对一年生草本植物小麦、水稻、玉米等从生物膜膜脂的组成和配比的角度来探讨抗寒机理已有很多的报告,对多年生木本植物的研究还不很多。以前的研究结果大部分都支持了脂肪酸不饱和度与抗寒性的相关性,但各种植物也不尽相同,甚至有一些相反的结论。为了比较杨树耐寒品种和不耐寒品种在化学和生理特性上的差别,本研究用现代仪器分析方法测定了耐寒欧美杨(45杨)和不耐寒美洲黑杨(69杨)的膜脂总脂肪酸、磷脂组分的配比和总磷脂含量,为判别杨树无性系新品种的抗寒能力提供了依据。  相似文献

Rheum palmatum Linn has been widely applied in the clinical treatment of diabetes mellitus. It has been found that emodin as the major bioactive component of R. palmatum L exhibits the competency to activate peroxisomal proliferator-activated receptor-γ (PPARγ) in vitro. So the aim of this study was to evaluate the anti-diabetic effects of emodin through the activation of PPARγ on high-fat diet-fed and low dose of streptozotocin (STZ)-induced diabetic mice. The diabetic mice were intraperitoneally injected with emodin for three weeks. No changes of food consumption and the body weight in emodin-treated mice were monitored daily during the entire experiment. At the end of experiment, the levels of blood glucose, triglyceride and total cholesterol in serum were significantly decreased after emodin treatment. However, serum high-density lipoprotein cholesterol (HDLc) concentration was significantly elevated. The glucose tolerance and insulin sensitivity in emodin-treated group were significantly improved. Furthermore, the results of quantitative RT-PCR analysis showed that emodin significantly elevated the mRNA expression level of PPARγ and regulated the mRNA expressions of LPL, FAT/CD36, resistin and FABPs (ap2) in liver and adipocyte tissues. No effects on the mRNA expressions of PPARα and PPARα-target genes were observed. Taken together, the results suggested that the activation of PPARγ and the modulation of metabolism-related genes were likely involved in the anti-diabetic effects of emodin.  相似文献

通过氢氧化钾‐甲醇溶液对桐油中的脂肪酸进行酯交换反应甲酯化,采用GC‐M S联用技术对脂肪酸甲酯进行了分析鉴定,鉴定了桐油中α‐桐酸、β‐桐酸、油酸、亚油酸、硬脂酸、棕榈酸等13种桐油脂肪酸组分。通过比较各产地桐油中的脂肪酸成分含量,结果表明,油中桐酸含量具有较大的差异,桐酸含量介于70.03%~82.00%之间;不同产地桐油中α‐桐酸、β‐桐酸、油酸、亚油酸等单个组分的脂肪酸也存在一定的差异,说明不同桐油中脂肪酸含量具有一定的产地特征。  相似文献

秦巴山区核桃种质的形态特征和生化特性   总被引:1,自引:0,他引:1  
本文以秦巴山区33份优良单株为材料,分析比较了核桃的形态特征、核桃种仁的物理特性和生化特性。结果表明:秦巴山区核桃坚果重量在10.48 g至12.72 g之间,最重的区域是广元市朝天区;核桃核仁重量在4.94 g至6.62 g之间,最重的区域是南江县;核桃仁坚比率在0.45至0.54之间,比率最大的区域是南江县;核桃壳厚在1.11 mm至2.00 mm之间,最薄的区域是广元市中区;核桃坚果平均直径在31.99 mm至33.70 mm之间,最长的区域是通江县;果面都较光滑,风味以香甜为主,核桃取仁都容易,大部分核桃的内褶壁退化,果形以近圆形、椭圆形和圆球形为主。南江县、青川县和万源县核桃的仁色浅,而朝天区、广元市中区和通江县核桃的仁色较深。秦巴山区各个区域核桃的丙氨酸、缬氨酸、异亮氨酸、亮氨酸、苯丙氨酸、蛋氨酸、脯氨酸、丝氨酸、甘氨酸、苏氨酸、酪氨酸、胱氨酸、精氨酸、天门冬氨酸、赖氨酸、组氨酸和氨基酸总量差异显著(P<0.05),而谷氨酸差异不显著(P>0.05)。说明各种氨基酸的形成除与外部环境条件密切相关外,还与核桃种质自身特性有关。秦巴山区各个区域核桃的棕榈油酸、油酸、亚油酸和花生四烯酸含量差异显著(P<0.05),棕榈酸、硬脂酸、亚麻酸和花生酸含量差异不显著(P>0.05)。秦巴山区不同区域核桃的粗脂肪含量、饱和脂肪酸、不饱和脂肪酸含量、多不饱和脂肪酸和单不饱和脂肪酸含量差异显著(P<0.05)。综上所述,秦巴山区核桃的种质资源非常丰富,不同区域核桃种质的形态特征和生化特性差异都较大,可针对不同类型核桃深加工产品的需要,选用不同区域的核桃,同时也为进一步筛选优良核桃种质和特异种质提供了可能性。  相似文献

