The effects of the severity and timing of leaf removal(LR) on the amino acids of Sauvignon Blanc grapes and wines were studied during the 2017 growing season. High-performance liquid chromatography(HPLC) was used to analyze the amino acids profiles of grape berries and wines. The basal leaves were removed at three time points(40, 56 and 72 days after flowering, named LR40, LR56 and LR72, respectively) at two severity levels(one at which the first, third, and fifth basal leaves of each shoot were removed(50% level); and another at which the first six basal leaves were removed(100% level)). The results showed that leaf removal had little impact on total soluble solids(°Brix), titratable acidity, pH or berry weight. The LR72-50% treated grapes had higher berry weight, titratable acidity and °Brix than those of the other treatments. The highest concentrations of total amino acids and of total amino acids except proline were detected in LR72-50% treated grapes(2 952.58 and 2 764.36 mg L~(-1), respectively); the lowest were detected in LR72-100% treated grapes(2 172.82 and 2 038.71 mg L~(-1), respectively). LR72-50% treatment significantly promoted the synthesis of aspartic acid, serine, arginine, alanine, aminobutyric acid and proline at both severity levels for grapes, the concentrations of all of these amino acids were increased relative to the control concentrations. The LR72-50%, LR40-100% and LR72-100% treated wines had higher total amino acids concentrations and higher concentrations of some individual amino acids, such as arginine, alanine and serine, than did the control wines. Of all the amino acids studied, glycine, tyrosine, cysteine, methionine and lysine were not significantly influenced by the timing or severity basal defoliation in grapes and wines. The present study reveals the effects of the timing and severity of leaf removal on the amino acids profiles of grapes and wines.  相似文献

为降低油菜种子中的亚油酸和亚麻酸含量并使之在异交条件下保持基本稳定,以高油品种CY2作为受体亲本,采用RNA干涉技术沉默油菜内源Bn FAD2基因的表达,抑制油酸脱饱和反应。育成的3个独立转基因株系的亚油酸和亚麻酸含量下降到6%以下,其中亚油酸含量较受体亲本降低78.2%~86.5%,亚麻酸含量降低53.4%~65.8%。将3个转基因株系与2个亚油酸和亚麻酸含量较高的常规品种正反交,F1种子中这2种脂肪酸的含量依然保持在与转基因亲本相仿的低水平。由此可知,通过RNA干涉技术沉默Bn FAD2基因的表达可赋予油菜低亚油酸和亚麻酸含量的异交稳定特性。这一特性的获得对于今后培育异交稳定的高油酸油菜新品种具有重要意义。  相似文献

The long‐term effects of salt stress (11 dS m?1) and drought stress (35 % WHC) were investigated for two maize genotypes, focusing on the relation between metabolic changes around the time of pollination and the impact on yield determinants at maturity. The relatively salt‐resistant hybrid Pioneer 3906 and the relatively drought‐resistant hybrid Fabregas were compared. The experiments were conducted in large plastic containers in a vegetation hall in two consecutive years (2011 and 2012). Plant height and leaf area were significantly reduced under both stress conditions. The transpiration rate was only slightly reduced under drought stress; but under salt stress, a significant reduction occurred 40–53 days after sowing. As a significant increase in sucrose concentrations was observed in the salt‐treated maize kernels 2 days after pollination, the availability of assimilates was not limiting and the plants could afford to save water by reduced stomatal opening. Although under both stress conditions the soluble acid invertase activity was reduced 2 days after pollination, concomitantly, an increase in hexose concentrations was observed. Thus, in these experiments, the delivery of hexoses by acid invertase activity did not limit kernel development. Differences in grain yield at maturity between salt and drought stress were most likely caused by salt‐specific effects (Na+ toxicity), Fabregas being more affected than Pioneer 3906.  相似文献

