Random amplified polymorphic DNA (RAPD) was used with the objective of identifying DNA markers linked to the sclerotinia crown and stem rot (SCSR) resistance of red clover. Bulked segregant analysis was used to detect polymorphism that should be linked to SCSR resistance. Two bulks were made by pooling previously extracted DNA. Each bulk (one resistant, and the other susceptible) consisted of eight genotypes from an F2 population obtained from a cross between a susceptible and a resistant parent. A binomial model was used to select RAPD fragments with a low probability of no linkage with SCSR resistance. Four RAPD fragments were retained as candidate markers of SCSR resistance. Three are associated with resistance and one with susceptibility.  相似文献

The F2 progenies of crosses between several cowpea (V. unguiculata) lines were investigated for variation of eye pattern and seed coat colour. It was found that three (W, H, O) and five (R, P, B, M, N) major genes control eye pattern and seed coat colour, respectively. The recessive gene (GO) for restricted eye pattern enables the underlying basic white or cream seed coat colour to be observed. A similar effect is obtained with the recessive gene (rr) for colour expression. The expression of mottling (V), possibly a seed coat pattern, may for be observed when it is combined with the genes for certain eye patterns. The significance of these findings in breeding for consumer preference for specific seed coat colour is discussed.  相似文献

A spontaneous mutant with apetalous flowers was obtained from a hybrid progeny in Brassica napus. The result of genetic analysis showed that the apetalous character was controlled by only one gene locus, petalous flower exhibited incomplete dominance over apetalous flower and that its expression was not affected by cytoplasmic factors. Sixteen agronomically important characteristics of the apetalous line Apet33-10 were compared with those of its petalled near-isogenic line Pet33-10. Results from 4 years of tests indicated that there was no difference between Apet33-10 and Pet33-10 in all tested agronomic characteristics, except for the pod number of main inflorescence and second-order branches at low sclerotinia disease incidence. The Sclerotinia sclerotiorum disease severity index of Apet33-10 was significantly lower than that of Pet33-10, and correspondingly, the plot yield of Apet33-10 was increased obviously in comparison to that of Pet33-10 if sclerotinia disease was serious. The pod number of main inflorescence of Apet33-10 was significantly lower than that of Pet33-10. On the other hand, the pod number of second-order branches of Apet33-10 was significantly higher than that of Pet33-10, hence no overage difference of the pod number per plant was observed between the two lines.  相似文献

安徽省油菜菌核病菌对多菌灵的抗药性监测   总被引:8,自引:1,他引:8  
从安徽省望江、合肥、太湖、巢湖、蒙城、凤阳、灵壁、肥西、宣城、旌德等10个市县的油菜病茎上分离得到60个油菜菌核病菌菌株,并对其进行了抗药性监测。结果表明,有18株对多菌灵表现出不同程度的抗药性,上述各地均有抗药性菌株分布,但所占的比例不一,以蒙城系列(MCS)监测到的抗药性个体比例所占比例最大(66.7%),太湖(THS)最小(9.1%),但监测到THS-1对多菌灵的抗药性最强,高达743.8347mg/L。结果提示安徽省各地油菜菌核病菌已经对多菌灵产生了抗药性,且个别菌株已达到极高水平。  相似文献

Summary Twenty-four entries of wild emmer possessing temperature-sensitive genes for resistance to yellow rust were studied in the seedling stage, at two temperature-profiles, with 15 pathogenic races from 11 countries in South America, Africa, Asia and Europe. It was shown that the majority of the resistance genes in these wild emmer entries were race-specific. In most of these entries a more resistant reaction was displayed at the higher temperature-profile; however in three entries a shift in reaction towards resistance was observed with certain races but towards susceptibility with some of the other races, suggesting that two different kinds of temperature-sensitive genes were involved in each of these entries. The similarity of temperature-sensitive genes occurring in wild emmer and in cultivated wheat is discussed.  相似文献