测定2341份我国东北地区大豆品种资源5种脂肪酸含量。以亚油酸含量最高,45—62%。平均含量依次为亚油酸>油酸>棕榈酸>亚麻酸>硬脂酸。分析比较结果,各脂肪酸含量,不同种皮色、脐色、结荚习性、叶形和花色品种间存在显著差异。棕榈酸、油酸、亚油酸和亚麻酸含量,不同栽培类型、不同粒形品种间有显著差异。硬脂酸、油酸和亚麻酸  相似文献

为了了解fad基因在胡麻蒴果发育过程中对不饱和脂肪酸的调控,对高、中、低三个不同亚麻酸(C18:3)含量的胡麻品种(’CDC Gold’,‘内亚7号’,’Linola’)进行了不同时期的品质测定,以及脂肪酸去饱和酶2a基因(fad2a)、脂肪酸去饱和酶2b基因(fad2b)、脂肪酸去饱和酶2c基因(fad2c)、脂肪酸去饱和酶3a基因(fad3a)、脂肪酸去饱和酶3b基因(fad3b)的qRT-PCR定量分析。结果表明,随着蒴果成熟,可溶性糖含量呈降低趋势,粗脂肪与粗蛋白不断积累,且差异显著(p<0.05)。fad2a基因、fad3a基因以及fad3b基因在各个时期中的表达符合正态分布。以0 d的蒴果为对照,在胡麻种子形成过程中,‘内亚7号’的三个基因在5 d和15 d的表达量迅速增加,到30 d时急剧减少,15 d的fad2a基因表达量为5.23倍,fad3a基因表达量是fad2a基因表达量的14.52倍,fad3b基因的表达量是fad2a基因表达量的16.14倍,表明这三个基因参与不饱和脂肪酸积累过程。在低亚麻酸含量品种‘Linola’30 d中,fad3a基因是fad2a基因表达量的52.71倍,fad3b呈下调趋势;在高亚麻酸含量品种‘CDCGold’30d中,fad3a基因与fad2a基因的表达量均呈下调趋势,fad3b的表达量为3.92倍;在中等亚麻酸含量品种‘内亚7号’中,fad2a基因表达量降低了0.31倍,fad3a基因是fad3b基因表达量的1.87倍。fad2b基因、fad2c基因熔解曲线不稳定,峰值低,可以在后续试验中继续探索。  相似文献

