Hereditary methemoglobinemia associated with nicotinamide adenine dinucleotide-cytochrome b5 reductase (b5R) deficiency is a rare autosomal recessive disorder in animals. Recently, nonsynonymous b5R gene (CYB5R3) variants have been reported to be associated with canine and feline hereditary methemoglobinemia. However, the underlying molecular mechanisms of canine and feline methemoglobinemia caused by these nonsynonymous variants have not yet been reported. Previously, we reported a Pomeranian dog family with hereditary methemoglobinemia, carrying CYB5R3 mutation of an A>C transition at codon 194 in exon 7, replacing an isoleucine residue with leucine (p.Ile194Leu). In this study, we investigated the enzymatic and structural properties of the soluble form of wild-type and Ile194Leu canine b5Rs to characterize the effects of this missense mutation. Our results showed that the kinetic properties of the mutant enzyme were not affected by this amino acid substitution. The secondary structure of the wild-type and Ile194Leu b5Rs detected by circular dichroism showed a similar pattern. However, the mutant enzyme exhibited decreased heat stability and increased susceptibility to trypsin hydrolysis. Moreover, the thermostability and unfolding measurements indicated that the mutant enzyme was more sensitive to temperature-dependent denaturation than the wild-type b5R. We concluded from these results that unstable mutant enzyme properties with normal enzymatic activity would be associated with hereditary methemoglobinemia in the Pomeranian dog family.  相似文献

The specific primers were designed according to Ovis aries DRA gene sequence deposited in GenBank and the multiple cloning site of the plasmid pYD1,which was a vector used for protein surface display on Saccharomyces cerevisiae.The gene encoding DRA was amplified by PCR using the genomic RNA of Ovis aries.The 762 bp fragment was cloned and released in GenBank and registration number was KR422362.The PCR product was inserted into the yeast surface display plasmid vector pYD1 by double enzyme digestion.It was indicated that DRA gene was successfully integrated into the genome.Dot mutation was made at both ends of exon 2 in DRA gene for making restriction enzyme cutting site and design the exon 2 specific primers according to mutated Ovis aries DRA gene sequence.Sequenced exon 2 amplification products based on DNA pooling of sheep large sample template was analyzed the polymorphic loci.The polymorphic exon 2 246 bp fragment was obtained by double enzyme digestion and connected to surface display restructuring mutation carriers pYD1-DRA by the same double enzyme digestion,and then we successfully constructed yeast surface display libraries.We transformed it into Saccharomyces cerevisiae EBY100 cell.Yeast monoclone was identified by PCR amplification and sequencing,and we confirmed that DRA gene had been integrated into Saccharomyces cerevisiae genome.After galactose induced,it was detected that DRA gene library had been successfully demonstrated on the yeast cell surface under the fluorescence microscope by immunofluorescence method.  相似文献

为明确60Co-γ亚不育剂量辐照对小菜蛾Plutella xylostella种群数量动态变化的影响,构建小菜蛾种群动态变化模型,在实验室条件下通过建立小菜蛾生命表获得种群特征参数、个体生理指标,预测亚不育剂量辐照对小菜蛾种群的影响。结果显示,亚不育剂量辐照组的羽化率显著高于完全不育剂量组,与空白对照组无显著差异,各处理羽化率无性别差异,亚不育剂量辐照组F1代孵化率显著低于空白对照组。亚不育剂量辐照组种群各阶段发育历期、存活率及繁殖力生命指标低于空白对照组。小菜蛾亚不育剂量辐照组世代平均历期(19.90 d)、世代净生殖率(2.35)、周限增长率(1.04 d-1)、内禀增长率(0.04 d-1)均低于空白对照组,而种群加倍时间(16.16 d)高于空白对照组。空白对照组世代平均历期、每日单雌产卵量与产卵时间均长于亚不育剂量辐照组。辐照组与对照组种群生命表参数对比,表明亚不育剂量降低了小菜蛾种群世代数和幼虫期的个体存活率,使F1代雌虫个体数减少。  相似文献

二斑叶螨寄主植物范围极广, 但其对百合科蔬菜造成危害的研究报道极少?本研究采集了来自北京海淀百合科蔬菜蒜和葱上的2个二斑叶螨种群, 生物测定结果显示, 2个种群对阿维菌素呈现出极高水平抗性, 对哒螨灵为中等以上抗性(LC50>5 000 mg/L), 对新型药剂联苯肼酯?虫螨腈和腈吡螨酯也表现出中等到高水平抗性?抗性基因突变频率检测发现, 与阿维菌素抗性相关基因G314D和G326E位点以及和哒螨灵抗性相关基因H92R位点的突变频率均高达100%, 为纯合抗性种群?因此, 本研究所测试的2个二斑叶螨种群对阿维菌素和哒螨灵单剂的敏感性极低, 需慎重选用这2种药剂, 而3种新型杀螨剂联苯肼酯?虫螨腈和腈吡螨酯在田间需轮换使用?  相似文献

