双生病毒是一类在全球范围内危害严重的单链环状DNA病毒。本研究从2017年采自云南的胜红蓟样品中(YN2017)获得了一条菜豆金色花叶病毒属病毒和一条β卫星的全基因组序列。该双生病毒的全长序列与烟草曲茎病毒的相似性最高,为99.60%,确定为烟草曲茎病毒的分离物。YN2017β卫星与中国胜红蓟黄脉β卫星的同源性最高,为90.8%。进一步分析发现,该β卫星的卫星保守区域(SCR)至富含腺嘌呤区(A-rich区)之间约1 kb的序列与中国胜红蓟黄脉β卫星相应序列的相似性高达97.2%;其包含的A-rich区上游与SCR之间约300 bp的序列与中国胜红蓟黄脉β卫星相应序列的相似性仅为70.2%,而与烟草曲茎β卫星相应序列的相似性最高,为97.3%。重组分析发现,所分离的β卫星是由中国胜红蓟黄脉β卫星和烟草曲茎β卫星重组产生。这是首次在中国发现由不同的β卫星重组产生的β卫星分子。  相似文献

锦紫苏又名彩叶草,是一种草本观赏植物,可以被马铃薯纺锤块茎类病毒科锦紫苏类病毒属的类病毒侵染。目前为止,共发现了6种锦紫苏类病毒,分别为锦紫苏类病毒-1~锦紫苏类病毒-6(CbVd-1~CbVd-6),它们具有共同的中央保守区(CCR),存在广泛的分子间重组,其嵌合体的发现为研究类病毒RNA重组提供了较为理想的试验材料。本文系统综述了锦紫苏类病毒的分类地位及分子特征、检测方法、与寄主互作等方面的研究进展,并对锦紫苏类病毒研究中的热点和难点问题进行了讨论和展望。  相似文献

Yellow vein mosaic disease (YVMD) caused by whitefly‐transmitted begomoviruses is an economically significant viral disease of okra. In this study, a survey of begomoviruses associated with YVMD was carried out in eight states and two union territories of India. A total of 92 full‐length DNA‐A components were sequenced and characterized. Sequence comparisons and population structure analysis revealed the existence of four begomovirus species. Two novel species were detected with several recombinationally derived genome fragments that probably originated from begomoviruses known to infect malvaceous and non‐malvaceous hosts. Among the four species, Bhendi yellow vein Maharastra virus (BYVMaV) and Bhendi yellow vein Madurai virus (BYVMV) were found to be predominant in okra, with BYVMV having a pan‐India distribution. There was evidence for a high degree of genetic variability and subpopulation structure within these four species. Neutrality tests suggested the occurrence of purifying selection acting upon these populations. The results of the current study have uncovered the diversity and genetic structure of okra‐infecting begomoviruses in India and generated potentially useful information for developing management strategies for YVMD.  相似文献

Ridolfia segetum is a frequent umbelliferous weed in sunflower crops in the Mediterranean basin. Field and remote sensing research was conducted in 2003 and 2004 over two naturally infested fields to determine the potential of multispectral imagery for discrimination and mapping of R. segetum patches in sunflower crops. The efficiency of the four wavebands blue (B), green (G), red (R) and near‐infrared (NIR), selected vegetation indices and the spectral angle mapper (SAM) classification method were studied using aerial photographs taken in the late vegetative (mid‐May), flowering (mid‐June) and senescence (mid‐July) crop growth stages. Discrimination efficiency of R. segetum patches in sunflower crops is consistently affected by their phenological stages, in this order: flowering > senescence > vegetative. In both fields, R. segetum patches were efficiently discriminated in mid‐June, corresponding to the flowering phase, by using the waveband G, the ratio R/B or SAM with overall accuracies ranging from 85% to 98%. The application of the median‐filtering algorithm to any of the classified images improved the accuracy. Our results suggest that mapping R. segetum weed patches in sunflower to implement site‐specific weed management techniques is feasible with aerial photography when images are taken from 8 to 10 weeks before harvesting.  相似文献

 通过RT-PCR扩增了黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)济南分离物(CGMMV-JN)的基因组片段。序列测定结果表明CGMMV-JN基因组全长6 424核苷酸(nt),5′-和3′-UTR分别为60和176 nt,含有4个ORF,分别编码129 kDa和186 kDa复制酶相关蛋白、29 kDa移动蛋白及17.4 kDa外壳蛋白。CGMMV-JN与另外29个CGMMV分离物全基因组核苷酸序列一致率为90.0%~99.7%。重组分析发现韩国KOM(AF417243)、以色列EC(KF155231)、印度(DQ767631)和我国河北的CHB(KJ658958)4个分离物在RdRp编码区存在重组。系统发育分析结果表明,这些分离物可分成3个组。选择压力分析结果表明cp基因处于正选择,其它基因处于负选择。本文研究的结果为黄瓜绿斑驳花叶病毒的监测及防控提供了理论依据。  相似文献

