利用爱字棉1517×德州047重组近交系(recombinant inbred lines, RIL)中G6群体构建的SSR遗传连锁图谱及基于混合线性模型的复合区间作图法对QTL进行定位,并对主效QTL,加性×加性上位性QTL及与环境互作效应进行分析,为利用分子聚合方法提高产量提供理论依据。对2006年、2008年以及2009年的产量性状进行分离分析,检测到24个不同年份的主效QTL,其中相关于单株籽棉、单株皮棉、衣分、子指以及单株铃数的分别检测到1个不同年份稳定存在的主效QTL;对3年的产量性状作环境因子联合分析,检测到14个主效QTL,其中6个与环境互作,检测到20对加加上位性QTL,其中7对与环境互作。不同年份检测的稳定且受环境影响小或不受环境影响的与近处标记紧密连锁的主效QTL可用于分子标记辅助选择,以提高育种的效率。  相似文献

Fusarium head blight (FHB) is an important disease of wheat (Triticum aestivum L.). The aim of this study was to determine the effects of quantitative trait locus (QTL) regions for resistance to FHB and estimate their effects on reducing FHB damage to wheat in Hokkaido, northern Japan. We examined 233 F1-derived doubled-haploid (DH) lines from a cross between ‘Kukeiharu 14’ and ‘Sumai 3’ to determine their reaction to FHB during two seasons under field conditions. The DH lines were genotyped at five known FHB-resistance QTL regions (on chromosomes 3BS, 5AS, 6BS, 2DL and 4BS) by using SSR markers. ‘Sumai 3’ alleles at the QTLs at 3BS and 5AS effectively reduced FHB damage in the environment of Hokkaido, indicating that these QTLs will be useful for breeding spring wheat cultivars suitable for Hokkaido. Some of the QTL regions influenced agronomic traits: ‘Sumai 3’ alleles at the 4BS and 5AS QTLs significantly increased stem length and spike length, that at the 2DL QTL significantly decreased grain weight, and that at the 6BS QTL significantly delayed heading, indicating pleiotropic or linkage effects between these agronomic traits and FHB resistance.  相似文献

 选用第7、13、25号染色体上纤维品质相关QTL(Quantitative trait locus)密集分布区间的48对SSR(Simple sequence repeat)引物,对48份棉花种质进行多态性检测,研究结果显示在实验材料中3对SSR引物具有陆海差异多态性,相关分析表明3个优质基因SSR位点与纤维长度和纤维强度达到极显著相关。通过与海岛棉带型比对、追踪,从分子水平上明确了这些品种中来源于海岛棉渐渗于陆地棉的优异基因区段,为下一步分子标记辅助聚合育种提供了理论参考。  相似文献

High-density marker-based QTL mapping can serve as an effective strategy to identify novel genomic information to facilitate crop improvement. In this study, we genotyped an F2 population (KB12-1 × PP12-1) using a RAD-seq approach and constructed a high-density linkage map for radish. After a series of filtering procedures were performed, 17,124 SNPs and 3,336 indels with aa × bb genotyping were retained to obtain bin markers. Then, a linkage map comprising a total of 1,221 bin markers in nine linkage groups spanning 1,467.3 cM with an average marker interval of 1.2 cM was constructed. We evaluated the resistance of the F2 mapping population to black rot using F3 progeny, and two major QTLs related to black rot resistance were identified based on this map. Among these QTLs, qBRR2 on Chr.2 explained 26.97% of the phenotypic variation with a LOD score of 11.93, and qBRR7 on Chr.7 accounted for 27.06% of the phenotypic variation with a LOD score of 11.83. The additive effect of qBRR2 was positive (14.97); however, qBRR7 had the opposite effect (−11.99). The high-density linkage map and the major QTLs qBRR2 and qBRR7 provide new important information for disease resistance gene discovery and utilization in genetic improvement.  相似文献

在已构建的结球甘蓝AFLP、SSR和SRAP标记高密度遗传图谱的基础上,运用MapQTL 4.0软件对结球甘蓝F2群体抽薹、开花时间两性状分别进行QTL定位和分析。最终检测到2个控制甘蓝抽薹时间性状的QTL( qbt-3-2、qbt-9-1),以及一个同开花时间性状相关的QTL(qft-9-1),分别位于连锁群LG3与LG9上,这3个QTL均为增效位点,共解释甘蓝抽薹、开花时间性状变异的17.5%,22.7%;同时得到与QTLs共分离的2个分子标记E46 M52-5、me26-em13-1,均可作为辅助选育甘蓝耐抽薹品种的标记使用,这将为结球甘蓝耐抽薹品种的选育提供理论依据。  相似文献

