The modernization has been the dream that Chinese nation has wished eagerly to realize all the time. In some sense, the modernization of China means urbanization. The reform and opening - up over the past 20 years , built and made the enormous achievement in urbanization of China, have changed the appearance of the city of China greatly. But to numerous traditional historical zones, what kind of urbanization way should on earth be taken, it has been the question long - term in dispute of the planning circles all the time. The 99% of all microorganisms in naturecannot be isolated by standard culture methods without conspicuous external characters to classify them morphologically. In addition, classification based on physiological or biochemical features are often fuzzy. Molecular techniques are used for uncultured microorganisms to explore the diversity of microbial communities and novel resources without the need of cultivation. DNA dynamics and cloning of PCR products obtained from environmental DNA is now routinely used to identify microbial populations in microbial ecology. For practical reasons the approach is not suited to study the complex dynamics of microbial communities in environment. Genetic fingerprinting techniques are better tools, which give complement for the traditional microbiological methods. The methods are rapid and relatively easy to perform, and they allow the simultaneous analysis of multiple samples, which makes it possible to compare the genetic diversity of microbial communities from different habitats, or to study the behaviour of individual communities over time.  相似文献

The inability to properly maintain irrigation systems over time forced the Government of Indonesia (GOI) to seek cost recovery from water users through introduction of an irrigation service fee. The plan is to introduce this fee in all of the technical irrigation systems of Indonesia, covering about 4 million hectares, over a 12 year period. Design and Introduction of this service fee in 4 pilot areas of the 4 major rice producing provinces (West, Central and East Java, and South Sulawesi) during 1989–1991 has shown that users are willing to pay. Results of first ISF collection from over 11,000 farmers in Central and East Java in pilot systems was an encouraging 95%. However that acceptance to pay by users depends on their structured and systematic involvement in defining systems needs. They need to understand a differentiation in payment if service levels are different. They appreciate use of collected funds in the system where collected. They understand the organization of water users associations but this requires their active role in fee determination and collection. They understand the establishment of federations of these associations. ISF requires also active involvement of Local Government officials as facilitators and intermediaries between service receivers (the water users) and service providers (personnel of the Public Works irrigation department) in the introduction period. Vegetative compatibility (vc) and mating types and genetic diversity of Cryphonectria parasitica isolates were determined using 183 isolates obtained from 215 infected chestnut trees growing in 13 provinces of Turkey. Based on the cultural aspects, 143 of these isolates were evaluated as virulent whereas the remaining 40 isolates were hypovirulent. When vc types of 183 isolates were classically differentiated, 135 of them matched to EU-1 (82.3%), 29 of them to EU-12 (17.6%) vc type, whereas 19 of them did not match to the two. When molecular vic markers were used, all the isolates were assigned to two EU vc types; 149 to EU-1 (81.4%) and 34 (18.5%) to EU-12. Of the majority of the isolates, 134 (73.2%) had mating-type MAT-1, while 44 (24%) isolates had MAT-2 and 5 (2.8%) isolates had both mating types. The population analysis based on two DNA marker systems, Inter-Primer Binding Site and Start Codon Targeted Polymorphism, showed no intraspecific genetic variation among the C. parasitica isolates. The prevalence of two dominant vc types revealed by this study shows that biological control with hypovirulent EU-1 and EU-12 isolates will be significant for the country. The results might be helpful to chestnut breeders carrying out resistance breeding studies to manage this disease based on hypovirulence attributed to Cryphonectria hypovirus 1.  相似文献

