MdHsfA3a/b/c等14个基因没有相应的EST外,其余72%的基因都有转录活性。选择压力检测和结构建模分析显示:在36个MdHsf蛋白的选择压力检测中,位点模型未鉴定到正选择位点的存在;而在显著水平下(P0.05),分支-位点模型在d和e进化分支上,共检测到5个正选择位点,它们是28R、30L、35D、51M、67V,其中28R和30L位于Hsf结构域中,35D、51M和67V位于Hsf结构域之外,这说明除了MdHsfA4d/e和MdHsf C1a/b发生快速进化外,其他成员受控于纯净选择,具有高度保守性。综合以上研究结果,苹果基因组中存在多种热激转录因子,其蛋白的保守基序和功能结构域具有较高保守性,大多具有转录活性,在进化上该家族受纯净选择主导。 相似文献

为评价60Co-γ射线和电子束辐照灭菌对葛根提取物品质的影响效应及其差异性,采用不同剂量的60Co-γ射线(0、2.8、5.7、8.3、10.6 kGy)和电子束(0、2.4、5.1、7.7、10.3 kGy)辐照处理葛根提取物,研究辐照对其微生物总数、抗氧化活性及指纹图谱的影响。结果表明,60Co-γ射线和电子束辐照均能有效降低葛根提取物中需氧菌总数、霉菌和酵母菌总数,随着吸收剂量增大,抑制作用增强。60Co-γ射线和电子束辐照吸收剂量分别为5.7 kGy和5.1 kGy时,葛根提取物均达到《中国药典》所规定的微生物限度标准(需氧菌总数为103 CFU·g-1、霉菌和酵母菌总数为102 CFU·g-1);吸收剂量分别为5.7 kGy和7.7 kGy时,需氧菌总数、霉菌和酵母菌总数均降至10 CFU·g-1以下。此外,大肠埃希氏菌(Escherichia coli)与沙门氏菌(Salmonella)均未检出。2种辐照方式均对葛根提取物的DPPH自由基(DPPH·)清除率、羟自由基(·OH)清除率以及总还原能力无显著影响(P>0.05)。不同剂量60Co-γ射线和电子束辐照处理葛根提取物特征峰的相似度达到1.0,且相对保留时间的相对标准差(RSD)均小于0.5%,表明2种辐照方式对葛根提取物指纹图谱均无显著影响(P>0.05)。综上所述,60Co-γ射线和电子束辐照对葛根提取物微生物具有较强的抑制作用,2种辐照方式均不会对葛根提取物的抗氧化活性及指纹图谱产生显著影响,不影响葛根提取物的有效性及质量稳定性。该研究为2种辐照技术,特别是电子束辐照在葛根药材、饮片及其中成药制剂灭菌中的合理应用提供了科学参考。 相似文献

Heat stress(HS)can be detrimental to the gut health of swine.Many negative outcomes induced by HS are increasingly recognized as including modulation of intestinal microbiota.In turn,the intestinal microbiota is a unique ecosystem playing a critical role in mediating the host stress response.Therefore,we aimed to characterize gut microbiota of pigs’exposure to short-term HS,to explore a possible link between the intestinal microbiota and HS-related changes,including serum cytokines,oxidation status,and intestinal epithelial barrier function.Our findings showed that HS led to intestinal morphological and integrity changes(villus height,serum diamine oxidase[DAO],serum D-lactate and the relative expressions of tight junction proteins),reduction of serum cytokines(interleukin[IL]-8,IL-12,interferongamma[IFN-g]),and antioxidant activity(higher glutathione[GSH]and malondialdehyde[MDA]content,and lower superoxide dismutase[SOD]).Also,16S rRNA sequencing analysis revealed that although there was no difference in microbial a-diversity,some HS-associated composition differences were revealed in the ileum and cecum,which partly led to an imbalance in the production of short-chain fatty acids including propionate acid and valerate acid.Relevance networks revealed that HS-derived changes in bacterial genera and microbial metabolites,such as Chlamydia,Lactobacillus,Succinivibrio,Bifidobacterium,Lachnoclostridium,and propionic acid,were correlated with oxidative stress,intestinal barrier dysfunction,and inflammation in pigs.Collectively,our observations suggest that intestinal damage induced by HS is probably partly related to the gut microbiota dysbiosis,though the underlying mechanism remains to be fully elucidated. 相似文献

研究了γ射线辐照红枣的杀菌效果及其对主要有机营养成分、矿物质、维生素、氨基酸含量及感官品质的影响。试验结果表明:红枣辐照杀菌效果显著,经3×103Gy辐照处理后,5个红枣样品的微生物数量均达到国家标准;辐照对红枣中总糖、蛋白质、矿物质及VA、VE、VB1、VB2含量和感官品质无显著影响;辐照后红枣中粗脂肪含量和氨基酸总量比辐照前有所增加,但不显著(P>0.05);VC含量随辐照剂量的增加显著下降(P<0.01),当辐照剂量为3×103Gy时,其含量比对照降低了20.5%;辐照对红枣的感官品质不仅无不良影响,而且在一定程度上还有改善作用。提出了红枣辐照杀菌的适宜剂量为3×103Gy。 相似文献
