通过溶液培养的方法,研究了Cd、Cu胁迫对路易斯安娜鸢尾幼苗生长及生理生化的影响.结果表明:Cd、Cu胁迫下,路易斯安娜鸢尾幼苗生长缓慢,地上、地下干重下降,叶片中的叶绿素含量均随胁迫浓度的增加而降低;植株叶片的丙二醛和游离脯氨酸含量、超氧化物岐化酶活性均随处理浓度、处理时间的延长而增加.  相似文献

试验旨在鉴定吉林省某雏鸡孵育基地病死雏鸡组织中分离出的1株致病性菌CCGGD201101株并测定其致病性。对疑似致病菌进行生理生化试验、16S rDNA测序鉴定,并人工接种昆明鼠,测定其半数致死量,验证细菌毒力。经鉴定该菌为鲍曼不动杆菌(Acinetobacter baumannii)。以鲍曼不动杆菌CCGGD201101分离株为研究对象,并以鲍曼不动杆菌标准株(ATCC 19606)为对照,测得半数致死量,进一步证明鲍曼不动杆菌病死鸡分离株CCGGD201101具有较强致病性。  相似文献

试验采用水培种植的方法研究不同NaCl浓度对羽衣甘蓝幼苗光合色素含量、可溶性蛋白质含量、过氧化氢酶(CAT)活性以及过氧化物酶(POD)活性的影响。结果表明:随着NaCl浓度的增加,叶绿素含量总体呈下降趋势;可溶性蛋白质含量、CAT和POD活性随着NaCl浓度的增加先上升后下降;在NaCl浓度为600mmol/L时可溶性蛋白质含量、CAT活性达到最高值,而POD活性则是在NaCl浓度为800mmol/L时达到最高。  相似文献

Oxidative stress is inevitable as it is derived from the handling, culturing, inherent metabolic activities and medium supplementation of embryos. This study was performed to investigate the protective effect of chitosan nanoparticles (CNPs) on oxidative damage in porcine oocytes. For this purpose, cumulus–oocyte complexes (COCs) derived from porcine slaughterhouse ovaries were exposed to different concentrations of CNPs (0, 10, 25 and 50 µg/ml) during in vitro maturation (IVM). Oocytes treated with 25 µg/ml CNPs showed significantly higher levels of GSH, along with a significant reduction in ROS levels compared to control, CNPs10 and CNPs50 groups. In parthenogenetic embryo production, the maturation rate was significantly higher in the CNPs25 group than that in the control and all other treated groups. In addition, when compared to the CNPs50 and control groups, CNPs25-treated oocytes showed significantly higher cleavage and blastocyst development rates. The highest concentration of CNPs reduced the total cell number and ratio of ICM: TE cells in parthenogenetic embryos, suggesting that there is a threshold where benefits are lost if exceeded. In cloned embryos, the CNPs25 group, as compared to all other treated groups, showed significantly higher maturation and cleavage rates. Furthermore, the blastocyst development rate in the CNPs25-treated group was significantly higher than that in the CNPs50-treated group, as was the total cell number. Moreover, we found that cloned embryos derived from the CNPs25-treated group showed significantly higher expression levels of Pou5f1, Dppa2, and Ndp52il genes, compared with those of the control and other treated groups. Our results demonstrated that 25 µg/ml CNPs treatment during IVM improves the developmental competence of porcine oocytes by reducing oxidative stress.  相似文献

为探明植物对锰胁迫的耐受性机制,以采自重金属污染区与非污染区的鸡眼草为试验材料,在不同锰浓度胁迫下[0 (对照)、1000、5000、10000、15000、20000 μmol·L-1] 开展盆栽试验,比较研究锰胁迫对两种来源鸡眼草表型、生理生化特性及锰积累特征的影响。结果表明:随着锰浓度的升高,1) 两种来源鸡眼草的根干重、芽干重、根冠比均呈降低的趋势;当锰浓度达5000~20000 μmol·L-1时,与对照相比,污染区、非污染区鸡眼草的芽干重分别下降4.34%~27.71%与16.33%~49.77%,根干重分别下降19.00%~66.06%与27.90%~77.54%,污染区下降幅度均较非污染区的小;2) 两种来源鸡眼草的叶绿素、可溶性糖、脯氨酸含量及超氧化物歧化酶(SOD)、过氧化物酶(POD)活性均呈先升后降的趋势,可溶性蛋白含量逐渐下降,丙二醛(MDA)含量则逐渐升高;3) 两种来源鸡眼草根、茎、叶的锰含量均增加;锰浓度为20000 μmol·L-1时,污染区和非污染区鸡眼草的叶锰含量分别为对照的16.53和13.41倍。因此,污染区鸡眼草锰耐受能力及富集能力均较非污染区鸡眼草高,较高的抗氧化酶活性是其耐受高锰胁迫的重要生理机制。  相似文献

