Effects of Adding Mannan Oligosaccharides to Diets with Different Concentrate to Forage Ratios on Ruminal Fermentation of Sheep in vitro

The test was aimed at determining the effects of adding MOS to diets with different concentrate to forage ratios on ruminal fermentation of sheep in vitro.The 4×6 two-factor experimental design was chosen.There were 4 diets with different concentrate to forage ratios(20:80, 30:70, 40:60 and 50:50) and added to 6 dosages MOS(0, 0.4%, 0.8%, 1.2%, 1.6% and 2.0%), respectively.The method of gas production in vitro was used to measure the total gas and CH₄ production, IVDMD, IVCPD and IVOMD after cultured for 3, 6, 9, 12 and 24 h.The results showed that the gas(except 12 and 24 h) and CH₄ production(except 9 h) were not impacted by the two factors(P>0.05).The IVDMD, IVCPD and IVOMD were significantly affected by concentrate to forage ratios(P<0.05), while the IVCPD at 24 h was significantly affected by MOS(P<0.05).Meanwhile, all the indexes were not influenced by the interaction of two factors(P>0.05).The IVDMD and IVOMD increased with the rising of concentrate approximately, while the IVCPD decreased with the rising of MOS roughly.The biggest associative effect was gained in 30:70 concentrate to forage diet when adding 1.2% MOS.     

巴西橡胶树乳管细胞HblMYC3互作蛋白筛选与功能分析

MYC是b HLH转录因子家族的亚家族成员,在植物茉莉酸信号转导过程中发挥着重要的调节作用。Hbl MYC3是从巴西橡胶树的乳管细胞中分离鉴定到的MYC类转录因子,其基因表达受割胶和茉莉酸上调。采用酵母双杂交方法初步筛选Hbl MYC3蛋白的互作蛋白,旨在进一步了解Hbl MYC3的功能。结果表明:Hbl MYC1、Hbl MYC2、DNAJ蛋白、谷氧还蛋白2、含A20和AN1锌指结构域的胁迫相关蛋白5、28 ku热和酸稳定的磷蛋白以及25 ku泛素连接酶E2等7种蛋白不同程度地与Hbl MYC3蛋白互作。基于这些互作蛋白的功能,推测Hbl MYC1或Hbl MYC2通过与Hbl MYC3形成二聚体对小橡胶粒子膜蛋白基因表达进行调控,其他蛋白参与胁迫条件下维持二聚体的稳定性和胁迫反应后降解该二聚体。     

Mycoplasma pneumoniae induces IL-1β production through activating NLRP3 inflammasome by ROS in RAW264.7 cells

AIM: To investigate whether Mycoplasma pneumoniae (Mp)-induced interleukin-1β (IL-1β) production in RAW264.7 cells is through the activation of NLRP3 inflammasome via reactive oxygen species (ROS). ME-THODS: RAW264.7 cells were randomly divided into 3 groups. In normal group, RAW264.7 cells were treated without Mp. In model group, RAW264.7 cells were treated with 1∶ 10 multiplicity of infection (MOI) of Mp. In NAC group, RAW264.7 cells were pretreated with N- acetylcysteine (NAC) at a concentration of 5 mmol/L for 30 min before infection with Mp. The RAW264.7cells were infected with Mp (1∶ 10 MOI) for 4, 8, 16 and 24 h in model group and NAC group, respectively. The intracellular ROS level was analyzed by flow cytometry. The mRNA expressions of NLRP3, ASC and caspase-1 were detected by real-time PCR. The protein levels of NLRP3, ASC and caspase-1 p20 were determined by Western blot. The levels of pro-inflammatory cytokine IL-1β in the supernatant were measured by ELISA. RESULTS: Compared with normal group, the production of ROS were significantly increased at 4, 8, 16 and 24 h after infection, the mRNA expression of NLRP3, ASC and caspase-1 were increased at 8, 16 and 24 h after infection, the protein levels of NLRP3, ASC and caspase-1 p20 were increased at 16 and 24 h after infection, and the releases of IL-1β were increased at 24 h after infection in model group (P<0.01). Compared with the model group, the level of ROS in NAC group decreased, so as the expression of NLRP3, ASC and caspase-1 at mRNA and protein levels and the releases of IL-1β in the supernatant at the corresponding time points. CONCLUSION: Mp may stimulate the ROS production to activate NLRP3 inflammasome in RAW264.7 cells.     

