Summary Starch-gel electrophoresis was used to study esterase, acid phosphatase, and peroxidase isozyme patterns in the primary leaves, stems, and roots of 13 species within the genus Phaseolus (P. acutifolius, P. adenanthus, P. angularis, P. atropurpureus, P. aureus, P. bracteatus, P. calcaratus, P. lathyroides, P. mungo, P. ricciardianus, domestic P. vulgaris, and wild P. coccineus and P. vulgaris). Most species showed unique banding patterns in each isozyme system. However, a close similarity in banding patterns was observed for the domestic P. vulgaris, wild P. vulgaris, and wild P. coccineus indicating close genetic relationships, possibly through species introgression or common descent.Great differences in isozyme patterns were found for different tissues from the same species in all systems indicating the high tissue specificity of isozymes. Upon compilation of bands from all the species, a very large number of isozymes were obtained in each system or tissue, making the electrophoretic isozyme technique of high potential in studies of isozyme variants in this genus.  相似文献

Summary Over a period of many years, data on dominance relationships of S-alleles in Brussels sprouts were collected at URL and IVT. The level of activity of S-alleles in heterozygotes was assessed on the basis of the number of pollen tubes that penetrated into the stigma. 209 out of 210 possible combinations between 21 S-alleles were used for this investigation. The S-alleles were grouped separately for activity in pollen and style on the basis of their sensitivity to lose activity in S-heterozygotes. Besides S-allele interaction per se, activity was found to be influenced by environment and genetic background.Results suggest that in stigma, co-dominance is the normal pattern and that deviations are caused by factors other than S-allele interaction as such.In pollen, only three truly recessive alleles were found. Besides several combinations with mutual weakening in pollen, examples of independent weakening were found.  相似文献

Summary Two cloned cassava genes (pCAS6, pHNL) and a barley gene (pBLT63) which is highly conserved in higher plants, were evaluated as potential multilocus probes for RFLP analysis of phylogenetic relationships of species within the genus Manihot. For M. rubricaulis, M. chlorosticta and M. esculenta subsp. flabellifolia dendrograms based on 13 probe-restriction enzyme combinations show very little variation between accessions of the same species. A close relationship is demonstrated between the Mexican species M. chlorosticta and the M. esculenta subsp. flabellifolia accessions, which does not support the classification of flabellifolia as a true South American wild species.  相似文献

Nineteen of the major strawberry (Fragaria × ananassa Duch.) cultivars grown in the UnitedStates and Canada were examined for AFLP markerpolymorphisms. For the AFLP reactions, the EcoRI-ACC primer was used in combination with fourMseI primers (MseI-CAC, MseI-CAG,MseI-CAT, or MseI-CTT). Each set ofprimers produced 46–66 scorable fragments ranging insize between 50 and 500 bp. The polymorphic fragmentsproduced from each set of primers were more thansufficient to distinguish among all the cultivars,demonstrating the usefulness of AFLP markers forcultivar identification. Similarity coefficients werecalculated based on data from 228 AFLP markers anddata from 15 previously characterized RAPD markers. The RAPD markers had been specifically selected forfingerprinting purposes because they succesfullydistinguish 41 strawberry cultivars, including the 19cultivars analyzed in this study. Separatedendrograms were constructed based on analysis of theAFLP and RAPD marker data using a neighbor-joiningalgorithm. The dendrograms were compared and found tobe very different. Correlations between similaritycoefficients calculated from AFLP marker data,similarity coefficients calculated from RAPD markerdata, and coefficients of coancestry calculated frompedigree information were evaluated. Interestingly,a better correlation with the coefficients ofcoancestry was observed with the RAPD marker data thanwith the AFLP marker data.  相似文献

Random amplified polymorphic DNA (RAPD) analysis was applied to eight commercial cultivars of pineapple, two intergroup hybrids and two wild species. Morphologically, pineapple is divided into the Cayenne, Queen, Spanish, Maipure and Abacaxi groups. Members of the first three groups have been analysed in this study. The cultivars ‘Tradsithong’, ‘Phuket’, ‘Sawee’ and ‘Tainan’, with spiny leaves, form the Queen group. In ‘Pattavia’, ‘Nanglae’ and ‘Petburi no. 2’ (Cayenne group), spines are confined to the leaf tips. ‘Intrachitdang’ is normally placed in the Spanish group, which is morphologically similar to the Queen group, but with inferior quality fruit. DNA amplification products were compared from 16 arbitrary 10‐mer primers from which a dendrogram was constructed. The results confirmed morphological classifications for seven of the eight commercial cultivars, with the Queen and Cayenne groups as separate clusters. However, the cv. ‘Intrachitdang’ was more closely related to the Cayenne group. Two hybrids from reciprocal Cayenne × Queen group crosses, were more closely allied to the Queen group. The two wild species were outside the groups. RAPD analysis can be exploited to investigate relationships within pineapple germplasm.  相似文献

