MicroRNA-9-5p mediates sympathetic overactivity by targeting KCNN3 in paraventricular nucleus of rats with T2D

AIM To investigate whether microRNA-9-5p （miR-9-5p） mediates sympathetic overactivity by targeting KCNN3 （potassium intermediate/small conductance calcium-activated channel, subfamily N, member 3） gene,which encoded small-conductance calcium-activated potassium channel 3 （SK3） protein, in paraventricular nucleus （PVN） of rats with type 2 diabetes mellitus （T2D）. METHODS A rat model of T2D was established by high-fat diet combined with intraperitoneal injection of 30 mg/kg streptozotocin. The levels of miR-9-5p and KCNN3 mRNA in PVN were detected by real-time PCR. The relationship between KCNN3 and miR-9-5p was predicted by TargetScan. Recombinant adeno-associated virus （rAAV）-miR-9-5p or KCNN3 were bilaterally microinjected into the PVN to observe the changes in plasma glucose levels and sympathetic drive indicators. The number of FosB and SK3 positive cells was measured by immunofluorescence staining. The protein expression of SK3 was determined by Western blot. The relationship between KCNN3 and miR-9-5p were confirmed by cell transfection and dual-luciferase reporter assay. RESULTS Compared with the rats in diabetes control （DC） group, the blood glucose, sympathetic drive indexes and the level of miR-9-5p in PVN were significantly increased, while the SK3 expression in PVN was obviously reduced in the diabetes mellitus （DM） rats. After microinjecion of rAAV-miR-9-5p in PVN, the sympathetic drive indexes, blood glucose, and the number of FosB-positive cells were increased significantly, but the SK3 protein expression was significantly reduced （P<0.05）. However, up-regulation of KCNN3 in PVN had the opposite effect. These responses were obviously enhanced in DM rats compared with DC rats. The results of cell transfection and dual-luciferase reporter assay demonstrated that miR-9-5p bound to the 3’-UTR of KCNN3 and inhibit its expression. CONCLUSION miR-9-5p was up-regulated in PVN of the rats with T2D, and it may mediate sympathoexcitation by targeting KCNN3.

黑线仓鼠胴体重长指标的研究

黑线仓鼠（Ｃｒｉｃｅｔｕｌｕｓｂａｒａｂｅｎｓｉｓ）是华北平原最主要的农田害鼠之一，本文用胴体重代替体重采用数理统计方法，分析了其胴体重与体长之比（即胴体重长指标）的年龄、性别、季节、年度变化，结果除年度变化外，均发现有显著性差异，雄性的该值较雌性的大得多，同性相比，Ⅳ龄组最大，在１９８７１９８９年间的种群密度下，其胴体重长指标与种群密度间没有相关关系     

用葡萄球菌甲蛋白协同凝集鉴别猪痢疾密螺旋体的研究

应用猪痢疾蜜螺旋体（Ｔ．Ｈ．）免疫血清结合葡萄球菌甲蛋白（ＳＰＡ）与Ｈ１０，２－５，Ｘ２３，Ｃ１７等Ｔ．Ｈ．菌株进行协同凝集试验，均出现明显凝集反应。而免疫血清结合不含ＳＰＡ的菌株，无特定病原（ＳＰＦ）血清结合含ＳＰＡ的菌株均无凝集反应。免疫血清结合ＳＰＡ与７种肠道菌均无凝集反应。免疫血清用量以０．２ｍｌ为合适。最低用菌量为３０倍稀释，即每个视野Ｔ．Ｈ．约４０个菌体。

土壤通用浸提剂与有效养分测试研究进展

土壤有效养分分析是推荐施肥的关键环节。其分析方法已经形成较为成熟的体系,测试技术开始向联合、通用浸提剂和多元素同时测定方向发展。通过对几种通用浸提剂的分析比较,Mehlich3土壤浸提剂以其多元素联合浸提、通用性强以及与参比方法和作物吸收量具有较好的相关性成为适合我国土壤有效养分分析的通用浸提剂,其与等离子体发射光谱联合应用的M3-ICP技术在我国推荐施肥中具有广阔的前景。     

Hmgn3和Hoxa10对小鼠子宫基质细胞蜕膜化过程中Foxo1表达的调控

利用小鼠子宫基质细胞体外诱导蜕膜化模型转染Hmgn3或Hoxa10过表达载体及siRNA片段,通过荧光定量PCR方法检测Hmgn3和Hoxa10对Foxo1表达的调控。结果显示:转染Hmgn3或Hoxa10过表达载体可促进Foxo1在子宫基质细胞蜕膜化过程中的表达,而转染Hmgn3或Hoxa10siRNA则可抑制Foxo1的表达。在小鼠子宫基质细胞中转染Hmgn3或Hoxa10siRNA后再添加孕酮,Foxo1的表达显著下降。同样,干扰Hmgn3或Hoxa10也可减低cAMP对Foxo1表达的调控。结果表明:Hmgn3和Hoxa10可通过Foxo1来影响小鼠子宫基质细胞的蜕膜化,孕酮和cAMP可通过Hmgn3和Hoxa10来调控Foxo1在子宫基质细胞中的表达。     

动物鲜味受体的研究进展及其基因表达调控

动物的鲜味受体包括代谢型谷氨酸受体(m GluR)和味觉受体异源二聚体(T1R1/T1R3),是C型G蛋白偶联受体,N末端捕蝇草模块(VFT)区域可与鲜味配体结合,识别鲜味。本文主要论述了鲜味受体的研究进展、鲜味识别转导机制及鲜味受体基因的表达调控等,以期为相关研究提供参考。     

钙蛋白酶3(CAPN3)基因多态性与牛胴体性状的关联分析

为了探讨钙蛋白酶3(Calpain3,CAPN3)基因作为影响牛肉质性状候选基因的可能性,寻找与牛肉质性状相关的分子标记,对CAPN3基因的5′端区域进行SNPs检测,分析不同基因型在不同品种或者组合群体间的分布规律。利用测序和PCR-RFLP的方法进行SNPs检测和基因型分析,计算等位基因频率、各位点多态信息含量。结果发现,2546位处发生点突变C→T。各位点的3种基因型与牛生产性状的最小二乘分析结果表明,AB与BB基因型个体在胴体质量指标上存在显著差异(P0.05)。初步推断CAPN3基因可能是影响牛肉质性状潜在的主效基因或与主效基因连锁,并且这个位点具有成为分子标记的潜在可能。     

Analysis of NO3 interception of the parasitic angiosperm Orobanche spp. using a positron-emitting tracer imaging system and 13NO− 3: A new method for the visualization and quantitative analysis of the NO3 interception ratio

Abstract

The root parasitic plants Orobanche spp. (broomrapes) seriously affect agricultural production. A visualization and quantitative analytical method for the interception of nutrients was established using a positron-emitting tracer imaging system and ¹³NO^? ₃. By using this analytical method that involves volume normalization with ¹⁸F^? images, the nitrogen nutrient interception ratio of the Orobanche spp. was calculated to be 73.6 ± 3.9% in a host–parasite system of red clover (Trifolium pratense L.).