This experiment was conducted to study the effects of dietary lactic acid bacteria (LAB) and fructooligosaccharides (FOS) on growth performance, nutrient digestibility and serum biochemical indices of Arctic foxes (Alopex lagopus) during early growth period. Sixty male post-litter Arctic foxes with the average body weight of (1.16±0.15) kg and at the age of (45±3) days were randomly assigned to 4 groups with 3 replicates per group and 5 foxes per replicate. The foxes were fed the following diets, respectively: basal diet (control group), basal diet+3×109 CFU / kg LAB (LAB group), basal diet+5 g / kg FOS (FOS group) and basal diet+3×109 CFU / kg LAB+5 g / kg FOS (LAB+FOS group). The experiment lasted for 30 days. The results showed as follows: 1) on the days 1 to 15, the diarrhea rate of foxes in the LAB group and LAB+FOS group was significantly lower than that in the control group and FOS group (P<0.05). On the days 16 to 30, the average daily gain of foxes in the LAB+FOS group was significantly higher than that in the control group and FOS group (P<0.05), but had no significant difference with LAB group (P>0.05); the diarrhea rate of foxes in the LAB+FOS group was significantly lower than that in the control group and FOS group (P<0.05), but had no significant difference with LAB group (P>0.05); compared with the FOS group, the feed to gain ratio of foxes in the LAB+FOS group was significantly decreased (P<0.05). 2) The dry matter digestibility of foxes in the LAB + FOS group was significantly higher than that in the control group and FOS group (P < 0.05), but had no significant difference with LAB group (P>0.05). There were no significant differences in nitrogen intake, fecal nitrogen, urine nitrogen, nitrogen deposition, net protein utilization and protein biological value among all groups (P>0.05). 3) Serum total protein (TP) content of foxes in the LAB+FOS group was significantly higher than that in the FOS group (P<0.05), but had no significant difference with control group and LAB group (P>0.05). Serum albumin (ALB) content of foxes in the LAB group was significantly higher than that in the control group and FOS group (P<0.05), but had no significant difference with LAB+ FOS group (P>0.05). Serum urea nitrogen (UN) content of foxes in the LAB+FOS group was significantly decreased compared with the control group (P < 0. 05). In conclusion, the combined supplementation of 3 × 109 CFU / kg LAB and 5 g / kg FOS in the diet is beneficial to reduce diarrhea rate, improve dry matter digestibility, improve body immune level, and promote the growth of Arctic foxes during early growth period. © 2023 Chinese Journal of Urology. To find a connection between polyamines and various protective effectors involved in the development of cold tolerance, eight different cereal genotypes, including wheat, barley and oat species, were investigated during the acclimation phase to low temperature. Exposure to low temperature induced different changes in the levels of polyamines, and other signalling molecules, such as salicylic acid and abscisic acid, and of other protective compounds, namely flavonols, sugars and antioxidant enzyme activity, and in the lipid composition of certain membrane factions. The most remarkable differences were observed in the oat varieties compared to the other cereal genotypes, which was manifested in the lack of spermidine accumulation and of decrease in trans‐Δ3‐hexadecanoic acid content, in lower initial and not cold‐inducible abscisic acid content and guaiacol peroxidase activity after cold treatment. Correlation analysis revealed that spermidine shows strong positive relationship with flavonols, abscisic acid and ascorbate peroxidase, while was in negative relationship with trans‐Δ3‐hexadecanoic acid. These results suggest that spermidine may have a crucial role in the cold acclimation signalling processes in cereals.  相似文献

Increasing pressures from agriculture and urbanization have resulted in drainage of many floodplains along the eastern Australian coastline, which are underlain by sulphidic sediments, to lower water tables and reduce soil salinity. This leads to oxidation of the sediments with a rapid decline in pH and an increase in salinity. Accurately mapping soil salinity and pH in coastal acid sulphate soil (CASS) landscapes is therefore important. One required map is the extent of highly acidic (i.e. pH < 4.5) areas, so that the application of alkaline amendments (e.g. lime) to neutralize the acid produced can be specifically targeted to the variation in pH. One approach is to use digital soil mapping (DSM) using ancillary information, such as an EM38, digital elevation models (DEM – elevation) and trend surface parameters (east and north). We used an EM38 in the horizontal (EM38h) and vertical (EM38v) modes together with elevation data to develop multiple linear regressions (MLR) for predicting EC1:5 and pH. For pH, best results were achieved when the EM38 ECa data were log‐transformed. By comparing MLR models using REML analysis, we found that using all ancillary data was optimal for mapping EC1:5, whereas the best predictors for pH were north, log‐EM38v and elevation. Using residual maximum likelihood (REML), the final EC1:5 and pH maps produced were consistent with previously defined soil landscape units, particularly CASS. The DSM approach used is amenable for mapping saline soils and identifying areas requiring the application of lime to manage acidic soil conditions in CASS landscape.  相似文献

