Genotypic diversity in Fusarium pseudograminearum and F. graminearum from Australia and the relationship between diversity and pathogen aggressiveness for head blight and/or crown rot of wheat were examined. Amplified fragment length polymorphism (AFLP) analysis revealed a high level of genotypic diversity within each species. Sixty-three of the 149 AFLP loci were significantly different between the two species and 70 of 72 F. pseudograminearum and 56 of 59 F. graminearum isolates had distinct haplotypes. When head blight and crown rot severity data from a recently published work on isolates representing the entire range of aggressiveness were used, only the genotypic diversity of F. pseudograminearum was significantly associated with its aggressiveness for the two diseases. Cluster analyses clearly demonstrated the polyphyletic structures that exist in both pathogen populations. The spatial diversity within F. graminearum was high within a single field, while frequent gene flow ( N m ∼ 14) and a low fixation index ( G st = 0·03) were recorded among F. pseudograminearum isolates from the adjacent states of New South Wales and Queensland. The differences in population structure between the heterothallic F. pseudograminearum (teleomorph G. coronicola ) and the homothallic F. graminearum (teleomorph G. zeae ) were not as pronounced as expected given their contrasting mating systems. Neither species was panmictic or strictly clonal. This points to sexual recombination in F. pseudograminearum , suggesting that ascospores of G. coronicola may also play a role in its biology and epidemiology. 相似文献

To develop an alternative method to feed withdrawal for molting layers, 2 flocks consisting of approximately 26,000 commercial laying hens each at 478 (68 wk, flock 1) and 466 (67 wk, flock 2) d of age were reared in an environmentally controlled windowless house and were fed wheat bran (WB) diet. Flock 1 hens were fed WB for 25 d, and flock 2 hens were fed WB for 21 d and then fed a mixture of WB and layer feed (1:1, wt:wt) for the last 4 d of the treatment. After that, the birds in both flocks were fed a normal layer feed. The photoperiod was reduced from 16 to 9 h in both flocks. Most of the birds in both flocks ceased egg production by 10 to 15 d of feeding the WB diets. Egg production in flock 1 gradually increased to 11.4% by 31 to 40 d and 71.4% by 41 to 50 d of the treatment, whereas the egg production in flock 2 hens lagged behind by almost 10 d. The mean egg production from 61 to 140 d exceeded 86% in both flocks. The houses in the farm were naturally contaminated with several serovars of Salmonella, not Enteriditis or Typhimurium. In both flocks with the WB treatment, no marked increase in Salmonella isolation from environmental samples was observed postmolt relative to premolt levels. The study demonstrated that feeding hens WB could be successfully used as an alternative to feed withdrawal to force-rest aging hens while not exacerbating a Salmonella problem in a commercial egg-production setting. 相似文献

An experiment was conducted to determine true and apparent ileal amino‐acid digestibility of a native cultivar of wheat (Mahdavi), autoclaved wheat (120°C for 30 min) and spaghetti by‐products available in Iran. One hundred 21‐day‐old broiler chickens were fed a standard corn–soybean meal starter diet from day 0 to 28 post hatch. At 28 days 80 chicks were distributed according to possessing very nearly the same average bodyweight to 20 wire cages. The experimental units were allocated at random to five dietary treatments with four replicates per treatment. The basal diet contained corn and soybean meal as the major ingredients. Three test diets were formulated containing wheat, autoclaved wheat and spaghetti by‐products as the sole source of dietary protein and each test diet was combined with a basal diet 50:50 on a weight basis to form three assay diets. The apparent and true digestibility of amino acid in the test ingredients were estimated from those in the assay diets basal/test diet mixtures, using the difference method. The apparent and true amino‐acid digestibility of the test ingredients were significantly different (P < 0.01). Autoclaving of wheat increase its amino‐acid digestibility (P < 0.01). Among the test ingredients, the average ileal amino‐acid digestibility of spaghetti by‐products was higher than wheat and autoclaved wheat but it was lower than values for the basal diet (P < 0.01). 相似文献

A cpDNA fragment of 34 genotypes belonging to Citrus and four related genera was amplified and sequenced. Chloroplast microsatellites were revealed with the length of repeats ranging from 25 to 44 bases. Other than the normal uninucleotide poly(A) repeats, a trinucleotide poly(TAA) motif was also found, the first report of such repeats in a plant chloroplast genome. According to SSR structure variations, 18 Chloroplast SSR Types (CST) were identified. The CST sequences were informative for better understanding the genetic relationships of chloroplast genomes among the analyzed genotypes and confirmed some previous hypotheses about the female parent of several hybrid accessions. 相似文献

以两个抗旱性不同的小麦品种西农1043、陕253为材料,采用盆栽与水培试验相结合的方式,研究水分亏缺对小麦不同生育期叶片光合及根呼吸速率等生理指标的影响。结果表明:相同水分处理下,抗旱品种西农1043与水分敏感品种陕253相比,具有较高的光合和根呼吸速率、以及相对较高的保护酶活性;水分胁迫下,两个小麦品种叶片光合和根呼吸速率及苗期叶保护酶活性均降低;两种水分处理下抗旱性强的西农1043根呼吸所占光合的比例相对高于陕253。 相似文献

A lesion-mimic mutant was obtained from a mutagenic treatment performed with ethyl methanesulfonate on the Argentine bread wheat ( Triticum aestivum ) cultivar Sinvalocho M.A. The HLP (hypersensitive-like phenotype) mutant exhibited tiny, discrete, white lesions in the absence of any pathogen, resembling the typical hypersensitive response (HR). The lesions only became evident once the fifth or sixth leaf emerged, and spread at random along the leaf blades and leaf sheaths of the developing plant, including tissues of the spike. Because the lesion-mimic mutant showed no lesions at the seedling stage, the phenotypes of both the mutant and its mother line were identical at this point. Histochemical studies showed that spontaneous hypersensitive-like lesions in the HLP mutant corresponded to cell death. In leaf-rust ( Puccinia triticina ) infection experiments performed at seedling and adult-plant stages, adult HLP plants showed enhanced resistance to leaf-rust attack compared with plants of Sinvalocho M.A. of comparable developmental stage, suggesting that the HLP mutation may confer increased resistance to the fungus. Because enhanced resistance coincided with the presence of spontaneous HR lesions, activation of HLP plant defence responses appeared to be tightly linked to this phenomenon. Final plant height and yield components in the lesion-mimic mutant did not differ from those of the mother line, indicating that the HLP mutation caused no detrimental pleiotropic effects that significantly affected agronomic performance.
