Soil incubations are often used to investigate soil organic matter (SOM) decomposition and its response to increased temperature, but changes in the activity and community composition of the decomposers have rarely been included. As part of an integrated investigation into the responses of SOM components in laboratory incubations at elevated temperatures, fungal and bacterial phospholipid fatty acids (PLFAs) were measured in two grassland soils contrasting in SOM quality (i.e. SOM composition), and changes in the microbial biomass and community composition were monitored. Whilst easily-degradable SOM and necromass released from soil preparation may have fuelled microbial activity at the start of the incubation, the overall activity and biomass of soil microorganisms were relatively constant during the subsequent one-year soil incubation, as indicated by the abundance of soil PLFAs, microbial respiration rate (r), and metabolic quotient (qCO2). PLFAs relating to fungi and Gram-negative bacteria declined relative to Gram-positive bacteria in soils incubated at higher temperatures, presumably due to their vulnerability to disturbance and substrate constraints induced by faster exhaustion of available nutrient sources at higher temperatures. A linear correlation was found between incubation temperatures and the microbial stress ratios of cyclopropane PLFA-to-monoenoic precursor (cy17:0/16:1ω7c and cy19:0/18:1ω7c) and monoenoic-to-saturated PLFAs (mono/sat), as a combined effect of temperature and temperature-induced substrate constraints. The microbial PLFA decay patterns and ratios suggest that SOM quality intimately controls microbial responses to global warming.  相似文献

稳定高效的多营养层级景观生态系统构建是近年来水族行业的发展热门,为探究海水水族箱在引入大型海藻后微宇宙生态系统结构和稳定性的变化,分别对不同处理组进行水质指标、浮游生物及细菌群落结构分析。结果表明:(1)有藻组水体溶解氧(DO)含量和pH上升趋势高于无藻组,并可维持亚硝态氮(NO2--N)含量在较低水平,实验结束时磷酸盐(PO43--P)及铵态氮(NH4+-N)含量显著低于无藻组(P < 0.05)。(2)有藻组细菌多样性及均匀度指数显著低于无藻组(P < 0.05);浮游生物群落均匀度指数存在显著组间差异(P < 0.05)。(3)细菌群落结构分析结果显示,无藻及有藻组第一、二优势菌门均为变形菌门(Proteobacteria)、放线菌门(Actinobacteria),无生物对照组分别为变形菌门和厚壁菌门(Firmicutes);各处理组浮游生物群落的第一优势门均为硅藻门(Bacillariophyta),至少占样品中浮游生物群落丰度的40%以上,第一优势属均为直链藻属(Melosira)。(4)CCA结果显示,无藻组微生物优势种丰度与营养盐含量呈正相关关系,与溶解氧(DO)及pH负相关,有藻组则相反。通过以上结果可推测,不同营养层级的微宇宙系统浮游生物及细菌群落组成不同,培养针叶蕨藻(Caulerpa sertularioides)能有效改善生态水族箱水质环境并影响水体微宇宙结构,有利于构建稳定高效并美观的海水景观生态系统。  相似文献

采集了鄂南不同母质和利用现状的6个红壤样,用2%PEG+6%Dextran两相分离技术(Aqueous two-phase partitioning technique,简写为A2PP)纯化细菌,测定细菌生物量,研究两相分离技术在土壤微生物研究领域的可应用性。结果表明:(1)采用0.1%胆酸钠、钠型离子交换树脂、玻璃珠与土壤一起在4℃下振荡2h,能较好地分散土壤细菌。供试土样细菌分离率介于0.41。0.60之间,不同母质发育的红壤相比,细菌分离率高低依次为:砂页岩〉花岗岩〉第四纪红色粘土;(2)A2PP技术能较好地纯化土壤中的细菌。6个供试原样的细菌多与土壤颗粒及有机质结合在一起,而两相分离技术能够得到较为纯净的细菌个体,土样细菌大多被分离存在于PEG相中,纯化率为63%~78%;细菌提取率介于0.31-0.48,不同母质发育土壤细菌提取率顺序与细菌分离率顺序相同;(3)供试土样的细菌形态都以小球状、小杆状细菌为主。  相似文献

[目的]研究病菌对植物的侵染对于培育高抗农作物新品种的重要意义。[方法]将病菌稀释液注入培养拟南芥幼苗的小瓶中,在不同时间监测植物的呼吸作用、光合作用及光系统Ⅱ的变化。[结果]病菌侵染拟南芥幼苗以后,呼吸作用速度普遍增加,呼吸作用速度增高能够提供更多的ATP以供植物抗病的需要。植物在受到病菌侵染以后也需要比平时更多的同化产物。此时需要光合作用速度提高,光合效率增加。但是病菌侵染以后,光合作用普遍降低。病菌侵染拟南芥幼苗以后,光系统Ⅱ的最大效率及通过光系统Ⅱ的光能所占光总量的比例普遍降低,光系统Ⅱ受到损伤,受到损伤的光合器官不能进行正常的光合作用。植物抗病对同化产物需要量的增加与植物光合器官受损伤而不能满足之间的矛盾最终会导致植物受侵染部位的坏死。[结论]病菌侵染拟南芥幼苗以后,光系统Ⅱ的最大效率及通过光系统Ⅱ的光能所占光总量的比例普遍降低,光系统Ⅱ受到损伤。  相似文献

