利用本实验室构建的C57BL/6背景下的PRNP基因敲除小鼠模型与C57BL/6野生型小鼠,通过避暗试验以及Morris水迷宫试验观察小鼠的行为。以此来研究PrPC蛋白的缺失对于小鼠学习与记忆能力影响,进而反映PrPC蛋白对于神经系统功能的影响。结果显示,PRNP敲除小鼠在避暗试验中对于电刺激的记忆比野生型小鼠强,且在第3天差异显著(P〈0.05)。在Morris水迷宫定位航行试验中,敲除鼠的逃避潜伏期明显长于野生小鼠,第3、5、6天差异显著(P〈0.05)。空间探索试验中,PRNP敲除鼠与野生小鼠相比较,首次穿越虚拟平台的时间以及穿越平台的次数差异并不显著。研究表明,PrPC蛋白缺失对于小鼠主动回避伤害的记忆能力有所提高,但是对于空间立体学习和记忆能力损害很大。 相似文献

Object recognition memory and contextual fear conditioning task performance in adult C57BL/6 mice exposed to cranial fast neutron irradiation (0.8 Gy) were examined to evaluate hippocampus-related behavioral dysfunction following acute exposure to relatively low doses of fast neutrons. In addition, hippocampal neurogenesis changes in adult murine brain after cranial irradiation were analyzed using the neurogenesis immunohistochemical markers Ki-67 and doublecortin (DCX). In the object recognition memory test and contextual fear conditioning, mice trained 1 and 7 days after irradiation displayed significant memory deficits compared to the sham-irradiated controls. The number of Ki-67- and DCX-positive cells decreased significantly 24 h post-irradiation. These results indicate that acute exposure of the adult mouse brain to a relatively low dose of fast neutrons interrupts hippocampal functions, including learning and memory, possibly by inhibiting neurogenesis. 相似文献

The aim of the study was to build a comprehensive picture of the appearance in the blood stream of Ag-specific plasma cells and memory B cells in the bovine model. For this purpose, we have developed a method allowing the detection and quantification of both cell types within individual calves immunised with ovalbumin. During the primary response, we detected a burst of ovalbumin-specific plasma cells at days 6 and 7 post-immunisation, which was followed by the production of specific Ab, whereas a gradual increase of memory B cells was only detected from day 15. As expected, a boost immunisation performed 7 weeks later induced a quicker and stronger secondary response. Indeed, a burst of plasma cells was detected in the blood at days 3 and 4, which was followed by a strong increase in Ab titres. Furthermore, a burst of memory B cells, and not a gradual increase, was detected at days 5 and 6 post-boost immunisation. Importantly, we showed a strong correlation between the anti-ovalbumin-specific IgG titres detected 5 months after secondary immunisation and the plasma cell numbers detected in the blood at the peak response after secondary immunisation. The detection and quantification of plasma cells following an mmunisation/vaccination strategy could constitute a very effective means for predicting the magnitude and longevity of an Ab response. 相似文献

Despite geotextiles having potential for soil conservation, limited scientific data are available to assess the effects of geotextiles in reducing runoff and water erosion. Hence, the objective of this review is to analyse the effects of plot length (L) and other possible affecting factors [cover percentage (C, %), slope gradient (S), rainfall duration (D), rainfall intensity (I), sand, silt and clay contents, soil organic matter (SOM) content and geotextile type (natural or synthetic)] on the effectiveness of geotextiles in reducing soil and water loss, based on reported experimental data. From linear regressions, C (%) and soil sand, silt and clay contents are found to be the most important variables in reducing SLR (ratio of soil loss in bare plots to that in geotextile treated plots) for splash, C (%) for interrill and D (min) for rill and interrill erosion processes, respectively. Soil clay and silt contents and D are key variables in decreasing RR (ratio of runoff from bare plots to that from geotextile treated plots) for interrill, and clay content for rill and interrill erosion processes, respectively. The linear relationship between mean b-value (geotextile effectiveness factor in reducing soil loss) and L of all studies was not significant (P > 0.05). The same is true for the relationship between L and SLR, and L and RR. However, when L is added to an equation as an interaction term with C (%), geotextile cover is significantly (P < 0.05) more effective in reducing SLR on shorter plots than longer ones for both interrill and rill and interrill erosion processes. Buffer strip plots (area coverage ∼ 10%) with Borassus and Buriti mats have the highest b-values. 相似文献

