Seed longevity is a very important characteristic controlling seed quality. However, the mechanism underlying this characteristic is poorly understood. In this study, 'FH7185', a storable rice variety, the germination rate of which was 66.63%, much higher than the control after artificial ageing under relative humidity 88% and 42°C for 21 days. Different methods were applied to reveal the involvement of proteins during ageing of rice seeds. In total, 35 differential proteins were identified from 2D‐PAGE, and 3,719 proteins from iTRAQ analysis. A comparison of these two methods showed that not all protein types could be detected on the 2D‐PAGE gels, and the dynamic range was somewhat limited. So the comprehensive proteome map from the iTRAQ analysis was used to identify the quantitatively regulated proteins that played roles in seed longevity, and found that the most marked change was the increased abundance of many metabolic enzymes, especially the ones involved in α‐linolenic acid metabolism in the embryos during ageing. OsLOX2 and OsLOX3 had a negative effect on seed longevity, which were lower in FH7185 than the control. The dehydrogenase (LOC_Os07g44430) which played a major catalytic role in lipid peroxidation, also changed significantly during ageing. Therefore, OsLOX2, OsLOX3 and Loc_OS 07G44430 may be involved in the regulation of seed longevity with a synergistic effect.  相似文献

ABSTRACT

Overexpression of Rubisco small subunit (RbcS) of C4 plant, sorghum (sorghum bicolor) was shown to enhance the catalytic turnover rate (k cat) of Rubisco in rice (Oryza sativa). In this study, the effects of other Rubisco small subunits of C4 plants, Napier grass (Pennisetum purpureum) and guinea grass (Megathyrsus maximus) on kinetic properties of Rubisco in rice were studied. The expression levels of Napier grass RbcS (NgRbcS) and guinea grass RbcS (GgRbcS) proteins accounted for 41% and 45% of total RbcS, respectively in homozygous overexpression lines. The k cat and K m for CO2 (Kc) of Rubisco were increased in all transgenic lines. Interestingly, the k cat was markedly higher in NgRbcS homozygous line, whereas K c was notably higher in GgRbcS homozygous line. Although its effects depend on species, these results suggest that the introduction of C4 RbcS are effective approaches to alter the catalytic properties of Rubisco in rice.  相似文献

In Japan anthropogenic-alluvial soils excluding irrigated rice soils are lagging for behind other genetic soil types in pedological investigations. Although there are some investigations of anthropogenic-alluvial soils used for mulberry plantations, orchards, vegetable gardens, and seeding beds for trees, the main object of these investigations is not pedological. Kanno et al. (1) reported that unirrigated anthropogenic-alluvial soils used for vegetable growing in the Chikugo Plain resemble the “Alochthonous brown warp soil” defined by Kubiena (2). First of all, it is of great importance to accumulate general data for unirrigated anthropogenic-alluvial soils and to develop the classification principles for them. Classification problems will be settled in accordance with the accumulated data. This paper deals with characteristics of an unirrigated anthorpogenic-alluvial soil found in the Kumamoto Plain and with classification principles for it. These principles have almost never been described for a pedological standpoint in Japan.  相似文献

The five‐year value in the compound annual growth rate of the biopesticides sector is predicted to be 16% by 2017 and to produce a global market worth $US 10 billion. Despite this, several impediments occur within the EU that negatively affect biopesticide research and innovation. At present, there are fewer biopesticide‐active substances registered in the EU compared with the United States, India, Brazil and China. The relatively low level of biopesticide research in the EU (6880 ISI papers) versus the United States (18 839), India (9501) and China (7875) relates to the greater complexity of EU‐based biopesticide regulations compared with these other countries. In this light, it is worth noting that tensions may exist between regulators that emphasise the beneficial nature of biopesticides in environmentally friendly crop management and those that adopt a more technologically based approach dependent on a chemical‐pesticide‐driven model. Compared with the other aforementioned countries, far fewer biopesticide products are available in the EU market, mainly as a direct result of the severe regulatory factors present there. The extent to which this trend will continue depends largely on a range of interacting political and/or regulatory decisions that influence environmentally friendly agricultural industries. © 2017 Society of Chemical Industry  相似文献

