采用MTT法、DNA ladder分析、流式细胞仪分析等方法,离体研究玉米赤霉烯酮对小鼠脾淋巴细胞增殖与凋亡的影响,结果发现:ZEA对离体培养LPS活化小鼠脾淋巴细胞增殖具有显著抑制作用(P<0.05),且抑制强度与其剂量和处理时间均呈依赖性关系;小鼠脾淋巴细胞经ZEA处理后,出现DNA断裂等典型细胞凋亡特征;ZEA对离体培养LPS活化小鼠脾淋巴细胞具有显著促凋亡作用(P<0.0 5),且促进强度与其剂量呈依赖性关系。这些结果表明,玉米赤霉烯酮对小鼠脾淋巴细胞有直接毒害作用。  相似文献

为探讨硒缺乏对鸡空肠中肥大细胞(MC)的形态学和超微结构的影响,将10只鸡随机分成低硒试验组(饲喂低硒日粮0.032 mg/kg)和正常对照组,在75日龄时断头处死立即取出空肠,进行甲苯胺蓝、长时间甲苯胺蓝、阿利新蓝-藏花红染色、阿利新蓝-高碘雪夫氏反应和醛品红-阿利新蓝5种染色并进行透射电镜观察.结果显示,采用不同染色方法所获得的MC数量、大小、形态、颜色及颗粒的黏多糖成分均不相同.电镜下,MC细胞胞浆内充盈大量的特征性颗粒,颗粒内容物呈均质状,电子密度高低不等,核圆形或椭圆形,胞质中可见高尔基复合体和较多的线粒体;试验组可见MC数量显著增加并与变性细胞紧密相邻,异染色质增多,核偏位或固缩,胞质稀疏,同时可见MC内颗粒内容物释放后的空隙或空泡.因此,组化染色提示肥大细胞存在不同亚型;试验组MC数量明显增加,这提示MC是重要的免疫细胞之一,在机体的免疫调节中发挥着重要的作用.  相似文献

A 6‐year‐old male neutered Australian Shepherd dog was presented for evaluation of a subcutaneous mass on the plantar aspect of the proximal left metatarsus. Fine‐needle aspirate smears contained numerous plump spindle cells and large multinucleated cells amongst a considerable amount of pink extracellular matrix. Histopathologic diagnosis of the tissue obtained during initial biopsy and eventual surgical cytoreduction of the mass was a benign giant cell tumor of the tendon sheath (GCTTS). Immunohistochemically, the synovioblastic neoplastic cells were diffusely strongly positive for vimentin and S‐100, were multifocally moderately positive for cytokeratin AE1/3, and were negative for CD18, muscle‐specific actin (MSA), and melanoma‐associated antigen (mutated) 1 (MUM‐1). The dog recovered from surgery and underwent definitive radiation therapy to treat the local residual disease. Eight months later, the mass had not recurred. The diagnosis of GCTTS in this case supports previously published reports describing GCTTS as a relevant disease entity in dogs, and provides the first documentation of cytologic findings with this tumor. Further investigation is needed to correlate pathologic features with clinical behavior and response to therapy in dogs.  相似文献

The response of advanced stage cutaneous squamous cell carcinomas (SCC) following treatment with photodynamic therapy (PDT) has been poor. It was the aim of this pilot study to determine whether an increase in the delivered fluence (i.e. energy density) would improve the duration of tumour remission in cats with advanced-stage SCC. Tumours were treated with aluminium phthalocyanine tetrasulphonate (AlPcS4) PDT at a fluence of either 100 J cm−2 or 200 J cm−2 and tumour response was evaluated at regular intervals. Those feline tumours treated with a fluence of 100 J cm−2 ( n = 8) had a significantly shorter median remission duration (69 days; range 0–619 days) than those feline tumours treated with 200 J cm−2 ( n = 6; 522 days; range 151–1057 days). It is our conclusion that a fluence of 200 J cm−2 is well tolerated and more effective when treating cats with advanced stage cutaneous SCC.  相似文献

本研究选用39只140~150g SPF级SD大鼠作为动物内毒素血症模型,运用组织学常规HE染色和PAS染色技术,采取定性、定位和定量相结合的方法,通过观察不同试验组大鼠肠黏膜杯状细胞(GC)的数量和分布的动态变化来探讨内毒素(ET)对肠道黏膜免疫系统的影响。结果表明,ET能够破坏肠道黏膜的正常结构,影响肠黏膜的正常功能,同时能够在不同程度上降低GC数量,从而破坏肠道黏膜免疫系统。  相似文献

This study compared concentrations of amyloid A in bovine milk with the cell-based indicators of intramammary inflammation, somatic cell count and California Mastitis Test. Mammary quarter data pertaining to 180 cows were categorised according to concentrations of serum amyloid A in the cow of origin. Ranked correlation, ranked regression and receiver operator characteristics all demonstrated acceptable agreement between milk amyloid A concentrations and cell-based indices. There were some indications of reduction in this agreement, in cows with raised concentrations of serum amyloid A. However, there were also indications that serum amyloid A did not significantly influence milk amyloid A. The results of the current study indicate that milk amyloid A exhibits good correlation with established cell-based indicators of intramammary inflammation.  相似文献

