Body condition and haematological indices provide powerful information when assessing wildlife health. Reference intervals for these indices can facilitate wildlife management, and would benefit initiatives to save the Critically Endangered geometric tortoise (Psammobates geometricus). We collected data from 126 geometric tortoises to establish baseline values reflecting variation over four seasons (spring 2000 to winter 2001) and among three groups (female, male and juvenile). We measured body condition index (BCI; mass to shell volume), blood urea nitrogen (BUN), plasma chloride, packed cell volume (PCV), haemoglobin concentration (Hb) and red blood cell count (RBC), and used PCV, Hb and RBC to calculate erythrocyte indices. BCI correlated poorly with haematological measures but had a strong inverse relationship with BUN. BCI did not vary among groups, but all groups had low condition indices in autumn, the end of the dry season. High BUN and chloride values in autumn indicate dehydration, particularly in males. Males had the highest PCV, Hb and RBC values, especially during summer and autumn when they moved long distances, likely pursuing mates. Female and juvenile indices were similar, except that female Hb was higher than juvenile Hb, possibly to meet female energy needs associated with their large size and reproductive metabolism. Low Hb levels in winter coincided with low temperatures and reduced movements. Our results illustrate how intrinsic and extrinsic factors influence the physiology of geometric tortoises and provide reference intervals to monitor their health.  相似文献

This experiment was conducted to investigate the effect of feeding phytoestrogens on milk composition, blood biochemical index and antioxidant capacity of dairy cows.A total of 40 Holstein cows were selected and randomly divided into 4 groups by health, age and lactation stage. The cows in control group was fed total mixed rations(TMR), and the other in 3 trial groups were fed TMR supplemented with 2.5 g dainzein(group Ⅰ), 35 g formononetin (group Ⅱ), 1.25 g dainzein and 17.5 g formononetin (group Ⅲ), respectively. The total duration of the trial was 127 d, the first 7 d of the trial was the preliminary period and the formal trial lasted for 120 d. The milk samples were collected in the trial of 0, 30, 60, 90, 120 d, respectively, and determination of milk composition. The blood samples were collected by jugular vein in the trial, and plasma was separated by 15 min, 3 500 r/min centrifugation, and saved at -20 ℃ for later use. The result showed as follows: Compared with the control group, the milk yield and milk protein yield in groups Ⅰ and Ⅲ were significantly increased (P< 0.05),the lactose yield in groups Ⅰ,Ⅱ and Ⅲ were significantly increased (P< 0.05), the T-AOC activity of blood in group Ⅲ was significantly increased (P< 0.05), the GSH-Px activity of blood in groups Ⅱ and Ⅲ were significantly increased (P< 0.05), the MDA activity of blood in groups Ⅰ and Ⅱ were significantly decreased (P< 0.05), the milk fat yield,lactose rate and TP, ALB, GLB of blood were not significantly increased (P> 0.05),the CAT, SOD activity of blood in trail groups had a upward trent. It was concluded that feeding phytoestrogens could enhance the milk yield, milk protein yield, lactose yield, the body's antioxidant capacity and immune function of Holstein cows.  相似文献

为了探讨种植业结构调整对于我国环境的影响,本研究运用生命周期评价方法,计算了甘肃省民勤县农户水平2014与2015年从农资生产到农户入仓范围生产1 kg玉米籽粒及1 kg紫花苜蓿鲜草的环境影响,并使用基于蛋白质和热量的计量单位—食物当量(FEU),比较分析了1个FEU玉米籽粒和紫花苜蓿生产的全生命周期环境影响差异。结果表明,生产1 kg玉米籽粒和1 kg紫花苜蓿鲜草全生命周期的一次性能源消耗(PED)分别为9.35和1.22 MJ,水资源消耗(WU)分别为889.33和144.37 kg,矿物和化石资源消耗(DAR)分别为0.13和0.02 kg antimony-eq,气候变化潜值(GWP)分别为1.21和0.10 kg CO2-eq,可吸入无机物(RI)分别为4.23×10-3和1.88×10-4 kg PM2.5-eq,光化学臭氧合成(POFP)分别为2.41×10-3和1.71×10-4 kg NMVOC-eq,环境酸化潜值(AP)分别为8.55×10-3和8.03×10-4 kg SO2-eq,淡水富营养化(FEP)分别为1.20和0.09 kg P-eq,生态毒性(ecotoxicity)分别为1.26×10-2和1.49×10-3 CTU。1个FEU紫花苜蓿生产的PED、WU、DAR、GWP、RI、POFP、AP、FEP和ecotoxicity则分别为玉米籽粒的20.50%、25.43%、21.08%、12.99%、6.98%、11.15%、14.76%、12.31%和18.58%。因而考虑到苜蓿的食物-经济比较优势,目前应给予其不少于粮食作物的种植补贴。并且如果将我国的部分玉米种植改为苜蓿种植,则是最便捷、经济的既能满足我国食物结构需求,又能减少农业生产的资源消耗与环境污染的措施。本研究同时也为在我国深入开展粮改饲提供了一定的立论基础。  相似文献

