拟南芥Rad23原核重组蛋白构建和活性检测

在拟南芥中,Rad23基因通过参与泛素/26S蛋白酶体途径调节植物生长发育和激素应答.为进一步研究该基因编码蛋白的分子作用机制及其生物学功能,构建了pCold-Rad23融合蛋白重组表达载体,将该载体转化到大肠杆菌BL21中诱导表达目的蛋白.SDS聚丙烯酰胺凝胶电泳检测结果表明,分子质量为94 ku左右的融合重组蛋白在大肠杆菌中获得高效表达.采用纯化的重组蛋白多次免疫昆明小白鼠的方法,获得了特异性强并且效价高的多克隆抗体,经蛋白印迹检测,在分子质量40 ku左右处出现特异的蛋白质条带,证明所制备的抗血清可以与拟南芥Rad23蛋白特异性结合.     

Characterization of Zm26Sub5 and Its Potential Roles in Regulating Pollen Fertility of S-CMS in Maize

Mitochondrial orf355-orf77 and the nuclear fertility restorer locus Rf3/rf3 mutually control the fertility of male gametes inCMS-S in maize (Zea maya L.). A fragment of gene Zm26Sub5 was identified through cDNA-AFLP from a set of Rf3/rf3near-isogenie lines in a previous study. In the present study, real-time PCR analysis revealed that Zm26Sub5 expression levels were much higher in pollen of S-MoI7Rf3Rf3 than in pollen of S-Mo 17rf3rf3, and also higher than in fresh leaves, mature leaves, and roots of both S-Mo 17Rf3Rf3 and S-Mo 17rf3rf3. In silico cloning of full-length cDNA was successfully implemented.Gene Zm26Sub5 was 1451 bp in size, of which 1329 bp encoded a protein with 443 amino acids. The structure of this genewas analyzed by comparing its full length cDNA to homologous genomie DNA sequence (GenBank accession: Ac209463.3).Subsequent sequence analysis led to sub cellular localization of protein ZM26SUB5, and construction of a phylogenetic tree.In silico mapping indicated that Zm26Sub5 was located on chromosome 5 and closed to a reported starch-filled pollen ratio QTL. ZM26SUBS, therefore might have potential roles in repressing mitochondrial PCD which is associated with sterile activity in pollen in S-type cytoplasm.     

基于绿色发展理念的浙江“26县”政策对城乡收入差距的影响

为探求缩小城乡收入差距的新动能与新路径,本文关注并分析了基于绿色发展理念的浙江“26县”政策,介绍了“26县”政策出台的现实背景,总结了“26县”政策的主要内容,阐述了“26县”政策影响城乡收入差距的作用机理,并利用2013—2019年浙江省83个县级行政区的面板数据,通过双重差分模型实证检验了“26县”政策对城乡收入差距的影响效果。研究发现：“26县”政策显著缩小了城乡收入差距;同时,“26县”政策显著缩小了经营净收入和转移净收入的城乡差距,但在缩小工资性收入和财产净收入城乡差距上的作用不显著。上述发现既可为迈向共同富裕的绿色发展之路提供现实依据,也可为激发乡村生态要素优势的政策制定提供有益参考。     

水产养殖用复合微生物粉剂中菌种的分子生物学鉴定

对两种市售水产养殖用复合微生物粉剂中微生物菌种进行鉴定。利用平板分离技术对粉剂中的微生物进行分离纯化,进行了形态学观察、16S rDNA和26S rDNA序列分析,并构建系统发育树。结果表明,两种微生物粉剂中菌群结构存在一定差异,S1粉剂中鉴定出5株芽孢杆菌和2株酵母菌,分别为枯草芽孢杆菌、地衣芽孢杆菌或索诺拉沙漠芽孢杆菌、葡萄牙棒孢酵母和库德里阿兹威毕赤酵母;S2粉剂中鉴定出3株芽孢杆菌、1株放线菌、2株酵母菌和3株肠球菌,分别为解木糖赖氨酸芽孢杆菌、环状芽孢杆菌、地衣芽孢杆菌或索诺拉沙漠芽孢杆菌、芬氏纤维微菌、酿酒酵母、库德里阿兹威毕赤酵母、鹑鸡肠球菌、屎肠球菌或乳酸肠球菌。明确水产养殖用复合微生物粉剂的菌群结构为同类产品的安全性及有效性评估提供理论基础。     

