为了探讨miR-24对猪骨骼肌发育的影响,对体外培养1日龄纯种长白公猪骨骼肌卫星细胞分别转染miR-24过表达载体和干扰片段,发现过表达miR-24后,成肌分化基因MyoD、Myogenin和Myf5表达量显著升高,骨骼肌卫星细胞分化趋势增强;干扰miR-24后,Myogenin的表达量显著降低;同时,靶基因预测结果显示Myogenin可能是miR-24的靶基因。  相似文献

【目的】研究HMGCS1基因对猪骨骼肌生长发育的影响及其靶标的验证。【方法】采用real-time PCR对通城猪不同组织以及背最长肌不同发育时间点的HMGCS1基因的表达变化进行检测。【结果】①HMGCS1基因在成年猪肺组织中的表达量最高;②在背最长肌发育中,该基因分别在胚胎期33 和65 d高表达,出生后20、30、40和60 d低表达;③双荧光素酶试验显示,转染miR-18a/b minics组荧光素酶活性低于对照组。【结论】HMGCS1参与了猪骨骼肌生长发育过程,并受miR-18a/b的调控,是猪的产肉性状的潜在候选基因。  相似文献

It is well known that an increase of and better grain can be obtained by the application of an adequate amount of quick-acting nitrogen fertilizers such as ammonium sulphate, thirty to twenty five days before heading, and this is a common practice of top-dressing, being called “Hogoe” in Japanese. It is also well known that excess of nitrogen supplied at this time makes plants weak against mechanical injury, insects and disease. For the application of “Hogoe”, therefore, an accurate diagnosis of the nitrogen nutrient condition of rice is required. In a series of investigations on the nitrogen metabolism, the author found that asparagine appeared in parallel with the increase of nitrogen concentration in rice plants, and considered that the detection of asparagine would be a good indicator for assessing the nitrogen requirement of rice lantstl).  相似文献

在室养条件下观察了MS-222、苯佐卡因和乌拉坦对稀有鮈鲫(Gobiocypris rarus)的麻醉效果。结果表明,乌拉坦不宜作为稀有鮈鲫的麻醉剂,而一定浓度的MS-222或苯佐卡因可导致稀有鮈鲫的快速麻醉;不同麻醉剂浓度、不同的作用时间下,稀有鮈鲫在行为上呈现不同的反应,据此可分为轻度镇静、深度镇静、轻度麻醉、中度麻醉、深度麻醉和髓质麻醉阶段;根据反应时间、恢复时间、维持时间和存活率等,建议使用60mg/L的苯佐卡因或100~110mg/L的MS-222作为深度麻醉剂量。  相似文献

Clove oil has been demonstrated to be an effective, inexpensive anaesthetic and euthanizing agent for a number of fish species, including rainbow trout, used in aquaculture and fisheries research. However, the potential for clove oil to cause perturbations in important plasma hormone concentrations has not been investigated. The effect of anaesthesia and euthanasia in trout with eugenol (the active ingredient in clove oil) on plasma cortisol, glucose, growth hormone (GH) and two thyroid hormones [tri‐iodothyronine (T3) and thyroxine (T4)] was compared with tricaine methanesulfonate (MS‐222) anaesthesia, and stunning by cranial concussion in two experiments. Effects on blood chemistry were different when comparing the particular anaesthetic method being used. Stunning fish significantly increased plasma cortisol and glucose levels (both P<0.05), while euthanizing fish using either clove oil or MS‐222 had no effect on these hormone levels. In contrast, the levels of GH, T3 and T4 hormones were unaffected regardless of whether fish were euthanized by stunning, MS‐222 or clove oil. Variation in effects between hormones were observed using clove oil eugenol. In fish sampled 10 min after anaesthetizing with 150 mg L?1 of eugenol, cortisol levels were significantly decreased (P<0.03), while there were no differences in either glucose or GH levels. Tri‐iodothyronine and T4 also showed significantly elevated levels (P<0.05) after 10‐min exposure to eugenol. These results highlight the importance of investigating the potential effects of any new anaesthetic or euthanizing compounds on blood plasma parameters, prior to using them in a research setting, or when comparing results to other studies which have utilized alternative anaesthetic compounds.  相似文献

MS-222、丁香油、苯唑卡因对养殖美洲鲥幼鱼的麻醉效果   总被引:23,自引:0,他引:23  
研究了MS-222、丁香油、苯唑卡因3种麻醉剂对美洲鲥Alosa sapidissima幼鱼的麻醉效果,并运用该3种麻醉剂的适宜剂量对美洲鲥幼鱼进行了运输试验。麻醉试验结果表明:在较高麻醉浓度(MS-222为75 mg/L以上,丁香油为20 mg/L以上,苯唑卡因为40 mg/L以上)下,鱼很快(30 min内)停止鳃盖张合运动,且停止鳃盖运动的鱼在空气中暴露一定时间(10 min内)后也能够复苏;在适宜的麻醉浓度(MS-222为20-30 mg/L,丁香油为8-10 mg/L;苯唑卡因为20-30 mg/L)下,鱼能够进入麻醉状态,且能保持很长时间(12 h);麻醉效果随着水温的升高而增强;在20 mg/L MS-222麻醉剂下,小规格鱼较大规格鱼更容易进入麻醉状态,而在10 mg/L丁香油和20 mg/L苯唑卡因麻醉剂下,小规格鱼却难进入麻醉状态。运输试验结果表明:麻醉运输组和对照组(非麻醉运输组)鱼血清中皮质醇的含量均显著高于基础组(运输前)(P<0.05);麻醉运输后鱼血清中皮质醇的含量虽均有一定程度的升高,但明显低于对照组,其中仅苯唑卡因麻醉运输组鱼血清中皮质醇的含量显著低于对照组(P<0.05)。试验结果表明,苯唑卡因更适合用于运输美洲鲥的麻醉。  相似文献

To determine the regulative role of miR-142-3p in lactation of dairy goat mammary epithelia cells (DGMECs),we selected the DGMECs in lactating dairy goat for material,used liposomal transfection techniques to silence miR-142-3p expression,and used Real-time quantitative PCR,Western blotting and Assay Kits to explore the changes of lactation after miR-142-3p inhibition.The results showed that when miR-142-3p was inhibited,the cell proliferation ability of DGMECs was increased,the secretion amount of β-casein and triglyceride were up-regulated.So we concluded that miR-142-3p could influence the lactation function of DGMECs,such as cell proliferation,β-casein and triglyceride secretion,by inhibiting the function of its target genes.  相似文献
