棉花新品种皖棉27号选育报告

介绍了皖棉27号的选育经过及其特征特性;阐述了其丰产稳产性以及优良的纤维品质;并介绍了其栽培技术要点。     

微生态制剂SC27对作物生长和土壤环境的影响

通过对大麦、水稻上的SC27的不同施用方法、不同施用量的田间小区试验，观察SC27对作物的产量、品质、生物性状及对土壤N、P、K利用率的影响，表明了SC27能提高化肥肥效，减少化肥环境污染；改善作物根系生长环境，调控土壤养分供应，大麦和水稻产量提高20％左右。     

搅拌参数对ZA27合金半固态组织与性能的影响

在连续冷却条件下以不同的参数对 ZA2 7合金熔体进行了电磁搅拌处理 ,并分析了搅拌参数对组织形貌及力学性能的影响。结果表明 ,最佳搅拌时间可选定在 5～ 15 m in之间 ,在连续缓冷条件下 ,半固态 ZA2 7的初生相随搅拌强度提高而细化。     

Effect of ClC-3 siRNA on cell cycle of HeLa cells

AIM: To investigate the roles of ClC-3 chloride channels in the regulation of cell cycle and the relationship between ClC-3 chloride channels and the cell cycle regulators, such as cyclin D1, cyclin-dependent kinase (CDK)4, CDK6, P21 and P27 in the HeLa cells.METHODS: ClC-3 genes were silenced by the siRNA technique in the HeLa cells. The transfection efficiency of ClC-3 siRNA was detected by real-time PCR. The cell cycle distribution was analyzed by the flow cytometry. The protein expression of ClC-3, P21, P27, CDK4, CDK6 and cyclin D1 was determined by Western blot.RESULTS: ClC-3 was knocked down by ClC-3 siRNA in the HeLa cells. Transfection of the cells with ClC-3 siRNA arrested the cells at G₀/G₁ phases, decreased the expression of cyclin D1, CDK4 and CDK6, and increased the expression of P21 and P27.CONCLUSION: ClC-3 plays an important role in the cell cycle of HeLa cells through the G₁-S transition point. ClC-3 may regulate the cell cycle progression by up-regulation of cyclin D1, CDK4 and CDK6 expression and/or by down-regulation of P21 and P27 expression.     

家蚕泛肽-核糖体蛋白S27a的表达及功能研究

泛肽(UB)-核糖体蛋白S27a(UBRPS27a)是泛肽和核糖体蛋白的融合蛋白,在真核细胞中表达时,被酶解成UB和核糖体蛋白S27a(RPS27a)。从家蚕蛹cDNA文库中获得了该基因并原核表达了该蛋白,命名为BmUBRPS27a。将纯化的融合蛋白免疫新西兰兔得到抗血清,通过亚细胞免疫荧光定位发现该蛋白存在于细胞核和细胞质。用荧光定量PCR对该蛋白在家蚕中的表达作了初步分析,发现RPS27a在活性增殖细胞中表达量很高。将合成了BmUBRPS27a的dsRNA进行RNAi,用脂质体转染家蚕卵巢上皮细胞Bm5后,细胞的形态发生了变化:细胞由圆形、椭圆形变成了梭形,甚至出现类似神经细胞的突触结构。该基因对维持细胞正常形态具有重要的作用。     

Effect of Different Test Materials on the Detected Results of Avian Leukosis Virus-p27 Antigen by ELISA

In order to study the effect of different test materials on the detected results of avian leukosis virus (ALV)-p27 antigen by ELISA, the cloacal swabs ALV-p27 antigen were detected by ELISA in 180-day local breeds hen, and the parts of the positive and negative chicken was selected to group test, the serum, egg whites and cell supernatant ALV-p27 antigen were detected by ELISA, the specific genes of ALV were detected by RT-PCR in blood.The results showed that serum, egg white and cell supernatant as ELISA test materials, the positive rate lower of than cloacal swabs, and serum ALV-p27 positive samples include all egg whites and cell supernatant positive samples in positive group.It was a significant correlation between ELISAs with serum and cell supernatant (linear equation:Y=1.8439X-0.1469, the correl was 0.937).In positive group, ALV-p27 gene positive rate lower than cloacal swabs ELISA, but higher than the serum, egg and cell culture medium, and ALV-p27 gene positive samples include serum positive samples by ELISA.ALV-J gp85 gene positive rate of 29.17%, and all positive samples were included in the serum ALV-p27 positive samples.The results suggested that the cloacal swabs as test material may occur false positive results, and egg whites and cell supernatant may occur undetected by ELISA ALV-p27 antigen assay in adult chicken, serum as test material in ELISA could more accurately reflect the state of adult chickens infected with ALV.     

抗锈高产种质汕油27在花生育种上的应用

 汕油27是汕头市农业科学研究所育成的中抗锈病、高产稳产、适应性广的花生新品种，由该品种衍生的许多品种在不同时期成为中国南方花生产区的当家种或推广良种。为了了解该优良种质在花生新品种选育中的作用，剖析了以汕油27作为直接或间接亲本选育的后代优良品种的系谱、遗传特性以及与汕油27的遗传关系，结果表明，由汕油27作为直接亲本先后衍生出优良花生新品种5个，进而由这些品种又衍生出的通过审定（或在省级区试中表现较突出）的花生新品种17个；在这22个优良衍生品种中，通过国家审定的有6个，包括粤油223、粤油79、粤油7号、粤油9号、汕油21和湛油30；所有衍生品种都保持了汕油27高产、抗锈病等优良特性。因此，作者认为汕油27是一个一般配合力较高的优良中心亲本；选准中心亲本对提高花生育种效率具有十分重要的作用。     

Proteomic analysis of anti-tumor effects of 11-dehydrosinulariolide on CAL-27 cells

The anti-tumor effects of 11-dehydrosinulariolide, an active ingredient isolated from soft coral Sinularia leptoclados, on CAL-27 cells were investigated in this study. In the MTT assay for cell proliferation, increasing concentrations of 11-dehydrosinulariolide decreased CAL-27 cell viability. When a concentration of 1.5 μg/mL of 11-dehydrosinulariolide was applied, the CAL-27 cells viability was reduced to a level of 70% of the control sample. The wound healing function decreased as the concentration of 11-dehydrosinulariolide increased. The results in this study indicated that treatment with 11-dehydrosinulariolide for 6 h significantly induced both early and late apoptosis of CAL-27 cells, observed by flow cytometric measurement and microscopic fluorescent observation. A comparative proteomic analysis was conducted to investigate the effects of 11-dehydrosinulariolide on CAL-27 cells at the molecular level by comparison between the protein profiling (revealed on a 2-DE map) of CAL-27 cells treated with 11-dehydrosinulariolide and that of CAL-27 cells without the treatment. A total of 28 differential proteins (12 up-regulated and 16 down-regulated) in CAL-27 cells treated with 11-dehydrosinulariolide have been identified by LC-MS/MS analysis. Some of the differential proteins are associated with cell proliferation, apoptosis, protein synthesis, protein folding, and energy metabolism. The results of this study provided clues for the investigation of biochemical mechanisms of the anti-tumor effects of 11-dehydrosinulariolide on CAL-27 cells and could be valuable information for drug development and progression monitoring of oral squamous cell carcinoma (OSCC).