在300 mmol/L Na Cl胁迫条件下,研究了水杨酸对豇豆[Vigna unguiculata(L.)Walp]幼苗生长发育的影响。结果表明,经水杨酸处理的豇豆幼苗株高、茎粗明显提高,促进了幼苗的形态构建。并且幼苗的超氧化物歧化酶和过氧化物酶活性均显著高于对照,丙二醛含量和电解质渗透率显著或明显降低;水杨酸在适宜的施用浓度范围内能够缓解豇豆幼苗的盐害症状,提高抗盐能力,对豇豆幼苗的生长有一定的促进作用,当水杨酸的处理浓度为100~150 mg/L时,作用效果最明显。  相似文献

以笃斯越橘(Vaccinium uliginosum)成熟果实为试材,以钼蓝比色法测定笃斯越橘果实还原型抗坏血酸(Reduced ascorbic acid,As A)含量进行条件优化。结果表明,3%偏磷酸-乙酸用量为100μL、5%硫酸和5%钼酸铵用量均为200μL、以去离子水补充至1 500μL,30℃干热恒温浴显色40 min,取出室温下放置1 h后,在700 nm下测定,所得数据稳定,准确性适合用于笃斯越橘果实As A含量的测定;测得含量为(40.20±6.23)mg/100 g FW。  相似文献

Routine agricultural practices are heavily dependent on the use of surfactants, many of which are toxic to humans and detrimental to the environment. In proof of concept work we have previously shown the potential of nanostructured liquid crystalline particles (NLCP) to safely interact with plant leaf cuticular surfaces with minimal impact on epicuticular waxes. Here we demonstrate the use of NLCP to effectively deliver the auxin herbicide 2,4-dichlorophenoxyacetic acid (2,4-D) to plant leaves in laboratory and field studies. In the laboratory, the physiological stress responses of lupin, Lupinus angustifolius (L.) (Fabaceae) towards NLCP spray applications were shown to be much reduced in comparison with application of two common surfactants. Phytotoxicity assays of 2,4-D loaded NLCP were used to validate the herbicidal effects on Arabidopsis thaliana (L.) Heynth. (Brassicaceae) and established a similarity with that of surfactant assisted 2,4-D delivery when tested at a concentration of 0.1%. Field trials were conducted to test the efficacy of NLCP-assisted delivery of 2,4-D in comparison with commercial surfactants for the control of the invasive weed wild radish, Raphanus raphanistrum (L.) (Brassicaceae), in wheat, Triticum aestivum (L.) (Poaceae) crop fields. Compared against Estercide 800, a commercially available 2,4-D formulation, NLCP assisted delivery of 2,4-D was effective at low concentrations of 0.03% and 0.06%. The crop yield remained similar for all the tested concentrations and formulations of 2,4-D loaded NLCP and Estercide 800. This is the first report to directly show that, as an alternative to conventional methods, NLCP can be used under both laboratory and field conditions to successfully delivery an agrochemical.  相似文献

The weight and composition of soybean seeds (Glycine Max L. Merrill) depend on changes in carbon and nitrogen assimilate supply during grain filling. Soybean pods and seeds are green, evidencing their capacity to capture light. However, the current physiological knowledge does not consider any effect of incident solar radiation reaching the pods on seed weight and composition. The objective of this work was to investigate the response of seed weight and composition to changes in assimilate supply from leaves, to the incident solar radiation reaching the pods and to the combination of both, changes in assimilate supply from the leaves and incident solar radiation on pods of soybean plants. Field experiments were performed during two growing seasons at Balcarce, Argentina. Treatments modified the amount of assimilates supplied by the leaves (plant shading, defoliation), the solar radiation reaching the pods (pod shading) or both (defoliation and pod shading) during seed filling. Plant shading and defoliation reduced seed weight, oil concentration and oil and protein content and increased the concentration of saturated and poli-unsaturated fatty acids while reduced oleic acid percentage. Pod shading increased the concentration of stearic acid and reduced the concentration of linolenic acid. When pods were shaded on defoliated plants, seed weight and oil and protein content decreased while fatty acid composition was similar to values obtained under defoliation treatment. Based on these results, a conceptual model that considers photoheterotrophic nature of reproductive structures of soybean is proposed. Seed weight, oil and protein content and oil fatty acid composition depended on assimilate availability for the seeds. The response of oil and protein content to assimilate supply depended on whether leaves were present or not. The effect of solar radiation incident on pods depended on the amount of assimilates available for the seeds: (i) when carbon allocated was low (defoliation treatments), pods contributed to seed carbon economy but solar radiation incident on them did not affect fatty acid composition; (ii) when carbon allocated to the seeds was high (intact plants), contribution of pods to seed carbon economy was not significant, but the amount of solar radiation incident on pods produced significant changes in fatty acid composition.  相似文献
