空间诱变水稻新品种华航一号和新品系华航八号与各自的原品种相比,灌浆期剑叶都表现出ATP酶活性较高;细胞质果糖-1,6-二磷酸酯酶(FBP酶)活性较低;白天蔗糖积累较少,淀粉积累较多;夜间光合产物输出的总量较多.水稻千粒质量和单穗质量与剑叶上午淀粉积累呈正相关,与剑叶夜间光合产物总量下降呈正相关,与剑叶上午蔗糖积累呈负相关.  相似文献

选用高温耐性品种农大228、082和高温敏感品种茉莉占、协青早B,比较研究了高温胁迫对根系质膜ATPase活性和NH4^+吸收速率的影响。结果表明,高温胁迫下,农大228和082根系质膜H^+-ATPase和Ca^2+-ATPase活性均比对照提高,而茉莉占和协青早B分别比对照大幅度下降,品种间差异显著(Duncan’s法检验)。根系质膜蛋白含量在高温胁迫后均表现下降。082的NH4^+吸收速率不受高温影响,热敏感型品种NH4^+吸收速率受高温胁迫明显下降。  相似文献

 铝胁迫下 ,耐铝性不同的两个小麦品种 ( Altas66,耐铝 ;Scout66,铝敏感 )根细胞质膜 H+ - ATP酶或 Ca2 + -ATP酶活性变化趋势相似。 2 0 μmol/ L 和 10 0 μmol/ L 铝处理 5 d,质膜 Ca2 + - ATP酶活性明显下降 ;H+ - ATP酶活性在 2 0 μmol/ L 铝处理时上升 ,10 0 μmol/ L 铝时下降。质膜磷脂含量下降 ,糖脂含量上升 ,但铝敏感的 Scout66磷脂 /糖脂比值下降较耐铝的 Altas66显著。 2 0和 10 0 μm ol/ L 铝处理后 ,Scout66的亚麻酸含量和脂肪酸不饱和指数明显下降 ,Altas66的脂肪酸不饱和指数则稍有上升  相似文献

Branchial plasma membranes from the freshwater cichlid teleostOreochromis mossambicus (tilapia) contain two Na+-dependent ATPases: Na+/K+ ATPase, and an amiloride-sensitive ATPase which is postulated to operate as a Na+/H+ (–NH4 +) ATPase. It is suggested that both enzyme activities are located in the basolateral membrane system of the chloride cells. K+ has opposing effects on the two enzymes: it stimulates Na+/K+ ATPase and inhibits Na+/H+ (–NH4 +) ATPase activity. Na+/H+ ATPase appears more sensitive to NH4 + at low concentrations than Na+/K+ ATPase and the stimulatory effect by NH4 + ions on the first enzyme could be important in facilitating NH4 + excretion by tilapia gills under physiological conditions.In vitro maximum stimulation by NH4 + is similar for the two enzymes (200%). In contrast to Na+/K+ ATPase, Na+/H+ ATPase activity is inhibited by supra-physiological (>20 mM) concentrations of NH4 +.  相似文献

ABSTRACT:   Enzymatic and structural properties of white croaker fast skeletal muscle myosin were determined and compared with those of walleye pollack counterpart. Ca2+-ATPase activity of white croaker myosin was decreased to approximately 70% of the original activity during 1 day of storage at 0°C and pH 7.0 in 0.5 M KCl and 0.1 mM dithiothreitol, whereas that of walleye pollack was decreased to approximately 20% under the same condition. The activation energy ( E a) for inactivation of white croaker myosin calculated by the Arrhenius plot for inactivation rate constant (KD) was 1.2-fold higher than that of walleye pollack. While Ca2+-ATPase showed a similar KCl-dependency for the two species, the maximal activity was observed at pH 6.2 and 6.3 for white croaker and walleye pollack, respectively. Actin-activated myosin Mg2+-ATPase activity of white croaker was approximately half that of walleye pollack at 0.05 M KCl and pH 7.0, although the two myosins showed a similar affinity to F-actin with K m of 1.7 and 1.4, respectively. Limited proteolysis with α-chymotrypsin cleaved heat-denatured white croaker myosin mainly at heavy meromyosin/light meromyosin (HMM/LMM) junction, whereas walleye pollack myosin was cleaved at several sites in LMM as well as at the HMM/LMM junction.  相似文献