To study the effect of weaning stress on dairy buffalo calves,5 healthy calves were chosen and weaned,and the blood samples were collected at 1 day before weaning,and 1,14 and 21 days after weaning to analysis blood physiology and biochemistry indexes and immune function.The results showed that the count of white blood cell,lymphocyte,erythrocyte,monocytes and hematocrit,aspartate aminotransferase activity,blood glucose,cortisol and immunoglobulin G contents of dairy buffalo calves were no significant difference between before and after weaning treatments (P > 0.05).The count of neutrophils at 21 d after weaning were significantly lower than that of 1 d before weaning of dairy buffalo calves (P < 0.05).However,lactate dehydrogenase and alanine aminotransferase activity in weaned treatments were significantly higher than those of animals in the treatment before weaning (P < 0.05).The serum urea nitrogen of dairy buffalo calves at 21 d after weaning was significantly higher than that of 1 d before weaning (P < 0.05) and hemoglobin content at 21 d after weaning was significantly lower than that at 1 day before weaning (P < 0.05).It indicated that the physiological and immunity function of dairy buffalo calves were challenged under weaning stress.  相似文献

The changes of cortisol, red blood cells (RBC), hemoglobin (Hb), hematocrit (Hct), heart rate (HR), respiratory rate (RR) and rectal temperature (RT) were evaluated after a reining training session in eight Quarter Horses. All parameters were measured before exercise (T0), immediately after exercise (T1), 1 h after exercise (T2), 2 h after exercise (T3) and 24 h after exercise (T4). One‐way analysis of variance (ANOVA) for repeated measures, followed Bonferroni's post hoc test, showed a significant effect of the reining training session (P < 0.0001) on cortisol, RBC, Hb, Hct, HR, RR and RT. Simple linear regression analysis showed the positive correlation (P < 0.05) between cortisol changes and variations of studied parameters in T1, T3 and T4. Exercise‐induced cortisol concentrations reflect the physiological response of reining training, suggesting that the changes observed are useful to assess the performance in reining horses and their reining training adaptability.  相似文献

Rhinoceros populations in Africa are under severe threat as a result of surging poaching rates and risk-mitigation strategies are continuously adapted in an attempt to ensure the survival of the species. This study compared faecal glucocorticoid metabolite (fGCM) levels of two age classes of limited free-ranging female white rhinos with fGCM levels of adult free-ranging female white rhinos. Subsequently, fGCM alterations in the limited free-ranging animals were monitored following routine dehorning as a measure of the animals’ short-term physiological stress response. Baseline fGCM levels differed significantly between tested groups, with both free-ranging and limited free-ranging adult animals showing significantly higher fGCM levels compared with limited free-ranging juvenile females. In contrast, baseline fGCM levels did not differ significantly between limited free-ranging and free-ranging adult individuals. Routine dehorning procedures resulted in a short-term stress response expressed by a significant increase in fGCM levels 48 h post-dehorning, with stress steroid levels returning to pre-dehorning concentrations 72 h after the procedure.  相似文献

稻粉虱Aleurocybotus indicus David et Subramaniam, 属半翅目粉虱科, 以刺吸式口器为害水稻叶片而导致产量损失, 是我国闽?赣?浙等稻区的一种偶发性水稻害虫, 近20多年已鲜有报道, 但2019年-2021年在中国水稻研究所富阳试验农场稻田暴发?本文描述了稻粉虱的为害症状和形态特征, 并配以原色照片; 克隆得到了稻粉虱线粒体DNA COⅠ基因片段, 发现其与烟粉虱Bemisia tabaci序列一致性最高(85.80%); 田间调查表明, 富阳试验农场及周边稻田稻粉虱均有不同程度的发生, 但前者发生量明显较高(每个分蘖最高可达251头), 推测可能与农场温室内的稻粉虱越冬虫源有关; 利用稻苗浸渍法评估了不同作用机制的7种杀虫剂对稻粉虱的毒力, 发现双丙环虫酯?氯虫苯甲酰胺?氟啶虫胺腈?氟啶虫酰胺和溴氰虫酰胺对稻粉虱成虫均有较好的杀虫活性, 其中前4种药剂推荐中剂量处理后72 h的校正死亡率均可达100%, 其LC50分别为3.028?3.769?6.405 mg/L和7.894 mg/L?本研究可为我国稻粉虱的识别诊断和化学防治提供重要依据?  相似文献