The effect of fertilizers on methane emission rates was investigated using an automated closed chamber system in Chinese rice fields (Human Province). Each of three experiments compared two fields treated with a first uniform fertilizer dose and a second fertilizer dose which was different for each of the two fields. The uniform fertilizer doses for both fields in each experiment comprised mineral (experiment 1), organic (experiment 2) and combined mineral plus organic components (experiment 3). In all three experiments the second fertilizer dose comprised organic amendments for field 1 and no organic amendments for field 2. The rate of increase in methane emission with a given amount of organic manure was found to depend on the total amount of organic manure applied. A single dose of organic manure increased the emission rates by factors of 2.7 to 4.1 as compared to fields without organic manure (experiment 1). In rice fields that had already been treated with organic manure, the application of a second dose of organic manure only slightly enhanced the emission rates in experiment 2 by factors of 1.1 to 1.5 and showed no detectable increase in experiment 3. The net reduction achieved by separation of organic and mineral fertilizers was maximized by concentrating the organic amendments in the season with low emission rates, i.e. early rice, and using exclusively mineral fertilizers on late rice when emission rates were generally higher. This distribution pattern, which was not associated with significant yield losses, resulted in an annual methane emission corresponding to only 56% of the methane emitted from fields treated with blended fertilizers.  相似文献

Deterioration of soil quality under resource-intensive modern agriculture in the face of global climate change poses a huge risk to food security. Because of the complex nature, estimators of soil quality often rely upon a limited set of soil attributes, along with statistical data reduction techniques, for developing quality indices, whilst overlooking biological aspects and regional climatic variability. This study screened the most suitable soil quality indexing approaches for a rice-oilseed-based cropping system in the lower Indo-Gangetic plains (IGP). For this, surface soil samples (0–15 cm) were collected from an ongoing long-term fertilizer experiment with a rice-mustard-sesame cropping system in the IGP. The following treatments were assessed for their effect on soil quality: T1-control, T2-NPK (recommended NPK doses), T3-NPKG (NPK + in situ green manuring), T4-NPKGB (NPK + in situ green manuring + biofertilizer) and T5-NPKF (NPK + farm yard manure FYM). We found that total organic carbon (TOC), β-glucosidase, CaCl2 extractable S, alkaline KMnO4 oxidizable N, activity of urease, amidase enzyme and mean weight diameter (MWD) were sensitive key indicators of soil quality. The NPKF treatment maintained the highest soil quality status (0.80–0.91), both under productivity and environmental protection goals, owing to the availability of decomposable carbon. Regression analysis showed a better agreement of equivalent rice yield with expert opinion (EO; R2 = 0.89) than principal component analysis (PCA; R2 = 0.76). Finally, we found that the expert opinion approach with the nonlinear scoring function was the best tool for soil quality assessment of the region.  相似文献

Summary Although denitrification has long been considered a major loss mechanism for N fertilizer applied to lowland rice (Oryza sativa L.) soils, direct field measurements of denitrification losses from puddled rice soils in the tropics have only been made recently. This paper summarizes the results of direct measurement and indirect estimation of denitrification losses from puddled rice fields and reviews the status of research methodology for measurement of denitrification in rice fields. The direct recovery of (N2+N2O)-15N from 15N-enriched urea has recently been measured at sites in the Philippines, Thailand, and Indonesia. In all 12 studies, recoveries of (N2+N2O)-15N ranged from less than 0.1 to 2.2% of the applied N. Total gaseous N losses, estimated by the 15N-balance technique, were much greater, ranging from 10 to 56% of the applied urea-N. Denitrification was limited by the nitrate supply rather than by available C, as indicated by the values for water-soluble soil organic C, floodwater (nitrate+nitrite)-N, and evolved (N2+N2O)-15N from added nitrate. In the absence of runoff and leaching losses, the amount of (N2+N2O)-15N evolved from 15N-labeled nitrate was consistently less than the unrecovered 15N in 15N balances with labeled nitrate, which presumably represented total denitrification losses. This finding indicates that the measured recoveries of (N2+N2O)-15N had underestimated the denitrification losses from urea. Even with a probable two-or threefold underestimation, direct measurements of (N2+N2O)-15N failed to confirm the appreciable denitrification losses often estimated by the indirect difference method. This method, which determines denitrification losses by the difference between total 15N loss and determined ammonia loss, is prone to high variability. Measurements of nitrate disappearance and 15N-balance studies suggest that nitrification-denitrification occurs under alternate soil drying and wetting conditions both during the rice cropping period and between rice crops. Research is needed to determine the magnitude of denitrification losses when soils are flooded and puddled for production of rice.  相似文献