建立高表达PCV2-Cap蛋白的CHO-K1悬浮稳转细胞系,鉴定蛋白的免疫原性,为开发新型且有效防治猪圆环病毒的亚单位疫苗奠定基础。构建重组质粒pEE12.4-PCV2-Cap,转染CHO-K1细胞,通过加压筛选,有限稀释,细胞悬浮驯化及Western blot检测得到高表达Cap蛋白的悬浮稳转单克隆细胞株,并对该细胞株进行发酵,纯化得到的目的蛋白,通过小鼠免疫验证其免疫原性。结果表明PCV2-Cap蛋白能够在CHO-K1细胞中正确表达;发酵过程中活细胞密度高达6×10~6个·mL-1,细胞活力在80%以上,PCV2-Cap蛋白表达量约为370 mg·L-1;利用间接ELISA检测小鼠免疫后血清中的抗体水平,证明了利用CHO-K1细胞生产的Cap蛋白具备良好的免疫原性。本研究成功构建了表达PCV2-Cap蛋白的CHO-K1悬浮稳转细胞系,并进一步对目的蛋白进行免疫原性分析,为猪圆环病毒亚单位疫苗的开发打下扎实的基础。  相似文献

天然抗菌肽具有较强的杀菌能力,但其较高的细胞毒性会使肽的细胞选择性降低。为了提高抗菌肽的选择特异性,本研究拟探讨不同疏水性氨基酸对α-螺旋抗菌肽生物学活性的影响及其抑菌机制。笔者采用α-螺旋肽GRX_2RX_3RX_2RG作为模板,分别以疏水性氨基酸色氨酸(Try,W)、苯丙氨酸(Phe,F)、缬氨酸(Val,V)、丙氨酸(Ala,A)、亮氨酸(Leu,L)和异亮氨酸(Ile,I)来替换X位置,得到了一系列富含疏水氨基酸的肽GW、GF、GV、GI、GA和GL。本研究检测了抗菌肽的二级结构、溶血活性、抑菌活性、盐离子活性,并对其抑菌的作用机制进行了研究。结果表明:通过CD光谱试验检测出GF、GI、GA和GL在细胞膜模拟环境中均表现出典型的α螺旋结构,而GV只在TFE中呈现α螺旋结构;溶血试验结果表明,GV和GA在浓度为128μmol·L-1未表现出溶血活性,而其他肽均表现出较高的溶血活性;抑菌试验发现GV的最小抑菌浓度(MIC)的几何平均值为3.7μmol·L-1,并具有最高的治疗指数(TI);盐离子稳定性试验表明,在NH_4^+、Zn2+和Fe3+中GV对大肠杆菌25922(E.coli ATCC 25922)的抑菌能力较稳定。进一步通过扫描电镜和内膜通透性试验对抗菌肽GV的抑菌机制进行分析,结果观察到GV能够通过穿透E.coli ATCC 25922和金黄色葡萄球菌29213(S.aureus ATCC 29213)促使细胞内容物流出而导致细菌死亡,以及通过时间和剂量依赖的方式穿透细菌内膜。综合以上结果,富含Val的GV具有较高的细胞选择性和成为高效抗菌药物的发展潜力。  相似文献

Because of its widespread use and potential adverse biological effects, bisphenol A (BPA) represents one of the most studied endocrine-disrupting compounds. Within the reproductive system, ovarian granulosa cells have been documented as a target of BPA action, but no consensus has been reached about functional modifications induced by BPA. On these bases, we studied the potential disrupting effects of BPA on the main granulosa cell functional activities, also taking into account a potential interference with the ovarian angiogenic process. Ovarian granulosa cells were isolated from porcine follicles and cultured in the presence or absence of BPA at different concentrations for 48 h. Cell proliferation was studied by measuring adenosine triphosphate content. Progesterone (P4) and estradiol 17β (E2) production was determined by radioimmunoassay. Vascular endothelial growth factor (VEGF) output was quantified by an enzyme-linked immunosorbent assay. Redox status was monitored by measuring superoxide anion and hydrogen peroxide, and by determining the activities of the scavenging enzymes superoxide dismutase, catalase, and peroxidase by colorimetric methods. Granulosa cell proliferation as well as redox status resulted unaffected by BPA. Concentrations of E2 were stimulated by the lower BPA concentration, whereas they were inhibited by the larger doses tested. P4 output was decreased by all BPA concentrations. To the contrary, VEGF production was stimulated. Data indicate that BPA can interfere with reproductive activity by affecting granulosa cell steroidogenesis in vitro; furthermore, BPA can exert a promoting effect on the ovarian angiogenic process by increasing VEGF output in pigs. A disruption of this finely tuned process seems particularly relevant because of the risk of uncontrolled neovascularization.  相似文献