将葡萄糖、氯化钠、氯化钾、柠檬酸钠四种物质分别配制成三个渗透压水平(278mOsm、328mOsm、378mOsm)的12种单方精液稀释液,在低温(1~4℃)下进行鸡精液稀释保存试验,在四种物质中各选出一种好的稀释液,再将所选四种单方稀释液按不同比例制成复方精液稀释液,再次进行保存试验。所有稀释液均测定渗透压、pH值、缓冲指数、精子存活时间、精子生存指数等指标。结果表明:①柠檬酸钠、葡萄糖对鸡精液的保存具有重要作用,而氯化钾在鸡精液稀释液中并不是必需的。②复方稀释液保存效果普遍优于单方稀释液。③低温下鸡精液稀释液渗透压应为等渗或接近等渗,且最适渗透压还与构成稀释液的物质种类和相互配比有关系。④稀释液pH值应接近中性,最好与精液pH值保持一致。⑤在缓冲指数为3.894—7.257的范围内,缓冲指数与精子存活时间和生存指数呈现显著的负相关。 相似文献

Estimating the ‘critical’ distance at which adjacent land-use degrades wetland water and sediment quality 总被引:1,自引:0,他引:1
Conversion of forested lands to agriculture or urban/residential areas has been associated with declines in stream and lake water quality. Less attention has been paid to the effects of adjacent land-uses on wetland sediment and water quality and, perhaps more importantly, the spatial scales at which these effects occur. Here we address these issues by examining variation in water and sediment nutrient levels in 73 southeastern Ontario, Canada, wetlands. We modeled the relationship between water and sediment nutrient concentrations and various measures of adjacent land-use such as forest cover and road density, measured over increasing distances from the wetland edge. We found that water nitrogen and phosphorous levels were negatively correlated with forest cover at 2250 meters from the wetland edge, while sediment phosphorous levels were negatively correlated with wetland size and forest cover at 4000 meters and positively correlated with the proportion of land within 4000 meters that is itself wetland. These results suggest that the effects of adjacent land-use on wetland sediment and water quality can extend over comparatively large distances. As such, effective wetland conservation will not be achieved merely through the creation of narrow buffer zones between wetlands and more intensive land-uses. Rather, sustaining high wetland water quality will require maintaining a heterogeneous regional landscape containing relatively large areas of natural forest and wetlands. 相似文献

AIM:To investigate the expression of synaptophysin in the CA1 region of hippocampus and prefrontal cortex (PFC) of rats with posttraumatic stress disorder (PTSD), and to explore the mechanism of spatial memory changes in PTSD rats.METHODS:Healthy adult SD rats (n=36) were randomly divided into 2 groups:control group and model group, with 18 rats in each group. The rats in model group was continuously given single prolonged stress (SPS) to construct the PTSD model. Morris water maze (MWM) was used to test the learning and memory ability of the rats in the 2 groups. The protein expression of synaptophysin in the hippocampal CA1 area and PFC was examined by immunohistochemistry, Western blot and immunofluorescence experiments. RESULTS:The latency of the rats in searching for the underwater platform in model group was significantly longer than that in control group from the 2nd day (P<0.01) in the MWM experiment, the target quadrant swimming time was significantly shortened (P<0.01), and the times of crossing the platform were also significantly reduced (P<0.01). The results of immunohistochemistry, Western blot and immunofluorescence experiments showed that the expression of synaptophysin was obviously reduced in the CA1 region of hippocampus and PFC in model group as compared with control group (P<0.05 or P<0.01).CONCLUSION:The reduction of spatial memory ability in PTSD rats may be associated with the decreased expression of synaptophysin in the CA1 region of hippocampus and PFC. 相似文献
