The possibility of using electrophoresis to characterize varieties of pepper, Capsicum annuum and Capsicum frutescens cultivated in Nigeria was investigated. The SDS- polyacrylamide gel electropherogram of extracted total seed proteins of 10 breeding lines in each of the 6 varieties investigated, revealed a pattern in which 12 polypeptide bands with apparent molecular weight range of 22 to 98 kilodaltons could be distinguished. The result showed that the six varieties could be characterized on the basis of presence/absence and staining intensities of 7 polypeptide bands. It is suggested that SDS- polyacrylamide gel electrophoresis of seed proteins provides a useful analytical technique for the characterization of varieties of pepper and there may be genotype duplicates in the collection of Nigerian Capsicum germplasm. 相似文献

鱼肉盐溶蛋白的凝胶特性与鱼糜制品的质地密切相关,而盐溶蛋白的凝胶特性又受到许多因素影响。为研究添加转谷氨酰胺酶(TGase)、海藻酸钠、卡拉胶、魔芋胶对宽体舌鳎盐溶蛋白凝胶特性的影响,本实验通过四元二次通用旋转试验设计,采用响应面法建立了宽体舌鳎盐溶蛋白凝胶保水性(Y)和凝胶强度(y)与各添加剂用量的二次多项回归方程,确定了TGase、海藻酸钠、卡拉胶、魔芋胶的适宜添加量分别为0.36%,1.00%,1.00%,0.71%,添加后舌宽体鳎盐溶蛋白凝胶网状结构致密均匀,保水性和凝胶强度分别为60.36%和55.20 g,比对照组分别增加了36.41%和351.80%。结果说明,合适的添加剂组合可以显著的增加宽体舌鳎盐溶蛋白凝胶的保水性和凝胶强度。 相似文献

In this study, the kinetics of soil organic matter (SOM) dissolution from soil samples in different states of moisture was investigated, using a continuous extraction method. The investigation distinguished three processes of SOM dissolution. They include an initial, fast process (probably hydrophilic dissolved organic matter) and two slow, rate limited processes, which probably correspond to hydrophobic dissolved organic matter (DOM). The second process indicates a slow, continuous release of DOM, whereas the third process is determined by a power law. The rate of the third process strongly depends on temperature and state of moisture. It is diffusion limited, with the diffusion control probably being located in the solid soil organic matter. This was explained by a gel structure, which slowly forms in the hydrating SOM and allows diffusion of mobile particles of SOM. The results show the importance of considering the moisture state of SOM for the kinetics of DOM dissolution. 相似文献

A route for the immobilization of 2,4‐dichlorophenoxyacetic acid (2,4‐D) on silica gel has been developed. In the first step the precursor was prepared by reacting the silylating agent 3‐(trimethoxysilyl)propylamine with silica gel. Nitrogen analysis of this anchored compound showed the presence of 1.16 mmol of amine groups per gram of support. The herbicide was covalently bonded to the amine groups previously anchored onto silica gel. Infrared, [13C] and [29Si] NMR spectra supported the reaction between the nitrogen of the amine group of the anchored silica with the carbon on the para‐position of the aromatic ring of 2,4‐D. 将PCR-DGGE技术和典范对应分析相结合,研究了珠江口水体颗粒附着微生物群落沿环境梯度的空间分布特征及其影响因素。DGGE指纹图谱分析表明,颗粒附着微生物群落沿环境梯度表现出明显的空间演替:不同站位之间既存在共同谱带,又具有各自的特征谱带;S4和S5两个站位的微生物群落结构与其相邻站位相比,DGGE指纹图谱变化明显,是颗粒附着微生物从海淡水混合区向海水区演替的中间过渡类型。对DGGE图谱中19条主带回收、测序表明,两个序列与已培养的微生物同源性≥99%,其余17条序列均与未培养的环境微生物种群具有很高的相似性(91%~100%);变形菌门和拟杆菌门是珠江口颗粒附着微生物的优势种群,其中变形菌门为绝对优势种群(78.9%)。典范对应分析显示,氮相关营养盐水平和盐度是影响水体颗粒附着微生物群落分布格局的两个主要因素。 相似文献

Digestive enzymes of Cirrhinus mrigala (Ham.) were studied during ontogenic development. Specific amylase activity was detected in first feeding fish. The enzyme activity decreased up to day‐18 and then it increased with the age of fish to reach the highest level on day‐34. Protease activity was 28.61 ± 8.90 mU mg protein?1 min?1 on day‐4 and increased with the age throughout the study period. Trypsin activity was 31.86 ± 1.12 mU mg protein?1 min?1 on day‐4. The activity decreased up to day‐10 and from day‐12 onwards increased up to day‐26. Chymotrypsin activity was 14.56 ± 2.74 mU mg protein?1 min?1on day‐4 and constantly increased up to day‐26. A significant increase in lipase activity was observed between days‐24 and 34. SDS‐PAGE and substrate SDS‐PAGE showed the diversity of protein (17.4–127.8 kDa) and protease activity bands (16.6–88.8 kDa) during ontogenesis. Soybean trypsin inhibitor, phenyl methyl sulphonyl fluoride, N‐α‐p‐tosyl‐l ‐lysine chloromethylketone and N‐tosyl‐l ‐phenylalanine chloromethylketone inhibited the protease activity up to 79.72–97.21, 65.55–94.83, 45.41–75.31 and 40.78–64.72%, respectively. Inhibition study in substrate SDS‐PAGE revealed the abundance of serine proteases and the presence of isoforms of trypsin and chymotrypsin. Ethylenediamine‐tetraacetate showed 5.56–22.78% inhibition of metal ion‐specific enzyme activity. 相似文献

We examined the ability of salmon growth hormone (sGH) in sodium alginate (SA) gel to accelerate the somatic growth of juvenile
abalone, Haliotis discus hannai, by feeding. After the feeding of sGH at 50 μg or 100 μg in 350 mg SA gel, immunoreactivity to sGH in body fluid was maximal
after 12 h, and was still detectable at 24 h. No immunoreactivity was observed in the control group. Following the feeding
of sGH at 0.5 mg or 5 mg/8 g of SA gel at 7- and 14-day intervals, there was a greater increase in shell length and body weight
than in the control. In abalone, sGH can be transported from food into the circulatory system and subsequently improve somatic