Oral squamous cell carcinoma (OSCC) is the most common oral epithelial malignancy in dogs. It exhibits locally aggressive biological behaviour with the potential to metastasize, and a reported 1-year survival rate of 0% when left untreated. Expression studies suggest that aberrant MAPK signalling plays a key role in canine OSCC tumorigenesis, which is consistent with BRAF and HRAS MAPK-activating mutations reported in some tumours. Several morphological subtypes of canine OSCC have been described, with papillary, conventional, and basaloid as the most common patterns. We hypothesized that mutational differences may underlie these phenotypic variations. In this study, targeted Sanger sequencing and restriction fragment length polymorphism assays demonstrate that up to 85.7% of canine papillary OSCC (n = 14) harbour a BRAF p.V595E mutation. Assessment of neoplastic epithelial cell proliferation using Ki67 immunolabelling (n = 10) confirmed a relatively high proliferation activity, consistent with their known aggressive clinical behaviour. These findings underscore a consistent genetic feature of canine papillary OSCC and provide a basis for the development of novel diagnostic and targeted therapeutic approaches that can improve the quality of veterinary care.  相似文献

应用化学诱变剂处理大豆种子,对M_1代植株的生育及形态均有强烈的抑制作用.M_1代所产生的形态变异主要是由于化学物质扰乱了植株生理而产生生理损伤所致.在M_2代植株各性状中以分枝数、株高、单株荚数的遗传变异系数较大,说明这些性状选择的潜力是大的,通过选择容易达到预期目标.诱变M_3代绝大多数性状仍在分离.加强选择,能选出早熟、大粒、农艺性状好、丰产的新品系.  相似文献

One of the most important breakthroughs in the history of genetics )was the discovery that mutations can be artificially induced in organisms (van Harten, 1998). Artificially induced mutations, by physical and chemical mutagens, have greatly advanced the understanding of genetics of higher organisms. Starting in the late 1960's, the International Atomic Energy Agency (IAEA) and the Food and Agriculture Organization (FAO) of the United Nations sponsored extensive research on mutation induction and their application to breeding of food and indnstrial crops that resulted in the introduction of new varieties of rice, wheat, barley, apples, citrus, sugar cane, banana, and others (more than 2 500 officially released new varieties are to be found in the FAO/IAEA Mutant Varieties Database) (hitp://www-mvd.iaea.org/MVD/default.htm). However, the usefulness of mutation techniques has been underappreciated in research communities, particularly during the last decade, when more and more researchers and breeders were rushing into molecular marker techniques and transgenic plants. 采用微波加热和常规加热方法聚合得到聚松香丙烯醇酯,分别对聚合物进行了红外、紫外、软化点和热失重分析。结果表明,微波加热聚合得到聚松香丙烯醇酯具有交联结构,松香的共轭双键参与了反应,软化点在300℃以上,微波加热反应的聚松香丙烯醇酯的相对分子质量在29 305以上,常规加热方法聚合得到聚松香丙烯醇酯相对分子质量在3 910以上,常规加热方法聚合时加入交联剂后得到的聚合物,相对分子质量在9 729以上,聚合物中残留有更多的双键,聚合物的热稳定性减弱。采用微波加热合成聚松香丙烯醇酯快速简便,聚合物性能优于常规方法。  相似文献

为探讨山羊MC1R基因c.676A>G突变与山羊皮肤色素沉积的关系,及其对黑色素合成和cAMP信号通路下游色素相关基因表达的影响:一方面采用直接测序法检测酉州乌羊及其杂交后代群体(120个个体)MC1R基因c.676A>G的突变情况,用色差仪测量不同基因型个体的腹部皮肤色度值,分析c.676A>G突变与皮肤色素沉积的关系;另一方面,构建野生型(MC1R c.676A)和突变型(MC1R c.676G)真核表达载体,采用脂质体介导法转染到小鼠皮肤黑色素瘤细胞(B16-F10)中进行过表达,利用酶标仪检测各组细胞黑色素含量的差异,并通过实时荧光定量PCR方法检测各组细胞中cAMP信号通路下游色素相关基因(MITF、TYR、TYRP1、DCT)的表达差异性。结果显示:酉州乌羊及其杂交后代群体中,c.676A>G位点均以GG基因型占优势,不同基因型群体的皮肤色度值差异不显著(P>0.05)。MC1R突变组(MC1R c.676G)细胞中的黑色素含量极显著(P<0.05)高于MC1R野生组(MC1R c.676A)。MITF、TYRP1和DCT基因在MC1R突...  相似文献