 This review discusses the analysis of whole-community phospholipid fatty acid (PLFA) profiles and the composition of lipopolysaccharides in order to assess the microbial biomass and the community structure in soils. For the determination of soil microbial biomass a good correlation was obtained between the total amount of PLFAs and the microbial biomass measured with methods commonly used for determinations such as total adenylate content and substrate-induced respiration. Generally, after the application of multivariate statistical analyses, whole-community fatty acid profiles indicate which communities are similar or different. However, in most cases, the organisms accounting for similarity or difference cannot be determined, and therefore artefacts could not be excluded. The fatty acids used to determine the biomass vary from those which determine the community structure. Specific attention has to be paid when choosing extraction methods in order to avoid the liberation of fatty acids from non-living organic material and deposits, and to exclude the non-target selection of lipids from living organisms, as well. By excluding the fatty acids which were presumed to be common and widespread prior to multivariate statistical analysis, estimates were improved considerably. Results from principal component analysis showed that determining the levels of fatty acids present in both low and high concentrations is essential in order to correctly identify microorganisms and accurately classify them into taxonomically defined groups. The PLFA technique has been used to elucidate different strategies employed by microorganisms to adapt to changed environmental conditions under wide ranges of soil types, management practices, climatic origins and different perturbations. It has been proposed that the classification of PLFAs into a number of chemically different subgroups should simplify the evaluating procedure and improve the assessment of soil microbial communities, since then only the subgroups assumed to be involved in key processes would be investigated. This study investigated the effect of management on -glucosidase, -glucosaminidase, alkaline phosphatase, and arylsulfatase activities and the microbial community structure in semiarid soils from West Texas, USA. Surface samples (0–5 cm) were taken from a fine sandy loam, sandy clay loam, and loam that were under continuous cotton ( Gossypium hirsutum L.) or in cotton rotated with peanut ( Arachis hypogaea L.), sorghum ( Sorghum bicolor L.), rye ( Secale cereale) or wheat ( Triticum aestivum L.), and had different water management (irrigated or dryland), and tillage (conservation or conventional). The enzyme activities were higher in the loam and sandy clay loam than in the fine sandy loam. Soil pH was not affected by management, but the soil organic C and total N contents were generally affected by the different crop rotations and tillage practices studied. The trends of the enzyme activities as affected by management depended on the soil, but in general crop rotations and conservation tillage increased the enzyme activities in comparison to continuous cotton and conventional tillage. The soil enzyme activities were significantly correlated with the soil organic C ( r -values up to 0.90, P< 0.001), and were correlated among each other ( r -values up to 0.90, P <0.001). There were differences in the fatty acid methyl ester profiles between the fine sandy loam and the sandy clay loam and loam, and they reflected the differences in the enzyme activities found among the soils. For example, a 15:0 ranged from 1.61±0.25% in cotton-peanut/irrigated/no-till in the fine sandy loam to 3.86±0.48% in cotton-sorghum/dryland/conservation tillage in the sandy clay loam. 高精料饲喂导致奶牛乳脂率降低的相关机制目前还不清楚.本研究通过探究高精料饲喂对肝脏内乳脂前体代谢的影响,揭示奶牛(Bos taurus)发生乳脂抑制的相关机制,将10头泌乳期(25+5)d荷斯坦奶牛随机分为2组,分别饲喂精粗比4∶6(对照组)和6∶4(实验组)日粮.饲喂20周后实施肝脏血管瘘手术,护理4周,进行如下实验:1)统计奶牛产奶量,每周测定乳成分;2)采集空腹进出肝脏血液,检测甘油三酯(triglyceride,TAG)和游离脂肪酸(free fatty acid,FFA)的含量;3)活体穿刺取肝脏组织,qRT-PCR检测肝脏中脂代谢相关基因的表达.结果表明,高精料饲喂6~l8周可提高产奶量,18周后乳脂率显著降低(P<0.05).高精料组奶牛肝脏TAG和FFA的净输出含量显著(P<0.05)或极显著降低(P<0.01);肝脏中参与脂肪酸合成代谢的固醇调节元件结合蛋白-lc基因(sterol regulating element binding protein-1 c,SREBP-1c)、乙酰辅酶A羧化酶基因(acetyl-coa carboxylase,ACC)和脂肪酸合成酶基因(fatty acid synthetase,FAS)表达显著(P<0.05)或极显著(P<0.01)下调;参与脂肪酸分解代谢的过氧化物酶体增殖物激活受体α基因(peroxisome proliferators-activated receptor α,PPARα)和肉毒碱棕榈酰转移酶-1基因(carnitine palmitoyltransterase-1,CPT-1)表达显著上调(P<0.05).高精料饲喂导致奶牛肝脏脂肪酸合成代谢减弱而分解代谢增强,肝脏内乳脂前体物发生重分配,乳脂前体物输出减少,影响了乳脂合成.本研究揭示了高精料饲喂导致奶牛乳脂率降低的机制,为改善奶牛乳脂率提供了理论基础.  相似文献

Stable isotope analysis has been used as a powerful tool in food web studies in terrestrial ecosystems. In addition the occurrence and abundance of fatty acids may serve as indicator for feeding strategies of soil animals. Here we combine both approaches and investigate the fatty acid composition, δ13C values of bulk tissues and individual fatty acids in soil organisms. The fungi Chaetomium globosum and Cladosporium cladosporioides were isotopically labelled by fructose derived from either C3 or C4 plants, and the fungal-feeding nematode Aphelenchoides sp. was reared on C. globosum. Fungi and nematodes were used as diet for the Collembolan Protaphorura fimata. The sugar source was fractionated differently by fungal lipid metabolism in a species-specific manner that points to a sensitivity of physiological processing to the non-random distribution of 13C/12C isotopes in the molecule. As a general trend stearic acid (18:0) was depleted in 13C compared to the precursor palmitic acid (16:0), whereas its desaturation to oleic acid (18:1 ω9) favoured the 13C-rich substrate.Fatty acid profiles of P. fimata varied due to food source, indicating incorporation of dietary fatty acids into Collembolan tissue. Individuals feeding on fungi had lower amounts in C20 fatty acids, with monoenoic C20 forms not present. This pattern likely separates primary consumers (fungivores) from predators (nematode feeders). The isotopic discrimination in 13C for bulk Collembola ranged between −2.6 and 1.4‰ and was dependent on fungal species and C3/C4 system, suggesting differences at metabolic branch points and/or isotope discrimination of enzymes. Comparison of δ13C values in individual fatty acids between consumer and diet generally showed depletion (i.e. de novo synthesis) or no changes (i.e. dietary routing), but the fractionation was not uniform and affected by the type of ingested food. Fatty acid carbon isotopes were more variable than those of bulk tissues, likely due to both the distrimination by enzymes and the different lipid origin (i.e. neutral or polar fraction).  相似文献
