In drought areas, in which topsoil moisture is low, barley seeds are generally sown in the subsoil (deep-seeding). In order
for the shoots of germinating seeds to emerge from the deep soil cover, the coleoptile and the first internode of the seedlings
must elongate as an adaptive response to deep-seeding. Here, we have mapped quantitative trait loci (QTLs) for these adaptive
characteristics. Elongation of the coleoptile and first internode was investigated using seeds sown under two soil cover conditions:
at a depth of 9 cm beneath a soil mixture; and, at a depth of 12 cm beneath vermiculite. We identified multiple alleles for
increased coleoptile and first internode elongation using a doubled haploid population of 150 lines generated from a cross
between the barley cultivars Harrington and TR306. Composite interval mapping analyses of the data revealed two moderate and
eleven small effect QTLs, with at least one QTL on each chromosome. The QTLs on chromosomes 5H and 7H had moderate effects
on coleoptile elongation (18.5–27.6% of PVE: phenotypic variance explained; 2.6–3.2 mm of Add: additive effect) and first
internode elongation (PVE: 16.6–19.6%; Add: 3.1–3.2 mm). The small effect QTLs showed PVEs of less than 15% and an Add range
of 1.2–3.2 mm for both characters. A marker assisted selection approach, using markers linked to the QTLs for seedling elongation
at deep-seeding, may eventually enable development of drought tolerant barley hybrids. 相似文献

Sprouting can be an important mechanism of regeneration in harvested cypress (Taxodium sp.) wetlands; however, limited information exists on factors influencing sprouting. We examined the influence of harvesting practices, tree size, and site conditions on stump sprouting in eight pondcypress (Taxodium distichum var. nutans (Ait.) Sweet) wetlands in Florida, USA. Stumps were cut at three heights: 20 cm below high water level, at high water, and 40 cm above high water. Half of each site was harvested using a sawhead feller and the other half using shearhead. We monitored sprouting for 2 years post-harvest. Stump height affected first year sprouting, but the effect disappeared during year 2. Felling equipment did not affect sprouting; however, the shearhead caused greater stump damage. Sprouting declined with increasing parent tree size; percentage of stumps with sprouts after 2 years was 68% for trees 10–14 cm dbh and 28% for trees 20–24 cm dbh. Stump sprouting ranged among sites from 23 to 54% after 2 years. Results indicate that sprouting may be an important, but inadequate, form of regeneration. At sites with short hydroperiods, dense understories, and/or predominance of trees greater than 20 cm dbh, reduced sprouting may occur, and planting seedlings may be beneficial. 相似文献

In species with seeds losing viability shortly after dispersal and exhibiting inherently low germination, quick decisions are required with respect to seeds that should be selected to maximize germination success and vigorous growth of seedlings. In ‘hollong’ (Dipterocarpus macrocarpus Vesque), I address the following questions: (a) are seeds that germinate randomly distributed within a seed population, (b) are subpopulations of viable and non-viable seeds separable, (c) does seed size predict which seeds germinate and (d) does seed size predict time required for germination and seedling vigour? Two estimators of seed size, diameter and weight, demonstrated a significant positive linear relationship endorsing assumption that accumulation of mass increases with increase in diameter of seeds. A threshold for selection of potential seeds for germination could not be derived from diameter measurements since seeds in an entire range of diameter did or did not germinate. All seeds <11 g did not germinate, but this threshold lies at the far lower end of the weight range and allows rejection of only a few seeds and acceptance of many seeds that will not germinate. A risk of potential seeds being rejected or non-viable seeds being picked exists if selection was derived from either diameter or weight. However, viable seeds could be better predicted from a scatter-plot of diameter on x-axis and weight on y-axis. Seeds showed a fan-shaped scatter and those developing the lower blade of the fan did not germinate while those following the handle were successful. Hence, two subpopulations segregated, but with a fuzzy edge. Mean diameter and mean weight of germinated seeds were significantly greater than those of ungerminated seeds. Most ungerminated seeds were those that had relatively smaller weight compared to their diameter. Although some large diameter seeds with small weight did germinate, but failed to develop into seedlings. Germination time and seedling vigour parameters (height, leaf number and collar diameter) were correlated both with diameter and weight. However, weight appears to have mattered more than diameter in germination success and early seedling growth. 相似文献

Using field plots where rhizomania had not previously been detected, different inoculum levels of beet necrotic yellow vein virus (BNYVV) were created by application of infested soil. A susceptible sugar beet cultivar (cv. Regina) was grown for two consecutive years (1988 and 1989), in the presence or absence of drip irrigation. In soil samples taken in spring 1989, the different initial inoculum levels of 1988 could be distinguished using a quantitative bioassay estimating most probable numbers (MPNs) of infective units per 100 g dry soil. The first sugar beet crop resulted in a tenthousandfold multiplication of inoculum of BNYVV (viruliferousPolymyxa betae). Mean MPNs of BNYVV ranged from 0.6 and 7 per 100 g soil for the lowest inoculum level to 630 and 1100 per 100 g for the highest level, in plots without and with irrigation, respectively. In spring 1990, MPNs had again increased. In both years, the initial inoculum level of 1988 had a significant linear effect on log-transformed MPNs of BNYVV determined. Log-transformed MPNs for 1990 and 1989 showed a positive linear correlation, despite a decreasing multiplication ratio at higher inoculum levels. Drip irrigation during one or two years enhanced the increase in MPN of BNYVV, which was reflected by the enhancement of multiplication ratios at all inoculum levels. Analysis was made of dsRNA in 37 cultivars and species of Ribes, that were healthy, naturally affected with the virus-like diseases, blackcurrant yellows, blackcurrant infectious variegation, gooseberry veinbanding or blackcurrant reversion, or graft-inoculated with scions from such diseased plants. Various dsRNA species, differing in size (from ca. 2 to 11kbp), number and staining intensity in gels, were detected in some or all assays of all plants, including those held as virus-tested stock. In different plant tissues from individual plants, the dsRNA species were usually similar in size and number but, in some sources, the dsRNA profile from flowers and/or bark differed greatly from the profiles of dsRNA obtained from leaves. No dsRNA species were associated consistently with any of these diseases. A 499kbp cDNA probe was obtained that in Northern blot analysis was specific to a ca. 5kbp dsRNA species present in the blackcurrant cv. Baldwin. It also detected a similarly sized dsRNA species in plants of many other blackcurrant cultivars, but it did not react with a similarly sized dsRNA species in redcurrant and gooseberry tissues. The 156 amino acid sequence encoded by the cDNA was very similar to sequences in the RNA-directed RNA polymerases of virus species in the family Totiviridae, especially Saccharomyces cerevisiae viruses L-1 and L-A. The significance of these findings and the possible origin of these dsRNA species are discussed. 相似文献