采用3-(4,5-二甲基-2-噻唑)-2,5-二苯基溴化四唑(MTT)检测法、荧光显微镜细胞形态观察法和流式细胞仪(flow cytometry,FCM)技术,研究了辣椒碱对离体培养斜纹夜蛾Spodopt-era litura(SL)细胞增殖及凋亡的影响。结果表明:辣椒碱对SL细胞24 h的IC50值为46.89μg/mL,与鱼藤酮差异不显著。40μg/mL辣椒碱处理SL细胞24h后,细胞形态明显不规则,呈现出凋亡的典型症状。10、20和40μg/mL辣椒碱处理SL细胞48h后,细胞早期凋亡率分别为13.56%、18.61%和3.81%,而晚期凋亡和坏死率分别为23.78%、34.28%和61.72%。辣椒碱处理细胞48h后,细胞周期变化明显,细胞生长阻滞于G2/M期。表明辣椒碱对斜纹夜蛾细胞具有良好的抑制作用,能致细胞凋亡并有一定的浓度依赖性。  相似文献

Background: Testing for canine blood types other than dog erythrocyte antigen 1.1 (DEA 1.1) is controversial and complicated by reagent availability and methodology. Objectives: The objectives of this study were to use available gel column technology to develop an extended blood‐typing method using polyclonal reagents for DEA 1.1, 1.2, 3, 4, 7, and Dal and to assess the use of gel columns for cross‐matching. Methods: Dogs (43–75) were typed for DEA 1.1, 1.2, 3, 4, 7, and Dal. Methods included tube agglutination (Tube) using polyclonal reagents, a commercially available DEA 1.1 gel column test kit (Standard‐Gel) using monoclonal reagent, and multiple gel columns (Extended‐Gel) using polyclonal reagents. Blood from 10 recipient and 15 donor dogs was typed as described above and cross‐matched using the gel column technique. Results: Of 43 dogs typed for DEA 1.1, 23, 25, and 20 dogs were positive using Standard‐Gel, Extended‐Gel, and Tube, respectively. Typing for DEA 1.2 was not achievable with Extended‐Gel. For 75 dogs typed for DEA 3, 4, and 7, concordance of Extended‐Gel with Tube was 94.7%, 100%, and 84%, respectively. Dal, determined only by Extended‐Gel, was positive for all dogs. Post‐transfusion major cross‐matches were incompatible in 10 of 14 pairings, but none were associated with demonstrable blood type incompatibilities. Conclusions: Gel column methodology can be adapted for use with polyclonal reagents for detecting DEA 1.1, 3, 4, 7, and Dal. Agglutination reactions are similar between Extended‐Gel and Tube, but are more easily interpreted with Extended‐Gel. When using gel columns for cross‐matching, incompatible blood cross‐matches can be detected following sensitization by transfusion, although in this study incompatibilities associated with any tested DEA or Dal antigens were not found.  相似文献

Spectral waveform analysis of blood flow velocity in the major arteries of six healthy, conscious immature micropigs was determined using Doppler ultrasonography. Doppler spectral tracings were recorded from the external iliac artery, femoral artery, and renal arcuate artery. Tracings were also taken from three parts of the common carotid artery and two parts of the abdominal aorta. Spectral Doppler parameters included peak systolic velocity, early diastolic velocity, peak systolic velocity-to-end diastolic velocity ratio, resistive index, and pulsatility index. In addition, the diameter of major arteries and indirect blood pressure were measured. These results from spectral Doppler analysis in major arteries may be useful as reference ranges in the future studies of vascular hemodynamics in immature micropigs.  相似文献

本试验选取16只3~4 kg健康泌乳家兔,通过人工造模的方法诱发其乳腺发炎,建立临床型乳腺炎模型。然后分别在造模前1 h和造模后4、12、24、36、487、2 h采集血液0.5 mL、血清1 mL,进行血常规、血生化检测和炎性细胞因子测定(ELISA)。结果显示:①外周血白细胞总数在造模后4 h显著高于造模前水平(P0.05),12~72 h内极显著高于造模前水平(P0.01);红细胞总数、红细胞压积、血红蛋白含量在造模前至造模后72 h内无显著差异(P0.05)。②血清磷离子含量、血清总胆红素含量、血清直接胆红素含量在造模前至造模后72 h内无显著差异(P0.05);血清钙离子含量在造模后4 h显著低于造模前水平(P0.05),12~72 h内极显著低于造模前水平(P0.01)。③血清TNF-α和IL-6含量造模后开始升高,12~72 h内极显著高于造模前水平(P0.01)。结果证明白细胞总数、血清钙离子含量、血清TNF-α和IL-6含量的变化可作为急性乳房炎炎症发展情况的指标。  相似文献

AIM: To investigate the effects of double thymidine deoxyribonucleoside (TdR) blocking on the cell cycle of human gastric cancer SGC-7901 cells.METHODS: SGC-7901 cells in the logarithm period were selected in the study and treated with TdR at concentration of 2 mmol/L, 4 mmol/L or 8 mmol/L for 15 h as the first blocking. After incubation in TdR-free medium for 10 h, the cells were treated with TdR at same concentrations again for another 15 h as the second blocking. The blocked cells were released by washing in fresh medium twice and incubation in TdR-free medium containing 10% fetal bovine serum for 12 h. The cells were collected and the cell cycle was detected by flow cytometry.RESULTS: By double TdR (2 mmol/L, 4 mmol/L and 8 mmol/L) blocking to synchronize the cell cycle, the cells in G0/G1 phase accounted for 77.3%, 77.5% and 77.0%, respectively. After further incubation for 12 h in TdR-free medium, the proportion of the cells in each phase of the cell cycles returned to normal range. CONCLUSION: The method of double TdR blocking is an ideal access in short term to acquire a large number of cells in G0/G1 phase.  相似文献
