Bovine somatic cell nuclear transfer (SCNT) is an important and powerful tool for basic
research and biomedical and agricultural applications, however, the efficiency of SCNT has
remained extremely low. In this study, we investigated the effects of cathepsin B
inhibitor (E-64) supplementation of culture medium on in vitro
development of bovine SCNT embryos. We initially used three concentrations of E-64 (0.1,
0.5, 1.0 μm), among which 0.5 μm resulted in the highest rate of blastocysts production after in
vitro fertilization (IVF), and was therefore used for further experiments.
Blastocyst development of SCNT embryos in the E-64 treatment group also increased relative
to the control. Moreover, the cryosurvival rates of IVF and SCNT blastocysts were
increased in E-64 treatment groups when compared with the control. On the other hand, we
found that IVF and SCNT blastocysts derived from E-64-treated groups had increased total
cell numbers and decreased apoptotic nuclei. Furthermore, assessment of the expression of
apoptosis-related genes (Bax and Bcl-xL) in bovine IVF and SCNT blastocysts treated with
E-64 by real-time RT-PCR analysis revealed suppressed expression of the pro-apoptotic gene
Bax and stimulated expression of the anti-apoptotic gene Bcl-xL. Taken together, these
finding indicate that addition of E-64 to embryo culture medium may have important
implications for improving developmental competence and preimplantation quality in bovine

在福建省将乐国有林场,通过连续3a对引自南京林业大学培育的体细胞胚胎发生杂种马褂木密度试验幼林进行观测研究.结果表明体细胞胚胎发生杂种马褂木适生性强,3a生林分保存率达到99%以上.林分生长良好,3a生时平均树高、胸径分别为2.26 m和2.3 cm,具有良好的推广应用前景.不同造林密度对体细胞胚胎发生杂种马褂木幼林生长总体而言有着显著影响,这种影响在林分2a生后开始得以体现,3a生时影响达到极显著水平. 相似文献

Increased endothelin-1 (ET-1) plasma concentration in human infants is associated with persistent pulmonary hypertension of the newborn, a problem also identified in calves derived from somatic cell clone technology. Increased ET-1 also is present in the amnionic fluid and plasma of the infant and mother in preeclampsia, a condition associated with abnormal placentation. Abnormalities in placentation are identified in clone calves. We measured ET-1 in fetal fluid, calf plasma, and surrogate dam plasma in 40 clone calves at the time of term delivery. Calves were subsequently identified as being either oxygen treated (O2) or non-oxygen treated based on their postpartum clinical course. Fetal fluid ET-1 concentration greater than 1.4 ng/mL carried a 3-fold increase in odds of the calf being treated with oxygen. Maternal plasma ET-1 concentration was greater in the O2 group (13 pg/ mL: [8-23 pg/mL] versus 25 pg/mL [12-40 pg/mL]; median, 25-75 percentile). Plasma ET-1 concentration in calves was not significantly different between groups. Fetal fluid ET-1 may serve as a marker for neonatal disorders of oxygenation in clone calves and the source of ET-1 may be the placenta. 相似文献

Summary Extensive pollinations were made in attempts to produce selected intergeneric or interspecific hybrids using Nicotiana and Petunia species that exhibit potential for somatic hybridization. Pollination between pairs of species was done by standard and bud-pollination methods. Nicotiana alata or N. tabacum when pollinated reciprocally with selected Petunia species failed to produce hybrids. Likewise, no interspecific hybrids were obtained between Petunia parviflora and 4 Petunia species or P. hybrida. 调查分析了大庆地区四个规模化牧场近3年奶牛DHI数据。结果表明,乳体细胞评分均值为5.05(其乳体细胞数均值为49万·mL-1)。75.75%的奶牛个体,其生鲜乳体细胞数小于50万·mL-1。胎次、泌乳阶段、自然月份等因素影响奶牛生鲜乳体细胞数。调查结果为牛场奶牛综合管理及制定科学可行的乳房炎防治措施提供依据。 相似文献

利用花楸树人工控制授粉后的未成熟合子胚进行体细胞胚的诱导培养,通过石蜡切片、扫描电镜和透射电镜对合子胚和体细胞胚的发生发育过程进行组织、细胞观察。结果表明:1)体细胞胚发生进程与合子胚相似,均经历了球形期、心形期、鱼雷形期,最终发育成成熟胚;2)直接发生和间接发生在愈伤组织表面的体细胞胚均为单细胞起源,间接发生在愈伤组织内部的体细胞胚存在单细胞起源,同时还存在可能的多细胞起源;3)子叶和愈伤组织表面发生的体细胞胚在发育早期均具明显的胚柄,鱼雷形胚期胚柄退化;4)愈伤组织内部发生的体细胞胚没有胚柄;5)与合子不同,胚性细胞的第1次分裂为均等分裂;6)多细胞原胚周围降解的细胞具有细胞程序性死亡的形态特征。 相似文献
