The goal of this study was to develop and characterize a cell line from the caudal fin tissue of zebrafish and also its application as an in vitro model to study the effect of H2O2 in wound healing. Fibroblastic cell line was developed using explant culture method from caudal fin tissue of zebrafish and characterized. This cell line was named as DrF cell line. The DrF cells treated with 0–10 µM/ml H2O2 were tested for viability, proliferation and motility by MTT assay, trypan blue assay and chemotaxis assay, respectively. Among the different concentrations of H2O2, 4 µM was found to be nontoxic to study cell migration in in vitro scratch wound assay. Furthermore, the expression of proliferating cell nuclear antigen (PCNA) and chemokine receptor (CXCR4) genes was carried by qPCR. The cell survival, proliferation and migration were extremely enriched at 4 µM level of H2O2. We observed accelerated wound closure in DrF cells treated with H2O2. The qPCR results indicated that H2O2 markedly up‐regulated mRNA expression of PCNA and CXCR4. The findings from our study suggest that H2O2 at low levels promotes cell survival, proliferation, migration and wound healing in DrF cells.  相似文献

在猪场中,正确的伤口处理对于伤口的恢复具有重要的意义。为了研究新型伤口处理剂对仔猪去势伤口的效果,特选取5~7日龄533头公仔猪,分为两组。试验组利用常规碘酊消毒去势后喷洒新型伤口处理剂,对照组则按常规操作单纯碘酊消毒。而后分别记录每天伤口恢复情况以及去势后15 d内表现关节肿的去势公仔猪比例。结果表明:新型伤口处理剂可以减少伤口出血,促进去势伤口的愈合,减少细菌通过伤口感染猪只的几率。  相似文献

The storage roots of cassava (Manihot esculenta Crantz) suffer from a rapid post-harvest deterioration that is a major constraint to their increased exploitation. In many ways this deterioration resembles wound responses in other better studied plant systems, though it appears to lack an adequate wound repair response. A cDNA clone (cMeHRGP1) for a hydroxyproline-rich glycoprotein expressed during the deterioration response was isolated and characterised. This clone proved to be an antisense pairing, coding for part of phosphoserine aminotransferase on its complementary strand. Messenger RNA corresponding to cMeHRGP1 accumulated in deteriorating cassava roots from day three after harvest, by which time the deterioration response was well advanced. There by confirming that aspects of the wound repair response were inadequate in harvested cassava roots. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The performance of laboratory X‐ray computed tomography (XCT) for the non‐destructive imaging of root wood was evaluated. Lateral roots of oriental cherry (Prunus serrulata var. spontanea) and Japanese zelkova (Zelkova serrata) were severed in spring and maintained in soil for 6 months. Without sectioning, XCT revealed the phloem, xylem and vascular cambium structures in the root wood. A virtual transverse section showed a ring of woundwood covering the severed, lateral root of the two trees. Different levels of X‐ray absorption were evident around the cut surfaces of P. serrulata; however, they were rarely detected in Z. serrata. More adventitious roots were observed on Z. serrata than on P. serrulata. Distinct white spots in the rays were only detected in Z. serrata. These results suggest that XCT has potential applications in forest pathology, providing virtual sections of wound closure, wood density distribution, organ redifferentiation, and mineral deposition in root wood.  相似文献

针对轴流泵在输送污水介质中的磨损和缠绕问题,设计了外特性相同但后掠角分别为40°和60°后掠叶片,并采用Particle颗粒模型进行固液两相流数值模拟,发现设计流量工况下60°后掠叶片固相分布情况要优于40°后掠叶片,60°后掠叶片压力面上的固相体积分数平均比40°后掠叶片上的固相体积分数小0.1,60°后掠叶片吸力面上的固相体积分数平均比40°后掠叶片小0.2。进一步对60°后掠叶片进行研究,发现随着颗粒直径的增加,叶片上的固相体积分数随之增加,且固相集中的区域都很相似;随着初始颗粒体积分数的增加,60°后掠叶片上的固相体积分数也随之增加,但初始颗粒体积分数越大,对后掠叶片压力面上固相体积分数的影响越小。为检验后掠叶片的抗缠绕能力,对60°后掠叶片进行缠绕试验,发现单独的后掠叶片形式的轴流叶轮不易发生缠绕,但当叶轮与套筒配合后,若面对大量棉线,容易在进口边轮缘处发生堆积。该研究为输送污水介质轴流泵的抗磨损和抗缠绕性能的研究提供了参考。  相似文献

This review discusses the structure of integrins as well as the various roles that integrins play in the corneal wound healing process. Objective To summarize the medical knowledge surrounding aardvarks to date, to describe the ophthalmic examination of a specimen with a chronic history of an upper eyelid lesion, of an assumed blind left eye, and to detail the anesthesia procedure performed. Procedure A 23‐year‐old aardvark was examined under general anesthesia and multiple ocular abnormalities were detected in the left eye (globe deviation, corneal opacities, iridodonesis, and aphakia). A thickening of the palpebral conjunctiva affecting the medial upper eyelid with erosion of the normal eyelid margin anatomy was identified. The adnexal lesion was resected by a wedge resection and histopathology was performed. Suture breakdown 3 days postoperatively required a second surgery, where buried sutures were used. The surgical techniques and postoperative care are discussed. Results The histopathology revealed mucosal hyperplasia and moderate neutrophilic and lymphoplasmacytic blepharitis. No causal organisms were identified. Following initial wound dehiscence and a modified surgical technique, the upper eyelid healed without complication and retained complete function. Conclusions The eyelid lesion involved a benign inflammatory and hyperplastic pathology of unknown etiology. Adjusting routine ophthalmic surgical techniques to wildlife and zoo animals can be challenging and complicated. It is important to understand the nature of the animals being managed, their circadian cycle, and habitat, to adjust and individualize the surgical approach, instrumentation, suture material, and perioperative treatment.  相似文献

Purpose To study the effects of topical administration of 1% morphine on corneal analgesia in rabbits submitted to lamellar keratectomy and to assess the expression of matrix metalloproteinase‐1, metalloproteinase‐2, metalloproteinase‐9 (MMPs), type IV collagen, and interleukin‐10 (IL‐10) during the treatment. Methods Morphine group (MG) received 50 μL of topical 1% morphine four times daily, while the control group received saline instead. Corneal touch threshold (CTT) and the wound area were assessed until corneal healing. Corneal samples were processed for routine histology, immunohistochemistry, zymography, and ELISA. Results Following keratectomy, CTT increased significantly from 6 to 96 h time points. Mean corneal re‐epithelization rate and scores of leukocyte infiltration did not differ significantly between treatment groups. Immunolabeling pattern for MMP‐1, MMP‐9, and type IV collagen was similar in both treatment groups. In the MG, zymography indicated significantly higher levels of active MMP‐2 on days 6 and 12; and in the latent MMP‐9, on days 3 and 6, and in the active MMP‐9, on day 6. Latent MMP‐2 and MMP‐9, and active MMP‐9 decreased to values close to those of healthy corneas on day 12, but levels of active MMP‐2 remained significantly elevated in the MG. IL‐10 levels measured on days 1–6 were reduced as compared to those of healthy corneal tissue and returned to levels close to those of healthy corneas on day 12. Conclusion Topical morphine promoted corneal analgesia for up to 4 days and did not delay corneal re‐epithelization. The re‐establishment of MMPs and IL‐10 to levels close to baseline values at the end of the study and the expression of type IV collagen in both groups reinforce that, with caution, 1% morphine can be used after lamellar keratectomy in rabbits.  相似文献
