AIM: To investigate the damage in human umbilical vein endothelial cells (HUVECs) induced by recombinant soluble human CD40 ligand (rshCD40L). METHODS: The cultured HUVECs were treated with rshCD40L for 12 h. The survival activity of the HUVECs was observed by MTS assay. The expression of E-selectin, intercellular adhesion molecule (ICAM)-1, tissue factor (TF) and tissue factor pathway inhibitor (TFPI) was measured by ELISA. The activity of superoxide dismutase (SOD) and the level of malondialdehyde (MDA) were detected by the methods of thibabituric acid (TBA). RESULTS: Compared with normal group, different concentrations of rshCD40L (0.5, 1, 2, 3 mg/L) had no obvious effect on the survival activity of the HUVECs (P>0.05). rshCD40L at concentration of 0.5 mg/L promoted the secretion of E-selectin, sICAM-1, TF and TFPI in the HUVECs (P<0.01). rshCD40L at concentration of 0.5 mg/L also increased MDA content and reduced the activity of SOD in the HUVECs (P<0.05). CONCLUSION: 0.5~3mg/L rshCD40L has no obvious effect on endothelial cell survival, but already causes endothelial dysfunction by increasing endothelial inflammation and exogenous coagulation reaction, inducing lipid peroxides injury and reducing antioxidant capacity.  相似文献

为了弄清调控萝卜杂交种种子产量的影响因素,以夏抗40天萝卜为研究对象,研究不同纬度地区(武汉、阜阳、泰安及张掖)夏抗40天萝卜种株分枝数量、结荚数量及种子质量等,以期揭示纬度对萝卜杂交种种子产量的影响。试验结果表明,夏抗40天萝卜种株种子产量构成因素中,总结荚数、单荚粒数、千粒质量以甘肃张掖最高,制种总产量也是甘肃张掖地区的最高,达58.2kg/667m2,其次是山东泰安。  相似文献

The effect of sublethal concentrations, 0.00141% (LC20), 0.00251% (LC30), and 0.0036% (LC40) of a dispersible formulation of an acylurea insect growth regulator, flufenoxuron (Cascade) on certain biochemical parameters in the larvae of Tribolium castaneum was investigated. When neonates were fed on diet treated with sublethal concentrations for 24 h, it was observed that at all concentrations tested, there was a significant reduction in chitin content on the 15th day of development. Total soluble protein content at LC20 and LC30 decreased with increasing age of the larvae. At LC20 and LC40 concentrations there was a progressive increase in the protein: chitin ratio as a function of increase in age of the larvae. SDS-PAGE analysis of the larval tissue extracts indicated gross quantitative changes in some of the protein bands (MW 50-97 kDa). Western blot analysis revealed significant increase in the level of HSP70 in the extracts of larvae fed on LC30 treated diet, on the 7th and 10th day of development in the decreasing order. Conversely, a significant decrease in the hyper-phosphorylated form of p34cdc2 kinase due to flufenoxuron treatment indicating modulation of cell cycle regulation was observed. Thus, sublethal concentrations of flufenoxuron alter expression of developmentally regulated proteins, HSP70 and p34cdc2 and chitin formation in a stage-specific manner thereby resulting developmental abnormalities in T. castaneum.  相似文献

该研究获得了合浦珠母贝(Pinctada fucata)热休克蛋白22 (heat shock protein 22,PfHSP22)基因cDNA序列,全长2 187 bp,其中开放阅读框699 bp,预测编码232个氨基酸。结构预测显示PfHSP22具有小热休克蛋白家族典型结构域“α-晶体结构域”(ACD)。氨基酸序列比对结果显示,PfHSP22与缢蛏(Sinonovacula constricta)的HSP22相似性最高(54%)。进化分析显示,PfHSP22与软体动物的亲缘关系较近。实时荧光定量PCR结果分析表明,常温条件下,PfHSP22在类胡萝卜素含量高的橘色组和含量低的白色组的各组织中均有表达,足中表达量最高,且橘色组均显著高于白色组(P<0.05),表明类胡萝卜素含量高低可能影响PfHSP22的基础表达量。高温胁迫时,橘色组和白色组各组织的PfHSP22表达量在前3 h基本呈现下降趋势,之后呈现出或升高或下降的复杂变化;其中橘色组一些组织的PfHSP22表达量显著升高时间晚于白色组,这可能是由类胡萝卜素含量差异引起。  相似文献