In mammals, the specificity of FSH–FSH receptor (FSHR), LH–LH receptor (LHR) and TSH–TSH receptor couples is such that no cross-activation occurs under normal physiological conditions. The interactions between fish gonadotropins and their receptors, however, appear to be less discriminatory. For example, the catfish FSHR is highly responsive to both catfish LH and catfish FSH, while the catfish LHR is specific for its cognate LH. Comparative structure–function studies aimed at elucidating the molecular basis of ligand promiscuity (in fish) and ligand selectivity (in mammals) are described in this paper.  相似文献

Length–weight relationships were estimated for 21 coastal fish species of the Azores, namely Abudefduf luridus, Bothus podas, Chromis limbata, Coris julis, Diplodus sargus, Echiichthys vipera, Gaidropsarus guttatus, Labrus bergylta, Mullus surmuletus, Phycis phycis, Pomatomus saltator, Sarda sarda, Scorpaena maderensis, Scorpaena notata, Seriola rivoliana, Serranus atricauda, Sparisoma cretense, Sphyraena viridensis, Synodus saurus, Thalassoma pavo and Trachinotus ovatus. Significant length–weight relationships were found for all species. Sexual dimorphism did not affect the length–weight relationships, except in the cases of S. cretense and C. julis. Length–length equations for converting size measurements (standard length (SL) and fork length (FL) to total length (TL)) are also presented for all fish species.  相似文献

应用SSR对浙江省分布的部分柿属植物(包括柿、浙江柿和金枣柿在内的3种46个基因型)进行了种质鉴定和亲缘关系分析。从已公开序列的20对柿属植物SSR引物中筛选出扩增谱带清晰和多态性丰富的引物11对;应用建立的技术体系,可将同物异名和同名异物的试材得以系统整理,一般认为是同一品种的兰溪大红柿与永康方柿,玉环长柿与永嘉长柿指纹图谱各有不同,应为不同种质。聚类分析结果表明,金枣柿(Diospyros sp.)与其他供试试材间的亲缘关系较远,支持其可能为一新种的观点;浙江柿(D.glaucifolia Metc.)与金枣柿的亲缘关系较远但与柿(D.kaki Thunb.)较近;部分供试日本柿种质与浙江省柿种质的亲缘关系较近。研究结果可为浙江省柿属植物种质资源保护和利用提供一定的科学依据。  相似文献

The potential use of RAPD markers for taxonomic studies in Malus was investigated using 18 accessions of wild species and 27 apple cultivars. 29 preselected random decamer primers were applied to three sets of Malus genotypes. Random amplified polymorphic DNA (RAPD) ‘fingerprints’ were analysed for polymorphic amplification fragments, and coefficients estimating genetic similarity were calculated on the basis of about 50 polymorphic RAPD loci in each set of genotypes. Cluster analysis by an unweighted pair-group method with arithmetic averages (UPGMA) revealed that, in the cultivars, the molecular classification was in good agreement with the known lineage. A dendrogram generated for the wild species gave relationships that were, in principle, in accordance with the known phylogenetic information. Closely related species from section I were clearly distinguishable from those of sections III and IV. On the molecular level, a high degree of genetic diversity was found among both different apple cultivars and wild species of the genus Malus. The results gave additional evidence for the hypothesis that M. pumila and M. sylvestris were involved in the origin of the cultivated apples.  相似文献

Randomly amplified polymorphic DNA (RAPD), inter-simple sequence repeat (ISSR) and a semi-random PCR system were used to analyze the genetic diversity of 16 Italian common bean landraces and their relationship to four commercial cultivars. Of the primers tested, 8 ISSR, 6 RAPD and 7 semi-random primers produced polymorphic and reproducible DNA fragments. A higher proportion of polymorphic bands were observed using ISSR (85%) and semi-random (90%) primers than RAPD (69%) method. The combination of any two semi-random markers allowed the identification of all 20 bean genotypes. In contrast ISSR (except for primer (CAC)3GC) and RAPD markers appeared to be less informative as more than two markers were necessary to achieve the same diagnostic level. Moreover, 7 ISSR, 2 RAPD and 8 semi-random exclusive bands were identified as putative population-specific markers. Semi-random and ISSR derived dendrograms showed similar tendencies in terms of genetic relatedness, whereas clustering of genotypes within groups was not similar when compared with the RAPD technique. Despite the different ability to resolve genetic variation among the investigated landraces, two major clusters with less than 60% (ISSR) and 40% (RAPD and semi-random) genetic similarity were formed with all three marker systems. The two groups were correlated with the phaseolin patterns and seed size of the landraces. The analysis showed that the cultivar ȁ8Lingua di Fuocoȁ9 and most of the landraces (13 out of 16) collected in Italy belong to the Andean gene pool, whereas only the three populations from Pratomagno belong to the Middle American gene pool.  相似文献
