香豆素为黄花草木樨(Melilotus officinalis)水浸提液的主效化感物质,能抑制多种杂草种子的萌发和幼苗生长。为探明香豆素的化感抑草作用机理,本研究采用室内培养、生物检测的方法,研究了简单香豆素水溶液对苏丹草(Sorghum sudanense)种子萌发及其幼苗生长和生理生化指标的影响,并采用透射电镜观察了苏丹草幼苗根和茎的超微组织结构。结果表明,20~100μg·mL~(-1)香豆素水溶液能够显著抑制早期苏丹草种子的萌发(P0.05),且随浓度升高抑制作用增强,能够显著抑制苏丹草根伸长,随处理时间的延长和浓度的升高其抑制作用也增强,根变粗扭曲,而对茎长的影响存在浓度效应和时间效应,在4d时,20~100μg·mL~(-1)浓度的香豆素水溶液均促进茎长,6和8d则表现为低浓度促进、高浓度抑制;与加蒸馏水的对照组相比,100μg·mL~(-1)香豆素溶液处理显著(P0.05)提高了过氧化氢酶(CAT)活性,并降低了超氧化物歧化酶(SOD)活性,对过氧化物酶(POD)抑制作用表现为先降低后升高;香豆素处理显著(P0.05)提高了可溶性糖的含量,同时显著(P0.05)降低了叶绿素的含量;在香豆素处理下,淀粉含量先降低后升高,吲哚乙酸氧化酶(IAAO)活性仅在第4天时低于对照组。香豆素水溶液破坏了苏丹草根细胞的超微结构,出现液泡体积变大,细胞器数量和种类减少的现象,细胞核的核膜发生明显的歪曲,细胞核变小甚至消失;香豆素水溶液处理下的苏丹草幼苗叶绿体内淀粉粒数目显著(P0.05)增加,体积变大。以上结果显示,100μg·mL~(-1)的香豆素水溶液可以通过改变苏丹草抗氧化酶系统、增加渗透调节物质、降低叶绿素含量、影响酶活性以及破坏细胞超微结构等途径调节苏丹草的生长。 相似文献

To test the efficacy of intraoperative intrafragmentary administration of bupivacaine (haematoma block) in controlling postoperative pain in dogs undergoing osteosynthesis of long-bone isolated diaphyseal fractures.Study design
Randomized, ‘blinded’, placebo-controlled, prospective study.Animals
A total of 23 client-owned dogs with isolated long-bone fractures.Methods
Dogs were allocated randomly to two groups: bupivacaine group (B) or placebo group (P). Group B dogs (n = 11) were administered an intraoperative intrafragmentary injection of 0.5% bupivacaine (1.1 mg kg–1) just before fracture fixation, whereas group P dogs (n = 12) were administered normal saline. Postoperative pain evaluations using the University of Melbourne Pain Scale (UMPS) and algometer were performed upon arrival to the recovery room and 1, 2, 4, 6, 8, 20 and 32 hours later. Algometer measurements were performed on: the incision site, a healthy region near the fracture line and the contralateral healthy limb. When the pain score exceeded 14 points in the UMPS, rescue analgesia was administered. The time-standardised area under the curve (AUCst) was used to compare UMPS scores and mechanical pain thresholds between the two groups.Results
None of the group B dogs required rescue analgesia, whereas eight of the 12 group P dogs did (p = 0.001). The pain threshold AUCst at the incision line was higher in group B [16.3 (2.9–41.6) N] than in group P [5.6 (2.5–17.4) N] (p = 0.029). The mean UMPS score AUCst was lower in group B (3.7 ± 1.8) than in group P (9.4 ± 4.6) (p = 0.016). In a small number of animals of both groups that were evaluated radiologically, adequate bone healing was noted.Conclusions and clinical relevance
An intraoperative bupivacaine haematoma block is a simple, quick and effective method that can be used to aid in postoperative pain control in dogs submitted to long-bone osteosynthesis. 相似文献13.

Transvenous cardiac pacing is a commonly employed treatment for symptomatic bradyarrhythmias in dogs. Medical management of these conditions has not proven to be successful leaving pacing as the only viable alternative. The technique of pacemaker implantation is not difficult, but successful pacemaker therapy requires an understanding of electrophysiology, cardiac fluoroscopic anatomy, pacemaker instrumentation, and programming. Proper patient selection and identification of the underlying rhythm disturbance is essential to a successful clinical outcome. To describe a novel ultrasound-guided posterior extraconal block in the dog.Study design