Feline immunodeficiency virus (FIV) infection of the domestic cat induces an immunodeficiency characterised by a gradual depletion of CD4+ T-helper lymphocytes. The virus targets T-helper cells by way of an interaction between its envelope glycoprotein (Env) and the cell surface molecule CD134 (OX40), a member of the nerve growth factor receptor/tumour necrosis factor receptor superfamily. The Env–CD134 interaction is a necessary prerequisite for the subsequent interaction with CXCR4, the only chemokine receptor identified to date to act as a co-receptor for FIV. As T-helper cell expression of CD134 and CXCR4 is restricted to activated cells, FIV targets selectively antigen-specific T-helper cells. With disease progression the cell tropism of the virus expands; this may be the result of changes in the way in which Env interacts with CD134, a less stringent Env–CD134 interaction enabling the Env to interact more readily with CXCR4 and thus broadening the cell tropism of virus. In contrast, viruses that are present in early infection may have a narrower cell tropism, reflecting a more stringent interaction with CD134. Accordingly, “early” viruses may target CD134-expressing cells more efficiently and be more resistant to neutralising antibody. It is these early viruses that may be transmitted and should be considered as candidates for the development of vaccine regimes and novel therapeutic agents.  相似文献

本文主要研究初产和经产、不同比例的过瘤胃淀粉和脂肪日粮及环境温湿指数对热应激奶牛HSP70表达的影响。试验选用54头中国荷斯坦奶牛,根据产奶量、泌乳天数和胎次相近原则,进行随机区组试验设计。试验奶牛分3个处理组(高过瘤胃淀粉低饱和脂肪酸组PF161、中等过瘤胃淀粉中等饱和脂肪酸组PF131和低过瘤胃淀粉高饱和脂肪酸组PF101),每个处理组18头奶牛。分别于正试期的第1天、第21天、第49天、第77天和试验结束时采集血样,应用酶联免疫吸附试验法检测HSP70的含量。结果显示:在正试期间,牛舍环境温湿指数均值为79.67,奶牛处于中度热应激;初产奶牛血液中HSP70的含量高于经产奶牛;中等过瘤胃淀粉中等饱和脂肪酸组的HSP70含量高于其他两组;HSP70含量随THI变化呈现升高-降低-升高-降低的变化趋势。  相似文献

选取250只60日龄蛋鸡,随机分为5组:空白对照组、维生素C组、复方中药Ⅰ、Ⅱ、Ⅲ组,每天记录各组的耗料量、蛋重、产蛋量、死淘数以及软、破蛋数量,采用ELISA法测定蛋鸡心脏组织HSP70表达量。结果显示,复方中药能够全面改善热应激状态下蛋鸡的生产性能,其中复方Ⅱ的效果最佳;中药复方组蛋鸡心脏组织热休克蛋白-70(HSP70)表达量均低于空白对照组,其中复方Ⅱ组下降的幅度最大,表明各复方中药均能提高热应激蛋鸡的生产性能,其作用机制可能与调控HSP70表达量有关。试验探讨了中药复方对慢性热应激下蛋鸡生产性能和心脏组织HSP70表达量的影响,研究成果具有指导生产实际价值。  相似文献

通过研究枸杞多糖(LBP)对免疫抑制小鼠血清中细胞因子IL-6、IL-12/IL-23p40和IL-10分泌的影响,探讨LBP的免疫调节机制,为LBP的开发利用提供理论依据。60只ICR雌性小鼠,随机分为5组,对各组小鼠进行连续4d每日腹腔注射环磷酰胺(40mg/kg),间隔3d后,再连续4d每日腹腔注射环磷酰胺(40mg/kg);同时,用高(40mg/1kg)、中(20mg/1kg)、低剂量(10mg/1kg)的LBP给小鼠连续灌胃2周,阳性对照和阴性对照分别用LPS(1mg/kg)和PBS,连续2周。小鼠眼眶取血,分离血清,ELISA检测血清中IL-6、IL-12/IL-23p40和IL-10的分泌水平。结果显示,与阳性对照和阴性对照组相比,LBP能提高免疫抑制小鼠血清中IL-6、IL-12/IL-23p40和IL-10的分泌水平,提示LBP能增强小鼠细胞免疫与体液免疫应答,为枸杞多糖作为免疫增强剂的开发应用奠定了一定的理论基础。  相似文献

Cancer is one of the most common reasons for death in dogs. One promising approach is oncolytic virotherapy. We assessed the oncolytic effect of genetically modified vaccinia viruses in canine cancer cells, in freshly excised tumour biopsies, and in mice harbouring canine tumour xenografts. Tumour transduction efficacy was assessed using virus expressing luciferase or fluorescent marker genes and oncolysis was quantified by a colorimetric cell viability assay. Oncolytic efficacy in vivo was evaluated in a nude mouse xenograft model. Vaccinia virus was shown to infect most tested canine cancer cell lines and primary surgical tumour tissues. Virus infection significantly reduced tumour growth in the xenograft model. Oncolytic vaccinia virus has antitumour effects against canine cancer cells and experimental tumours and is able to replicate in freshly excised patient tumour tissue. Our results suggest that oncolytic vaccinia virus may offer an effective treatment option for otherwise incurable canine tumours.  相似文献