The biological and molecular relationships between a large number of Potato virus Y (PVY) isolates were examined, concentrating mainly on isolates associated with potato tuber necrotic ringspot disease (PTNRD). Following detailed analysis of the coat-protein gene, four main groups were identified which broadly corresponded to the phenotype of the different isolates. The groups comprised the ordinary strain (PVYO), the necrotic strain (PVYN), the C strain (PVYC) and a group of recombinant (between ordinary and necrotic) isolates. In the latter group, all members were associated with PTNRD. However, four nonrecombinant isolates were also identified which were associated with PTNRD or tuber necrosis. Three were from tubers showing PTNRD symptoms in the field, while the fourth originated from symptomless tubers, but could cause necrotic rings on tubers under glasshouse conditions. The results show that although coat-protein recombination is always found associated with the PTNRD phenotype, some nonrecombinant isolates have very similar biological properties.  相似文献

[目的]分别构建84K杨的微管蛋白TUA5和TUB16与红色荧光蛋白mCherry融合的植物过表达载体,瞬时表达验证载体在植物体内表达后的荧光信号,为研究杨树微管功能奠定基础。[方法]以毛果杨微管蛋白TUA5和TUB16的基因序列为模板,设计84K杨的TUA5和TUB16基因的引物,提取野生型84K杨的RNA并反转录成c DNA,同源克隆得到84KTUA5和84KTUB16基因,分别连接在pCAMBIA 1300载体mCherry荧光标签的N’端和C’端,转化到大肠杆菌TOP10感受态细胞中,通过菌落PCR和测序鉴定获得阳性单克隆,并通过电击法将重组质粒转化到农杆菌GV3101中,瞬时转化烟草后进行荧光观察。[结果]克隆得到了84KTUA5和84KTUB16基因,成功与pCAMBIA 1300-mCherry载体连接,烟草瞬时表达荧光观察结果显示:仅目的基因与mCherry标签C’端相连的融合蛋白能够成功表达,且荧光明显。[结论]成功构建了84K杨TUA5和TUB16基因与pCAMBIA 1300-mCherry的融合表达载体,为进一步研究杨树微管功能提供了背景材料。  相似文献

Present status of genetics of rust resistance in flax   总被引:5,自引:0,他引:5  
Summary Present knowledge of host genes conferring resistance to rust in flax and their genetics are reviewed. There are at least 34 genes conferring resistance to rust occurring in seven groups, namely, K, L, M, N, P, D and Q. Expression of these host genes is affected by temperature, genetic background and by the inhibitor gene present in certain rust strains. Recombination analysis indicates that genes within each of the M and N groups are probably closely linked and that of the L group are genetically complex. When testcross progeny between two genes of the L group were screened, susceptible and modified recombinants were recovered. Some of these susceptible recombinants yielded rare resistant revertants in their progeny. Mechanism of such reversion is not defined but appears to follow a definite pattern. It is also indicated that some of the recombinants represent new specificity. A molecular approach of cloning host genes in flax is described.  相似文献

Cytological modes of the origin of 2n gametes were investigated in six different genotypes of F1 hybrids between Oriental and Asiatic (OA) lilies (Lilium, 2n = 2x = 24). Chromosome pairing between the parental genomes was very low, the average frequency range from 0.3 to 1.2 bivalents per cell among the genotypes. Within a genotype the frequency of bivalents varied from 0 to 6 in some cases. The normally occurring haploid pollen grains were totally sterile. In contrast, in different genotypes, variable percentages of 2n pollen were found and shown to be fertile as estimated from pollen germination. A cytological analysis of Metaphase I and subsequent stages of meiosis using genomic in situ hybridization (GISH) revealed that there was intergenomic recombination between the alien genomes. Following Metaphase I stage, three different types of abnormal cytological events led to the formation of 2n pollen: (i) Post-Metaphase I division (PMI), (ii) Post-Metaphase II division (PMII) and (iii) Asymmetric Cytokinesis of the pollen mother cell followed by chromosome division. All three cytological events led to first division restitution (FDR) gametes. Based on in vitro pollen germination it was proved for two genotypes that 2n pollen was viable only during the first day of anthesis. It was possible to use 2n pollen successfully for backcrossing. Implications of 2n pollen for intergenomic recombination in BC1 progenies are discussed.  相似文献