A total of 13 Beagle Cross cadaver heads.Methods
After describing the ultrasound bony landmarks and posterior extraconal local regional technique in one head, 12 heads were used to evaluate the spreading of contrast and evaluate potential complications. A 5–8 MHz microconvex ultrasound probe was positioned caudal to the orbital ligament, with the beam orientated transversely, and then tilted caudally until the orbital fissure was visualized. After identifying the bony structures consistent with the orbital fissure, a needle was advanced using an in-plane technique and 0.5 mL of a 50:50 mixture of iohexol and methylene blue was injected. Computed tomography (CT) and dissection were used to evaluate successful injections and potential complications. The injection was considered successful if radiopaque contrast medium was 5 mm from the orbital fissure. Potential complications were defined as the presence of radiopaque contrast within the globe or the intracalvarial tissues.Results
The CT images confirmed contrast at the target site in 15/24 (63%) of the injections. Only two injections were found in the temporalis muscle; the rest of the injections were located in the extraconal space. No potential complications such as intracranial spreading of contrast, intravascular or intraocular injection were found.Conclusions and clinical relevance
The technique can deliver contrast close to the main nerves which provide sensory and motor innervation to the eye. Further studies are needed to evaluate this technique in clinical cases. 相似文献15.
Exergy, the thermodynamic function that represents the distance of an open system from equilibrium, is proposed as an ecological indicator for summarizing the complex dynamics occurring in a disturbed community during its recovery processes. These complex dynamics can be difficult to capture by using classical indices. In this study, we sampled macrobenthos using the BACI scheme (before versus after; control versus impact) in tidal wetlands of west Chongming Island, China, an area that has been disturbed by ecological engineering measures. Exergy storage is estimated for benthic communities. The control area is proposed as dynamic reference for estimating local exergy storage of the benthic community. Three different methods were used for estimating exergy on the basis of coefficients: (a) taken for taxa groups; (b) estimated from coding genes given for broad taxonomic groups; and (c) estimated from genome size taken as close as possible to the taxonomic level of the species, providing a basis for inferring similarities. We found a decrease of local exergy content in the disturbed area 9 days after the disturbance. Subsequently, the reference exergy of the benthic community increased (i.e. in the surrounding control area) in accordance with the proposed hypothesis regarding the dynamics of exergy storage during a system's development. Estimates of local exergy arrived at using the three methods provided comparable results. Based on this, we then discuss the feasibility of using the more readily available genome size data for estimating exergy. This result may have important implications for broader application of this indicator to biological systems. Moreover, the adjacent control samples seem to represent an appropriate dynamic reference for estimating the local exergy of disturbed communities. We found that exergy was a useful indicator that integrates the processes underlying the recovery of the benthic community after disturbance. 相似文献

以“桂优62号”桑种为材料,在不同浓度高分子量的聚乙二醇(PEG6000)模拟的水分胁迫条件下,以摇床培养方法对其进行萌发培养。测定不同浓度PEG溶液(30%、25%、20%和双蒸水)中萌发种子的相关抗性生化指标,实验结果经统计分析表明,当PEG浓度为30%时,萌发种子中的脯氨酸、可溶性蛋白、可溶性糖和丙二醛的含量均极显著地高于CK(双蒸水),说明在高浓度PEG溶液处理下能够萌发的种子具有较强的抗水分胁迫的能力,可以作为桑树抗旱育种的选择依据之一。 相似文献

