【目的】白粉病(powdery mildew)是番茄生产上的重要病害,严重影响番茄的产量。番茄基因组测序工作的完成为抗病基因挖掘提供了重要的信息资源。ARP2/3(actin-related protein 2 and 3)复合体是肌动蛋白微丝骨架动力学的主要调控因子,能够参与包括响应外界胁迫等多种细胞学过程。本研究通过对番茄ARPC5(actin-related protein C5)进行克隆和抗病功能验证,为番茄基因组信息完善、抗病机制解析和分子育种等方面打下基础。【方法】从番茄LA1777(Solanum habrochaites)cDNA中PCR扩增ShARPC5,使用DNAMAN 6.0进行多序列比对;MEGA 6.0构建系统发育树;应用在线工具ProtComp v. 9.0进行亚细胞定位预测。利用实时荧光定量PCR(qRT-PCR)比较接种白粉菌(Oidium neolycopersici,On-Lz)后高感品种Moneymaker(MM)和高抗品种LA1777中番茄ARPC5的表达特征,分析白粉菌侵染与ARPC5表达的相关性。应用病毒诱导的基因沉默(virus-induc...  相似文献

Moromycin B (Mor B), saquayamycin B1 (Saq B1), saquayamycin B (Saq B), and landomycin N (Lan N), four angucyclines produced by the marine-derived actinomycete Streptomyces sp., are a class of polyketone compounds containing benzanthracene. Here, the structure–activity relationship of these four compounds was analyzed in human colorectal cancer (CRC) cells. Saq B1, which showed the strongest cytotoxicity with an IC50 of 0.18–0.84 µM for CRC cells in MTT assays, was employed to test underlying mechanisms of action in SW480 and SW620 cells (two invasive CRC cell lines). Our results showed that Saq B1 inhibited CRC cell proliferation in a dose- and time-dependent manner. Notably, lower cytotoxicity was measured in normal human hepatocyte cells (QSG-7701). Furthermore, we observed proapoptosis, antimigration, and anti-invasion activities of Saq B1 in CRC cells. At the same time, the protein and mRNA expression of important markers related to the epithelial–mesenchymal transition (EMT) and apoptosis changed, including N-cadherin, E-cadherin, and Bcl-2, in Saq B1-treated CRC cells. Surprisingly, the PI3K/AKT signaling pathway was shown to be involved in Saq B1-induced apoptosis, and in inhibiting invasion and migration. Computer docking models also suggested that Saq B1 might bind to PI3Kα. Collectively, these results indicate that Saq B1 effectively inhibited growth and decreased the motor ability of CRC cells by regulating the PI3K/AKT signaling pathway, which provides more possibilities for the development of drugs in the treatment of CRC.  相似文献

安塞试验站2005~2006连续两年采用N、P、K三因素最优设计,田试不同施肥量对南瓜Vc和干物质两项营养品质的影响,探讨南瓜优质的N、P、K肥效反应模式。结果表明,N肥对南瓜Vc含量的影响最大,K肥对干物质含量的影响最大,N与P交互作用和N与K交互作用对南瓜Vc、干物质含量影响显著。筛选出南瓜优质的较佳施肥量为:施氮90~95 kg/hm2,施磷75~80 kg/hm2,施钾25~30 kg/hm2。N∶P2O5∶K2O=1.00∶0.83∶0.28  相似文献

黑土坡耕地土壤流失形态分析——以一次降雨为例   总被引:7,自引:0,他引:7  
以东北黑土区典型漫岗坡耕地为研究对象,测定次降雨条件下整个集水盆地出口处的径流体积及其含沙量,研究侵蚀泥沙的粒径分布特征、碳氮富集度以及可溶性养分的流失状况。结果表明,研究区只有在强降雨的情况下才会在集水盆地尺度上发生水土流失现象。20.5 mm降雨条件下(2006年8月1日)径流泥沙含量为1.0~2.5 g/L,泥沙C、N富集系数分别为2.10和3.31。径流液中DOC的浓度介于2.47~3.93 mg/L之间,TN浓度介于1.61~3.28 mg/L之间,径流携带养分量在整个养分流失过程中是不可忽视的流失途径。侵蚀泥沙(未加分散剂)的团聚体组成和原土壤有很大差异,径流泥沙的流失主要是粒径为0.053 mm以下的团聚体,尤其是0.002~0.02 mm的粉粒团聚体为主。  相似文献

The cytotoxicity-bioassay-guided fractionation of the ethanol extract from the marine sponge Guitarra abbotti, whose 1-O-alkyl-sn-glycerol ethers (AGEs) have not been investigated so far, led to the isolation of a complex lipid fraction containing, along with previously known compounds, six new lipids of the AGE type. The composition of the AGE fraction as well as the structures of 6 new and 22 previously known compounds were established using 1H and 13C NMR, GC/MS, and chemical conversion methods. The new AGEs were identified as: 1-O-(Z-docos-15-enyl)-sn-glycerol (1), 1-O-(Z-docos-17-enyl)-sn-glycerol (2), 1-O-(Z-tricos-15-enyl)-sn-glycerol (3), 1-O-(Z-tricos-16-enyl)-sn-glycerol (4), 1-O-(Z-tricos-17-enyl)-sn-glycerol (5), and 1-O-(Z-tetracos-15-enyl)-sn-glycerol (6). The isolated AGEs show weak cytotoxic activity in THP-1, HL-60, HeLa, DLD-1, SNU C4, SK-MEL-28, and MDA-MB-231 human cancer cells. A further cytotoxicity analysis in JB6 P+ Cl41 cells bearing mutated MAP kinase genes revealed that ERK2 and JNK1 play a cytoprotective role in the cellular response to the AGE-induced cytotoxic effects.  相似文献