The effect of induced autotetraploidy on the response of red clover to Sclerotinia clover rot (SCR) was studied under laboratory, glasshouse and field conditions. To assess the response in the laboratory, the detached leaf technique was employed, whereas in the glasshouse and in the field, 6—14 weeks old seedlings and adult plants, respectively, were artificially inoculated with the pathogen grain inoculum. The results of the studies showed that great genotypic variability occurs within diploid (2×)and tetraploid (4×)red clover in response to that disease. Generally, tetraploid red clover, in comparison to diploid, is more resistant to SCR. The laboratory, glasshouse and field experiments showed that the effect of induced autotetraploidy on SCR resistance is positive. The hardening of plant with light and temperature enlarged the differences in reponse to SCR between ploidy levels, family pairs and individual strains with every testing method. Other factors, like plant age and flowering time, influencing red clover response to SCR are also discussed.  相似文献

Sclerotinia sclerotiorum is considered the most devastating pathogen of sunflower grown in humid environments. In this study, progenies of partial hybrids between Helianthus maximiliani, a wild species that has been shown to be resistant to S. sclerotiorum, and H. annuus were characterized by amplified fragment length polymorphism (AFLP) analysis to identify whether there are introgressions from H. maximiliani into the cultivated sunflower at the molecular level. Wild species‐specific fragments as well as fragments not found in either parent were detected. Progenies tended to cluster together according to the original partial hybrids in the dendrogram by the use of bootstrap procedures. The progenies were studied for their reaction to S. sclerotiorum using artificial inoculation of sunflower heads. Some of the progenies showed a higher level of resistance compared with resistant inbred lines. It was possible to identify two AFLP‐fragments which seem to be linked to Sclerotinia resistance.  相似文献

Summary S1 to S5 inbred lines, derived from a maize population bred for its overall resistance to three tropical viruses, were screened for resistance to maize streak virus (MSV) by artificial plant infection using viruliferous leafhoppers. Symptoms were rated and intra-line frequency distributions studied for all pedigree inbred lines. Mortality due to MSV was very low among these inbreds. Symptoms appeared later, developed slower and were less severe than in the susceptible control hybrid. Results of a study of 500 S1 and 93 S2 lines suggested that resistance is under genetic control via a system involving loci with major genes (with dominance for resistance) controlling high to complete resistance, associated with a genetic system involving loci with minor genes controlling partial resistance. Lines expressing complete resistance to MSV were developed from 5 cycles of inbreeding and selection. Sclerotinia sclerotiorum (Lib.) de Bary is a major limiting factor for sunflower (Helianthus annuus) production. This study was conducted to (1) evaluate a large number of current sunflower lines for their reaction to Sclerotinia, and (2) determine the optimum allocation of resources for their selection. A set of 85 inbred lines was screened under artificial leaf infection in three environments. The test revealed ample variation for all resistance traits in the current germplasm. A line selected from NDBLOS, an oilseed sunflower germplasm poo, and its derivatives were most resistant to Sclerotinia. Lines derived from interspecific crosses with the wild species Helianthus tuberosus and Helianthus argophyllus also showed satisfactory Sclerotinia resistance. The evaluation of five plants per plot in two replications and four environments appears to be a reasonable allocation of resources in order to optimize the selection response.  相似文献

Summary The latent period (LP) is a crucial component of partial resistance. Five cultivars, L94, Sultan (Su), Volla (Vl), Julia (Ju) and Vada (Va), representing the known range in partial resistance and LP were crossed in a diallel, and the F1, F2 and F3 tested. The LP effectuated by the five cultivars is about 9, 101/2, 101/2, 13 and 151/2 days, respectively. The crosses Su×L94, Vl×L94 and Ju×L94 had an F2 positively skewed. Their F2 means were similar or only slightly larger than the F1 means. The F2 frequency distributions in the crosses Vl×Su, Ju×Su and Ju×Vl were normal or nearly so with F1 and F2 means similar to each other and to the mid-parent value. The crosses involving Va as a parent again showed a positive skewness but with F2 means considerably larger than the F1 moans.Most F2's ranged from the low parent to the high parent values without transgression. In the crosses Va×L94 (reported earlier) and Ju×L94 the parental values were not recovered among 216 and 154 F2 plants, respectively. The cross Ju×Va showed transgression beyond the low parent, Ju.From these data it is concluded, assuming no linkage, that seven loci are involved. The effect of this locus on LP seems considerably larger than that of the other loci. There are indications of physiological barriers, which means that LP's shorter than the one of L94 or much longer than that of Va are not possible.The effect of + genes in genotypes governing LP's close to these barriers (with very few or very many + alleles respectively) is smaller than in genotypes governing intermediate LP's.  相似文献