Selected information was compiled from canine urinalyses and urine cultures conducted between January 1969 and December 1995. Eight thousand three hundred fifty-four microbial isolates (bacteria and fungi) included 4,873 isolates from females and 3,481 from males. Ten bacterial genera accounted for 96.3% of the urinary isolates, including Escherichia coli (44.1%), Staphylococcus spp. (11.6%), Proteus spp. (9.3%), Klebsiella spp. (9.1%), Enterococcus spp. (8.0%), and Streptococcus spp. (5.4%) as the 6 most common isolates in both genders of dogs. Among these 6 genera, female dogs were generally predisposed over males, although males had more urinary tract infections (UTIs) caused by Klebsiella spp. Distributions of ages at UTI diagnosis tended to be similar between genders. Infection with a single microbial species was responsible for >72% of UTIs in both genders. Among females, 40 breeds and a mixed-breed group represented 90.2% of all positive urine cultures, 88.4% of the individual dogs with UTIs. and 88.2% of the microbial isolations. Among males, these same 41 breed groups represented 87.9% of all positive urine cultures, 87.6% of the individual dogs, and 88.2% of the microbial isolations.  相似文献

We performed a comparative experiment to investigate: (1) how the ubiquitous soil bacterium Bacillus subtilis weathers granite; and (2) which granite-forming minerals weather more rapidly via biological processes. Batch system experiments (granite specimen in a 500 ml solution including NaCl, glucose, yeast extract and bacteria B. subtilis at 27 °C) were carried out for 30 days. Granite surfaces were observed by SEM before and after the experiment. B. subtilis had a strong influence on granite weathering by forming pits. There were 2.4 times as many pits and micropores were 2.3 times wider in granite exposed to B. subtilis when compared with bacteria-free samples. B. subtilis appear to preferentially select an optimum place to adhere to the mineral and dissolve essential elements from the mineral to live. Plagioclase was more vulnerable to bacterial weathering than biotite among the granite composing minerals.  相似文献

从广东省揭西县坪下牛蛙养殖场收集患黄杆菌病的病蛙,其症状是歪头、狂游、双眼白内障、肝脏肿大充血,腹水严重。从病蛙脑髓、眼底等处分离到62菌株。经急性毒性试验证明分离菌株对健康牛蛙有较强的致病性。选用其中有代表性的981229—1等菌株进行系统的生理生化试验结果显示,981229—3等菌株革兰氏染色阴性、短直杆菌、无动力、无芽孢、好氧型、不利用葡萄糖。内毒素试验阳性。经VITEK-AMS-60自动化检测系统鉴定为脑膜炎脓毒性黄杆菌(Flavobacterium meninosepticum)。选取47种抗菌药物,通过K—B纸片扩散法对分离菌进行耐药性试验结果显示,病原菌对40种试验药物耐药(85.11%)、中等敏感药物2种(4.26%)、敏感药物5种(10.64%)。以试管稀释法、测定分离菌株对5种敏感药物的平均 MIC分别是:利福平 0.24、环丙沙星 0.70、氧氟沙星 0.70、氟啶酸 1.20和麦迪霉素7.80μg/ml。  相似文献

为研究日粮中添加不同纤维分解菌对鹅生产性能的影响,试验选用0日龄健康农安本地鹅64只,随机分成4组,每组16只。试验组从0日龄每3 d饲喂一次纤维分解菌,喂菌量为109 CFU/mL,纤维分解菌分别为:从鹅肠道内分离扩增的芽孢纤维分解菌(A组);从白蚁肠道内分离扩增的芽孢纤维分解菌(B组);从牛粪中分离扩增的芽孢纤维分解菌(C组);对照组用等量的蒸馏水代替。测定鹅日增重、采食量、料重比。结果表明,A、B、C组提高了鹅的日增重、采食量,显著降低了料重比,B组作用效果较好、A组次之,C组较差。  相似文献

The exotic earthworm invasion in hardwood forests of the northern United States is associated with many ecosystem-level changes. However, less is known about the effects of the invasion on the composition of the soil microbial community through which ecosystem-level changes are mediated. Further, earthworm effects on soil microbial community composition have not been well studied in the field. To evaluate changes in bacterial and fungal abundance associated with the earthworm invasion we quantified bacterial and fungal biomass by microscopic counts in paired earthworm-invaded (earthworm) and earthworm-free (reference) plots in five forest stands in central New York (USA). Earthworms significantly increased the ratio of bacteria to fungi on an area basis (per m2), by more than two times in mid-summer and early autumn. While this effect was associated primarily with the lack of the fungal-dominated organic horizon in earthworm plots, a higher ratio of bacteria to fungi in the surface 5 cm mineral soil also contributed as it developed between spring and mid-summer. Earthworm reduction of fungal biomass was confirmed by substantially lower growth of fungal hyphae into mesh sand bags in earthworm compared to reference plots. Burrowing activity by the earthworm Lumbricus terrestris increased the ratio of bacteria to fungi over the short-term within earthworm plots, introducing small-scale spatial heterogeneity associated with burrows. Our study suggests that the exotic earthworm invasion in these northern hardwood forests markedly increased the ratio of bacteria to fungi by eliminating the fungal-rich organic horizon, and was associated localized increases in bacterial vs. fungal abundance in mineral soil, setting the stage for future research into linkages between the earthworm invasion, bacterial and fungal abundance, and ecosystem processes.  相似文献