Sub18是以陆地棉遗传标准系TM-1为背景, 含海岛棉3-79第18染色体的置换系材料。本研究以TM-1为受体亲本, 置换系Sub18为供体亲本, 借助分子标记辅助选择技术培育了一套以TM-1为背景, 含海岛棉3-79第18染色体不同长度片段的置换系。这套置换系由45个株系构成, 共78个置换片段。其中27个株系导入单片段, 占总株系的60%; 9个株系导入2个片段, 占20%; 9个株系导入3个及以上片段, 占20%。导入片段总长度为467.6 cM, 约为该染色体遗传长度的4倍, 每个株系内被替换的染色体片段长度不完全相同, 平均遗传长度为5.99 cM, 最短的为0.9 cM, 最长的20.35 cM。其中13个株系表现开放花蕾性状, 涉及的最短导入片段长5.05 cM。对TM-1、Sub18以及培育的45个导入系进行农艺性状调查和QTL联合定位分析, 鉴定出纤维强度(qFS-C18-1)、整齐度(qFU-C18-1)、马克隆值(qFMi-C18-1)、成熟度(qFMa-C18-1)、皮棉重(qLW-C18-1)、籽指 (qSI-C18-1)和衣分 (qLP-C18-1) 7个加性QTL和5个上位性效应QTL。研究结果为进一步精细定位目标QTL、克隆QTL以及重要性状分子设计育种奠定了基础。  相似文献

[Objective] The purpose of this study was to map quantitative trait loci (QTL) related to chlorophyll content based on Soil and Plant Analyzer Development (SPAD) readings in cotton. [Method] The 195 BILs (Backcross Inbred Lines) were produced by a cross between Gossypium barbadense Hai 7124 and G. hirsutum CRI 36, using CRI 36 as the recurrent parent for backcrossing with F1 to produce BC1F1, followed by seven generations of selfing. The genetic linkage map was constructed in a previous study. QTLs of chlorophyll SPAD value in the first flowering and boll development stages were identified with inclusive composite interval mapping (ICIM) method of the BIP and MET models in IciMapping 4.1 software, respectively. [Result] In total, nine chlorophyll SPAD reading QTLs were identified on 6 chromosomes. The q-SPAD-A11-1 detected at the first flowering stage overlapped with q-SPAD-A11-2 detected at the boll development stage, contributing 5.08% and 5.75% of the phenotypic variation, respectively. The q-SPAD-D08-2 physical position ranged from 48.71 to 53.65 Mb on chromosome D08, which overlapped with a chlorophyll content QTL detected in a previous study. [Conclusion] The novel stable QTLs, q-SPAD-A11-1 and q-SPAD-A11-2 detected in this study provide an important piece of information for fine mapping chlorophyll content in cotton.  相似文献

Making use of the markers linked closely to QTL for early-maturing traits for MAS (Marker-assisted selection) is an effective method for the simultaneous improvement of early maturity and other properties in cotton. In this study, two F2 populations and their F2:3 families were generated from the two upland cotton (Gossypium hirsutum L.) crosses, Baimian2 × TM-1 and Baimian2 × CIR12. QTL for early-maturing traits were analyzed using F2:3 families. A total of 54 QTL (31 suggestive and 23 significant) were detected. Fourteen significant QTL had the LOD scores not only > 3 but also exceeding permutation threshold. At least four common QTL, qBP-17 for bud period (BP), qGP-17a/qGP-17b (qGP-17) for growth period (GP), qYPBF-17a/qYPBF-17b (qYPBF-17) for yield percentage before frost (YPBF) and qHFFBN-17 for height of first fruiting branch node (HFFBN), were found in both populations. These common QTL should be reliable and could be used for MAS to facilitate early maturity. The common QTL, qBP-17, had a LOD score not only > 3 but also exceeding permutation threshold, explaining 12.6% of the phenotypic variation. This QTL should be considered preferentially in MAS. Early-maturing traits of cotton are primarily controlled by dominant and over-dominant effects.  相似文献