AIMS: To compare two methods of applying rubber rings to the pedicles of yearling stags to induce analgesia in the antler prior to removal. To compare the application of a rubber ring with that of a lignocaine ring block of the antler pedicle on the efficacy and time course of the analgesia produced in yearling stags. METHODS: Rubber rings were applied to the pedicles of 36 yearling stags that required velvet antler removal. The standard method, a doubled-over ring expanded and lowered down from the distal end of the antler and released midway down the pedicle, was compared with a cable-tie method on the other pedicle, where a ring was pulled around the pedicle by an electrical cable tie threaded through the ring. Brief electrical stimulation (train-of-four mode) was applied proximal and distal to the ring before, and at regular intervals for 1 hour after, application of the ring to a level that produced an auriculopalpebral reflex response. In a second experiment, each pair of antlers per yearling stag (n=36) was allocated to one of three pairs of treatments, viz no treatment (control) and the cable-tie method as described above, control and local anaesthesia (a ring block of 2 ml 2% lignocaine per cm pedicle circumference), or the cable-tie method and local anaesthesia. Electrical stimulation (tetanic mode) was applied to each antler approximately 25 mm distal to the pedicle/antler junction before, and at intervals up to 1 hour after, application of treatments at a level required to produce a head/neck avoidance behavioural response. In a third experiment, the two electrical stimulation protocols used above were directly compared by measuring the response of stags (n=8) to one protocol on each pedicle/antler prior to, and at intervals for 1 hour after, application of a rubber ring. At the end of each treatment in all three experiments, analgesia of the antler was established as a nil behavioural response of the stag to a saw cut to the antler (the ‘nick test’). RESULTS: For both methods of application of a ring the minimum electrical stimulation required distal to the ring to elicit a reflex response increased from around 16 to 55 mA by 60 minutes. In contrast, the electrical stimulation required proximal to the ring remained low (~17.0 mA) throughout. No stag subjected to either of the methods of application responded to the nick test 60 minutes after application of the ring. The electrical stimulation required to produce a behavioural response increased very rapidly in stags treated with local anaesthetic and at a slower rate in those treated with the cabletie method but showed no significant increase in control stags. After 4 and 30 minutes, for local anaesthesia and the cable-tie method, respectively, 95% of stags were not responding to 80 mA. A significantly greater proportion of stags with antlers treated with local anaesthetic and the cable-tie method did not respond to the nick test than controls, and there was no significant difference in the frequency of the response between stags with treated antlers. The minimum current required to produce a response proximal to a rubber ring was slightly higher on average for train-of-four electrical stimulus (mean 18.1 (SD 2.6) mA) than for the tetanic mode (mean 11.9 (SD 2.5) mA). The increase in minimum current required to produce the respective response to stimulation distal to the ring was similar for both methods, although the maximum predicted value (67.4 mA) was lower for train-of-four than for the tetanic mode (84.5 mA). No stag responded to the nick test =60 minutes after application of the ring. CONCLUSIONS: The cable-tie method was no different from the standard method as a procedure for producing analgesia in the antlers of yearling stags and should be accepted as an appropriate procedure for applying analgesic rings to yearling stags. The analgesia produced in the antlers of yearling stags by rubber rings applied by the cable-tie method to the pedicle was similar to that of a lignocaine ring block, but the time course for the development of analgesia was markedly different. Given that a lignocaine ring block is accepted as an adequate method of pain relief for antler removal, the application of rubber rings followed by a period of =30 minutes after application can be advocated as a viable alternative for pain relief. 相似文献

ObjectiveTo describe a lateral ultrasound (US)-guided approach to the radial, ulnar, median and musculocutaneous (RUMM) nerves through a single proximal in-plane insertion in cats and to determine whether one or two injection points are required to successfully stain all the target nerves.Study designProspective study.AnimalsA total of eight client-owned healthy cats and 12 cat cadavers.MethodsIn live cats, the US anatomy of the brachium, the landmarks and the site for needle accesses were determined. Then, 12 thawed feline cadavers were used to assess the spread of dye solution and nerve staining following the US-guided proximal-lateral-humeral RUMM injection using one and two injection points. Each cadaver was injected with 0.15 mL kg–1 of a 0.25% new methylene blue solution in either a single injection aimed for the radial nerve of one limb (G1) or via two sites delivering 0.1 mL kg–1 and 0.05 mL kg–1 aimed for the radial and musculocutaneous nerves of the opposite limb, respectively (G2). Upon dissection, staining of the target nerves around their circumference for length of >1 cm was considered successful.ResultsSonoanatomy was consistent with anatomy upon dissection and target nerves were identified in all cadavers. Staining was 100% successful for the radial, median and ulnar nerves in both groups, and 41.7% and 100% for the musculocutaneous nerve in G1 and G2, respectively.Conclusions and clinical relevanceThis novel lateral US-guided approach for the proximal RUMM nerve block allowed a good identification of the nerves and related structures, and it provided a consistent muscular structure through which the needle could be easily guided. Then, anesthetized dogs were administered four bilateral treatments separated by 1 week, saline or 2% lidocaine 1 mL per injection: injections of saline at the maxillary and ethmoidal foramina (Control), injections of lidocaine at the maxillary foramina and saline at the ethmoidal foramina (MBLOCK), injections of saline at the maxillary foramina and lidocaine at the ethmoidal foramina (EBLOCK) and injections of lidocaine at all foramina (M-EBLOCK). The ventral nasal meatus was bilaterally stimulated using cotton swabs, and HR, SAP, MAP, DAP and fR were continuously recorded. Values for each variable were compared before and after stimulation using Wilcoxon signed-rank test. Changes in variables among treatments were analyzed using Mann–Whitney U and Kruskal–Wallis tests (p ≤ 0.05).ResultsComputed tomography revealed iohexol distribution around the openings of the target foramina in all cadavers. In living dogs, HR, SAP, MAP, DAP and fR significantly increased after stimulation within each treatment (p < 0.03). Physiologic responses were significantly attenuated, but not absent, in the M-EBLOCK [HR (p = 0.019), SAP, MAP, DAP and fR (all p ≤ 0.001)] compared with those in the Control.Conclusions and clinical relevanceConcurrent injections of lidocaine at the maxillary and ethmoidal foramina attenuated HR, arterial pressure and fR responses to nasal stimulation in Beagle dogs. 相似文献