以从兰科植物出的内生真菌与大花蕙兰接种,均能形成菌根,其中GC941和GC945两种菌只侵染根的表皮细胞,在细胞内呈菌丝结结构,而GC934菌在侵染前期同前两和种真菌,后期菌丝可部分侵染皮层细胞,并呈疏松的分枝状结构。3种内生真菌可使幼苗茎叶干重比增施矿质营养但不接种真菌的处理(CK2)提高173.2%-250.1%,并对植株吸收N,P,K养分有促进作用,其中GC945菌使幼苗吸收N和K的量比CK  相似文献

采用二因素随机区组设计,利用尿素、过磷酸钙不同配比施用于3 a生宽叶羌活实生苗进行施肥试验.结果表明:单施尿素时施肥量120 kg/hm2是3a生宽叶羌活药材高产的最佳施用量,单施过磷酸钙时,施用量400 kg/hm2是宽叶羌活药材产量最佳使用量.随着尿素和过磷酸钙用量的增加、3 a生宽叶羌活药材的羌活根产量、挥发油含量、醇浸出物含量逐渐增加,在尿素施肥量120 kg/hm2、过磷酸钙施用量400 kg/hm2时3 a生宽叶羌活药材根产量、挥发油含量、醇浸出物含量均达到最大,之后逐渐降低.在青藏高原地区,3 a生羌活药材产量、挥发油含量、浸出物含量最高的肥料配比为:尿素施肥量120 kg/hm2、过磷酸钙用量400 kg/hm2.  相似文献

Lipid peroxidation is associated with the development of some pathologies, such as cardiovascular diseases. Reduction in oxidative stress by antioxidants, such as Arthrospira (formely Spirulina), helps improving this redox imbalance. The aim of the study was to evaluate the effect of the Arthrospira liquid extract “Spirulysat®” on oxidative markers—in particular, oxidized LDL (oxLDL)/total LDL cholesterol—and isoprostanes and to investigate its impact on lipid and glucose metabolism in the metabolic syndrome subject. A controlled, randomised, double-blind design was conducted in 40 subjects aged 18 to 65 years with metabolic syndrome after a daily intake of Spirulysat® or placebo for twelve weeks. Blood and urinary samples were collected at three visits (V1, V2, V3) in the two groups for parameters determination. Although the Spirulysat® group showed a decrease at all visits of the oxLDL/total cholesterol ratio, there was no significant difference compared to the placebo (p = 0.36). The urinary isoprostanes concentration in the Spirulysat® group was reduced (p = 0.014) at V3. Plasma triglycerides decreased at V3 (p = 0.003) and HDL-cholesterol increased (p = 0.031) at all visits with Spirulysat®. In conclusion, Spirulysat® did not change the oxidized LDL (oxLDL)/LDL ratio but decreased the urinary isoprostanes, plasma triglycerides and increased HDL cholesterol, suggesting a beneficial effect on metabolic syndrome.  相似文献

Four novel monocyclic cyclopropane acids, namely, sydocyclopropanes A–D (1–4), along with one known congener hamavellone B (5), were isolated from the Aspergillus sydowii MCCC 3A00324 fungus, which was isolated from the deep-sea sediment. The gross structures of novel compounds were established by detailed analyses of the spectroscopic data (HRESIMS and NMR spectra), and their absolute configurations were resolved on the basis of the quantum chemical calculations of ECD and NMR data, in association with DP4+ probability analyses. Sydocyclopropanes A–D, featuring the 1,1,2,3-tetrasubstituted cyclopropane nucleus with different lengthy alkyl side chains, were discovered in nature for the first time. All compounds exhibited antiviral activities against A/WSN/33 (H1N1), with IC50 values ranging from 26.7 to 77.2 μM, of which compound 1 exhibited a moderate inhibitory effect (IC50 = 26.7 μM).  相似文献

为了解江苏常州地区肉鸡禽白血病病毒(ALV)感染情况,本研究采集了该地区2019年7-12月、2020年1-4月肉鸡血清样本1257份,通过ELISA试剂盒检测ALV-A/B、ALV-J、抗原p27阳性率。结果显示:在接受调查的两个地区均存在ALV感染及混合感染,且ALV-A/B感染率(13.13%)均高于ALV-J(5.01%);不同月份阳性率存在一定差异,ALV-A/B、ALV-J均为春夏季阳性率较高,秋冬季逐渐下降趋势;抗体阳性样本中有54.39%的p27抗原阳性率。本次调查为江苏常州地区商品肉鸡ALV净化及防控提供一定的参考依据。  相似文献
