Effects of lupeol and miR-145-5p on proliferation and apoptosis of prostate carcinoma cells

AIM To investigate the effect of lupeol combined with microRNA-145-5p (miR-145-5p) on the proliferation and apoptosis of prostate carcinoma LNCaP cells. METHODS After hsa-miR-145-5p and lupeol were applied to LNCaP cells for 24, 48 and 72 h, the cell viability inhibitory rate was detected by MTT assay. PI single staining plus flow cytometry was used to detect the cycle distribution. The flow cytometry with annexin V/PI double staining and TUNEL experiment were used to detect apoptosis. Transwell method was used to detect cell migration and invasion abilities. Wound healing experiment was used to detect cell migration ability. The cell colony formation assay was used to calculate the colony formation inhibitory rate. RESULTS The cells in cell control group, non-specific control group and solvent group did not show effective LNCaP cell viability inhibition, migration inhibition and invasion inhibition, while the viability, migration and invasion abilities were significantly inhibited, and early apoptosis in vitro were induced in hsa-miR-145-5p group and lupeol group. In particular, the combination (hsa-miR-145-5p+lupeol) group showed more effective proliferation inhibition than the single-drug groups. CONCLUSION Both hsa-miR-145-5p and lupeol inhibit the proliferation, migration and invasion of LNCaP cells, and induce early apoptosis in vitro. Lupeol enhances the proliferation-inhibitory and apoptosis-inducing effects of hsa-miR-145-5p on LNCaP cells.     

图像匹配技术在土地利用动态监测中的应用方法研究

利用遥感技术进行土地利用动态监测是利用多源多时相的遥感数据,利用栅格数据和矢量数据相结合对土地进行全面性的调查与监测,掌握其数量、质量、权属、利用现状等各种信息,它能够主动发现土地利用变化的信息,使我们可以及时掌握有关土地变更的各种信息。可是当我们发现了疑似变更了属性的非法建设用地等图斑时,这些变化图斑或不确定图斑,我们很难在影像上获取它们的准确的空间位置。本文以SPOT-5遥感影像为数据源,通过图像匹配技术来解决在土地利用动态监测中的非法建设用地变化图斑的空间定位的问题。     

我国高粱两大主干恢复系晋粱5号和晋辐1号的选育及利用

通过对晋粱 5号和晋辐 1号选育经验和遗传系谱的剖析 ,找出了晋粱 5号和晋辐 1号的亲缘关系及组配杂种优势大、适应性广的原因。进一步讨论了不育系和恢复系遗传基础扩大问题。     

茄子新品种龙杂茄5号的选育

龙杂茄5号是以自交系15号为母本，98—5为父本配制的早熟茄子一代杂种，果实长棒形，紫黑色，光泽度好，耐老化，果肉绿白色，细嫩，籽少，果纵径25—30cm，横径5—6cm，单果质量150—200g，每667m^2产量4000kg左右，中抗黄萎病，对褐纹病的抗性较对照强，耐低温、弱光，适于黑龙江省保护地栽培。     

5-硝基糠醛二醋酸酯的合成及其结构测定

本文研究了由糠醛制备5-硝基糠醛二醋酸酯的新方法,发现了熔点分别为91-92℃和105-106℃两种不同晶型的5-硝糠醛二醋酸酯,并测定了它们的晶体结构。     

Sequential preovulatory expression of a gonadotropin-releasing hormone-inducible gene,Nr4a3, and its suppressor Anxa5 in the pituitary gland of female rats