以园林植物石楠为试材,通过混合2种中性盐(NaCl和Na2SO4)和2种碱性盐(NaHCO3和Na2CO3)分别模拟不同强度的盐、碱胁迫条件,对园林植物(石楠)进行3年(2015—2017年)的胁迫处理,以期探讨盐碱胁迫对园林植物土壤酶活性和微生物的影响。结果表明:1)土壤中有机酸含量随着盐碱浓度的增加呈逐渐降低趋势。随胁迫强度增加,盐、碱胁迫均使土壤中Na+浓度大幅度上升,K+浓度大幅度下降,但碱胁迫下Na+浓度上升幅度和K+浓度下降幅度均大于盐胁迫。2)随着盐碱浓度的增加,土壤中氨基酸类、碳水化合物类、羧酸类、聚合物类、胺类、酚酸类碳源呈逐渐降低趋势,总体而言,碳水化合物和羧酸类碳源是盐碱胁迫下园林植物土壤微生物的主要碳源,其次为氨基酸类、酚酸类和聚合物类,胺类碳源的利用率最小。3)土壤微生物物种丰富度指数(H)、均匀度指数(E)和碳源利用丰富度指数(S)、土壤微生物量碳、微生物呼吸和微生物代谢熵随盐碱浓度的增加而增加。4)土壤糖苷酶、几丁质酶、亮氨酸氨基肽酶、碱性磷酸酶、酚氧化酶δ和过氧化物酶δ随盐碱浓度的增加呈逐渐降低趋势。5)交互作用分析显示,盐胁迫、碱胁迫、盐胁迫×碱胁迫对有机酸、Na+、K+、Na+/K+、物种丰富度指数(H)、碳源利用丰富度指数(S)、土壤微生物量碳、土壤微生物呼吸、土壤微生物代谢熵、糖苷酶和碱性磷酸酶具有显著的影响(P<0.05),其中碱胁迫对土壤酶活性和微生物群落多样性的影响高于盐胁迫,并且盐胁迫×碱胁迫的影响高于单独盐胁迫或碱胁迫.  相似文献

为了研究养殖刀鲚冰鲜保存过程中细菌群落组成及变化情况,采用Illumina Miseq高通量测序方法对冰鲜条件不同贮藏时间(0、3、6、9、12、15、18 d)刀鲚肌肉样本细菌的16S rRNA基因的2个高变区(V3—V4)进行测序,比较其细菌多样性及门、属水平的细菌相对丰度,并进行主成分分析、Heatmap分析以及特定腐败菌分析。结果显示,7组样品细菌分布于40门787属,冰鲜条件下刀鲚肌肉的优势致腐败菌属为假单胞菌、微杆菌属和气单胞菌属。主成分分析显示,刀鲚腐败前后的微生物结构存在差异。Heatmap图分析显示新鲜样本的细菌丰度最大。3组冰鲜贮藏6 d前的样品群落结构较相似。贮藏12 d时常见致腐菌(微杆菌属、假单胞菌属、气单胞菌属)占比显著提高,其细菌的多样性及丰富度最低。随着冰鲜保存时间的延长,刀鲚肌肉的细菌菌落结构产生了动态变化。冰鲜保存6~9 d后刀鲚肌肉的细菌菌落结构变化较大,研究结果可以为预测冰鲜刀鲚货架期提供参考。  相似文献

以香港大学城市环境与人类健康实验室这一新兴学术共同体作为研究切入点,使用文献综述与案例研究的方法深入探讨学术共同体在推动风景园林学科的发展和进步中扮演的关键角色。研究结果发现:该实验室在以往的风景园林学科内容、框架和知识体系基础上,运用新兴技术并结合定量的统计学方法,将城市环境与居民健康紧密相连,为快速城市化地区的环境和人类健康问题提供高效、精准、人文关怀且切实可行的解决方案,并极大地拓展了风景园林学科的研究领域;实验室的核心关注点在于城市感知:探究城市环境特征如何以及在何种程度上通过心理、生理等途径影响人类健康;在健康城市领域,这支先锋团队担任着理论研究、技术探索以及教育者的角色。  相似文献

以南方地区的蓝莓品种为主要对象,利用SSR标记分析现有品种种质的遗传多样性。设计合成了56对蓝莓SSR分子标记引物,进行扩增和多态性引物筛选,将多态性好的引物用于66份蓝莓不同类型种质的遗传多样性分析。结果表明,56对引物均能扩增出预期大小条带,其中条带清晰多态性较好的引物为25对,占比引物数的44.64%,多数引物扩增的位点数在5~7个之间。25对引物在66份蓝莓供试种质中检测多态性,共检测到165个等位变异,平均每个位点有6.6个等位变异,PIC值变幅为0.656~0.947,平均值为0.777。多态性最好的引物Vc26可以检测到15个等位基因数目。北方高丛、南方高丛和兔眼类型66份蓝莓种质的遗传相似系数位于0.60~0.96之间,品种间遗传差异较丰富。利用UPGMA聚类将3种类型种质进行分析,发现除了兔眼类型品种‘红粉佳人’外,其余明显分为3个类群,且同一类型的种质基本聚在一起。结合不同类型种质的系谱分析,发现分类与品种种质的遗传成分相似有一定的相关。研究利用SSR分子标记揭示了蓝莓不同类型种质的遗传多样性,对今后杂交育种亲本选择利用有一定借鉴意义。  相似文献