对虾白斑综合征病毒VP24和VP26原核表达及抗体制备

滨海开发带土壤重金属分布特征及来源分析

以江苏省大丰市为例,研究了土壤中8种重金属的空间分布特征及其与土地利用的关系,并通过主成分分析方法,对其可能的来源进行了探讨。结果表明:研究区8种土壤重金属Cu、Zn、Pb、Cd、Cr、As、Hg、Ni平均含量分别为17.40、74.38、18.14、0.105、55.58、8.33、0.074、25.73mg/kg,不同采样点之间变异不大。沿垂直海岸线方向,随距海岸线距离增加,Cu、Zn、Pb、Cr、Hg、Ni 6种重金属含量逐渐升高,As含量逐渐降低,Cd含量则先升高、后降低。Cu、Zn、Pb、Cr、Hg、Ni 6种重金属均在水田土壤中含量最高,Cd在旱地土壤中含量最高,As则在滩涂土壤中含量最高。相关分析表明,土壤As含量与其他重金属元素含量的相关性均不显著,土壤Cd含量与Zn、Pb、Hg、Cr含量的相关性显著,与Cu、As、Ni含量的相关性不显著,其他各元素间相关性均达极显著水平。基于主成分分析结果,认为研究区土壤Cu、Zn、Pb、Cr、Hg、Ni 6种重金属元素含量受土壤母质影响较大,Cd含量与农业生产中磷肥施用关系密切,As含量的累积受磷肥施用的影响,但以水稻种植为主的耕作土壤As含量总体上呈下降趋势。本研究可为滨海开发带土地利用规划提供指导。     

Enhancement and confirmation of white spot syndrome virus detection using monoclonal antibody specific to VP26

A portion of the VP26 gene (VP26F109) encoding a structural protein of white spot syndrome virus was expressed, purified by SDS‐PAGE and used for immunization of Swiss mice for monoclonal antibody (MAb) production. Three groups of MAbs specific to different epitopes on VP26 were selected; these MAbs can be used to detect natural WSSV infection in Penaeus vannamei using dot blotting, Western blotting or immunohistochemistry without cross‐reaction with other shrimp tissues or other common shrimp viruses. The detection sensitivity of the MAbs was ranged 7–14 fmole per spot of the rVP26F109 as determined using dot blotting. A combination of three MAbs specific to VP26 with MAbs specific to VP28, VP19 and ICP11 increased the detection sensitivity of WSSV during early infection. Therefore, the MAbs specific to VP26 could be used to confirm and to enhance the detection sensitivity for WSSV infection in shrimp with various types of antibody‐based assays.     

122份小麦品种(系)抗叶锈病基因Lr26、Lr34、Lr38分子标记检测

运用抗叶锈病基因Lr 26、Lr 34、Lr 38的特异性分子标记,对122份小麦品种(系)进行了检测,以明确各品种(系)的抗叶锈性.结果表明:122份供试材料中,含有Lr 26的有33个,含有Lr 34的有3个,含有Lr 38的有37个;同时含有Lr 26和Lr 38的有30个,同时含有Lr 26、Lr 34和Lr 38的有3个.本研究结果可为小麦的抗叶锈育种提供理论参考.     

The effects of sulfometuron on the root growth of loblolly pine

The effect of sulfometuron (Oust) on the root growth of loblolly pine (Pinus taeda L.) seedlings was studied in the field and in two soil types in the greenhouse. In the greenhouse study sulfometuron was applied to the foliage only, the soil only, and to both foliage and soil at the rates of 0.10, 0.21, and 0.42 kg ai/ha. Twenty-eight days after application root growth significantly decreased for all methods of application and with increasing rates e.g., 0.10, 0.21, and 0.42 kg ai/ha sulfometuron reduced new root length by 42%, 53%, and 64%, respectively. Application in the field at the rate of 0.30 kg ai/ha resulted in a 68% reduction in root growth. However, by the end of the growing season treated seedling root and shoot biomass were 115% and 64% greater, respectively. To ensure earlier season survival the rate of sulfometuron applied should be kept at a minimum level that will allow both weed control and adequate root growth. Low rates would be particularly important during a dry planting season.     

高抗SMV抗旱大豆新品种晋豆26号的选育与利用

晋豆26号大豆新品种是山西农业大学大豆研究室于1988年以复61为母本、晋大28（晋豆16号）为父本进行有性杂交,采用系谱法选育而成的。该品种的突出特点是高抗SMV、抗旱性强、高产稳产、综合性状优良,具有4500kg/hm2的产量潜力。