Functional relationship between nuclear receptor subfamily 4 group A member 3 (Nr4a3) and annexin A5 (Anxa5), which are two gonadotropin-releasing hormone (GnRH)-inducible genes, has been established while evaluating pituitary gonadotropes in relation to follicle-stimulating hormone beta (Fshb) expression. However, the physiological variations that arise due to the differential expression of these genes in the pituitary gland during rat estrous cycle remain unknown. This study aimed to evaluate the Nr4a3 and Anxa5 mRNA expression during the estrous cycle in rats in comparison with the expression of the gonadotropin subunit genes, luteinizing hormone beta (Lhb) and Fshb. Nr4a3 mRNA expression showed a single peak at 1400 h of proestrus during the 4-d estrous cycle. Anxa5 mRNA level was elevated along with increased Fshb mRNA expression after the decline of Nr4a3 mRNA until 2300 h. Lhb mRNA expression levels were not significantly changed during the estrous cycle. Notably, addition of a GnRH antagonist at 1100 h completely eradicated luteinizing hormone secretion at 1400 h and 1700 h of proestrus, and significantly reduced the Nr4a3 mRNA expression level at both the time points. These results suggest that GnRH is, at least partly, responsible for the increase in pituitary Nr4a3, and that the interaction between NR4A3 and ANXA5 is required to regulate Fshb expression during the preovulatory gonadotropin surge.

采用光度分析的工作曲线准确测定2-重氮-1-萘醌-5-磺酰氯的含量。于丙酮和无水乙醇的混合溶剂中，波长为380nm，该物质在4～30μLg/ml内与吸光度成直线关系。相关系数为0．9999，平均回收率100.24％，相对标准偏差为0.53%。     

In vitro rescue of immature avocado (Persea americana Mill.) embryos

An in vitro culture protocol was developed as a means of avocado embryo rescue. Different factors including presence of cotyledons, medium texture and cold or gibberellic acid pretreatments, were studied. To better understand the germination process in this recalcitrant species, immature zygotic embryos at different stages were used in these experiments. Optimum results were dependant on the embryo developmental stage. Whereas smaller embryos (5 mm long) germinated better in M1 liquid medium, 15 mm long embryos responded better when precultured in B5m medium supplemented with 1 mg l⁻¹ GA₃, and fully mature embryos were capable of germinating directly in solid M1 medium. Our results suggest the existence of two types of dormancy in avocado embryos: an embryo-dormancy caused by cotyledons, and another type of dormancy, mainly occurring in 35 mm long embryos and revealed by the formation of dwarfing rosette seedlings, that can be released by a GA₃ pretreatment.     

Effects of DARPP-32 on the drug sensitivity of SH-SY5Y cells

AIM: To investigate the effects of DARPP-32 on the drug sensitivity of human neuroblastoma cell line SH-SY5Y. METHODS: The plasmid containing cDNA of DARPP-32 gene and the small interfering RNA eukaryotic expression vector specific to human DARPP-32 gene were constructed and transfected into SH-SY5Y cells. Stable transfectants were identified by RT-PCR and Western blotting analysis. MTT assay and flow cytometry assay were used to investigate the effects of DARPP-32 on the drug sensitivity and the drug accumulation in cell models. The expressions of Bcl-2，Bax, P-gp and MRP were analyzed by RT-PCR and Western blotting. RESULTS: The stable clones with increased or decreased DARPP-32 expression were successfully established. Up-regulation of DARPP-32 significantly enhanced the drug sensitivity and the drug accumulation of SH-SY5Y cells due to down-regulation of P-gp and Bcl-2 expressions. Down-regulation of DARPP-32 significantly reduced the drug sensitivity of SH-SY5Y cells and decreased drug accumulation in SH-SY5Y cells. CONCLUSION: DARPP-32 might mediate the sensitivity of SH-SY5Y cells to conventional chemotherapeutic agents.

早杂5号大白菜是由两个自交不亲和系早5-4和011W-8配制而成的一代杂种.其中011W-8是利用游离小孢子培养技术,从庆农601中培育出的一个纯系;早5-4是从台湾引进的耐热2号中选出的优良单株,经多代自交选育出的自交不亲和系.该品种中桩类型、早熟耐热,叶淡绿色,叶面光滑无毛,叶柄宽,绿白色,生长势强,整齐度高,抗病毒病、霜霉病、软腐病.夏秋作小白菜栽培,播后30天收获,每667 m2产量2 000 kg,秋季作半结球栽培,生育期22天,667 m2产量2 200 kg.