基于3G通信技术的现代图书馆工作新思路

3G移动通信技术所具有的高带宽、高数据传输率为现代图书馆的发展提供了新的历史机遇和挑战,传统的数字图书馆在基础设施、管理架构和服务模式等方面已经远远不能满足3G时代读者的个性化需求,因此对传统图书馆进行3G适应性的改造势在必行。结合3G移动通信的技术特征和传统图书馆的不足提出新环境下图书馆工作的新思路。     

基于变分法的FY-3C土壤水分产品适用性分析

FY-3C作为我国风云三号首颗业务卫星,其上搭载的微波成像仪(MWRI)可提供全天候土壤水分数据。【目的】获取高质量土壤水分数据可以对合理利用土壤水资源提供参考,为农田干旱监控和预报提供基础参数。【方法】选取山东省农业气象站土壤水分数据对FY-3C土壤水分产品进行检验,为获取更高质量FY-3C土壤水分产品,选用变分订正方法对FY-3C土壤水分产品进行偏差订正。【结果】FY-3C升降轨土壤水分产品与地面站土壤水分相关系数R分别为0.481 6和0.408 2,RMSE分别为0.099 6和0.091 0 cm~3/cm~3。订正后FY-3C升降轨土壤水分产品与地面站土壤水分R分别为0.701 4和0.892 4,RMSE分别为0.021 7和0.011 cm~3/cm~3。对2016年3—4月山东省干旱过程订正前、后FY-3C土壤水分变化情况进行对比,订正后FY-3C土壤水分更准确地反映出此次干旱过程。【结论】FY-3C土壤水分产品可以准确反映土壤水分随时间的变化趋势,订正后FY-3C土壤水分产品与地面站土壤水分间误差减小、相关性提高。     

滇西北退耕还林后续政策分析及发展对策

滇西北是云南省最为贫困的少数民族聚集地区之一,实行退耕还林政策10多年来取得了巨大的成效,随着退耕还林政策补助陆续到期,部分退耕农户生计将出现困难。对退耕还林后续政策进行分析,认为退耕还林补偿标准与现有物价水平不符,套种的规定过于保守,针对政策上的不足与实施过程中存在的作业设计不够规范、监管不力、档案管理不够规范等问题提出相关对策和建议。     

春白萝卜新品种雪单3 号的选育

雪单3 号是利用双单倍体育种技术选育的春白萝卜新品种。母本ED0108A 是以梅花春萝卜× 白玉春萝卜进行花药培
养获得的DH 系为轮回父本，与不育系进行多次回交，转育成的雄性不育系。父本ED3128 是由迟花春萝卜× 天鸿春萝卜进行
花药培养获得的DH 系。该品种生育期60 d（天），植株开展度45 cm，裂叶，小叶11 对，叶片数20，叶簇平展，叶色深绿；
肉质根长圆柱形，白皮白肉，根长36 cm，横径7.1 cm，单根质量1.1 kg，高抗黑腐病、芜菁花叶病毒病、黄瓜花叶病毒病和花
椰菜花叶病毒病，抗霜霉病，群体整齐度高。每667 m² 产量4 000～5 000 kg。适宜在湖北、湖南、河南、山东等地区早春栽培。     

土壤中缓效钾的批量快速测定

通过对NY/T 889—2004标准中前处理方法的改进,建立了批量、快速、准确测定的土壤中缓效钾的方法。以石墨消解代替油浴消煮测定标准土中的有效钾,以验证改进后方法的可行性。结果表明,改进后的方法一次可以测定36个样品,且3 min内沸腾时间对结果无显著影响。因此,改进后的方法操作简单、条件易控制、准确快速,能实现对土壤中缓效钾的批量、快速测定。