ABSTRACT:   Myosins were prepared from fast skeletal muscles of grass carp thermally acclimated to 10, 20 and 30°C in the laboratory as well as from those seasonally acclimatized and collected in January (winter) 2003 and May (spring), August (summer) and November (autumn) 2002. The maximal initial velocities ( V max) of actin-activated Mg2+-ATPase activity for myosins from the 10°C-acclimated and winter grass carp were 1.7–1.8-fold as high as those from the 30°C-acclimated and summer fish. The inactivation rate constant ( K D) of Ca2+-ATPase for myosin from the 10°C-acclimated grass carp was three to fourfold higher than those for myosins from the fish acclimated to 20°C and 30°C, whereas myosin from winter grass carp was about sevenfold as high as that for myosin from summer fish. Myosins from spring and autumn fish showed K D values comparable to those of the fish acclimated to 30°C and 10°C, respectively. In differential scanning calorimetry analysis, the transition temperature ( T m) was observed near 38°C and 45–46°C with most myosins. However, the lowest T m at 32–33°C was given as one of the major endotherms in myosins from the 10°C-acclimated, autumn and winter fish. These responses of grass carp to changed environmental temperatures were almost similar to those for common carp reported previously.  相似文献

Testosterone (T) administration to maleAnabas testudineus significantly stimulated the activities of cytochrome oxidase, -glycerophosphate dehydrogenase (-GPDH) and Mg2+ adenosine triphosphatase (Mg2+ ATPase) and inhibited lactate dehydrogenase (LDH), cytosolic and mitochondrial malate dehydrogenases (MDH). The activities of succinate dehydrogenase (SDH), glucose-6-phosphate dehydrogenase (G-6-PDH) and catalase were unaffected by testosterone treatment. Administration of estradiol-17 (E2) in female fish, significantly stimulated cytochrome oxidase activity, inhibited Mg2+ ATPase, SDH, catalase and cytosolic and mitochondrial MDH activity, and was without effect on other enzymes studied.The simultaneous injections of actinomycin D or chloramphenicol and T or E2 prevented the hormonal influence on hepatic enzyme activities. The present study demonstrates that inA. testudineus sex steroids influence hepatic oxidative metabolism by a mechanism sensitive to the action of inhibitors of protein synthesis.  相似文献

Flavonoids (morin, quercetin and phloroglucinol) were tested for their ability to modulate the function of P-glycoprotein ATPase of the insecticide resistant pest Helicoverpa armigera (Ha-Pgp). Flavonoids in the presence of ethylparaoxon or cypermethrin significantly reduced both larval weight as well as survival rate 40-50%. Morin and quercetin inhibited the activity of Ha-Pgp ATPase by 80-90%, whereas phloroglucinol inhibited ATPase activity by 40% at 100 μM concentration. These flavonoids inhibited the verapamil, ethylparaoxon and cypermethrin-stimulated Ha-Pgp ATPase activity. Morin, quercetin and phloroglucinol binding were quantitated by quenching of the intrinsic Trp fluorescence of purified Ha-Pgp ATPase. Drug transport was monitored in proteoliposomes containing Ha-Pgp ATPase using the high affinity fluorescent substrate tetramethylrosamine (TMR) in real time. Addition of the morin and quercetin mediated the collapse of the TMR concentration gradient generated by Ha-Pgp ATPase. The inhibition studies on Ha-Pgp ATPase activity may contribute towards understanding new strategies of the pest to overcome insecticide resistance.  相似文献

通过水培的方法研究了可降解螯合剂EDDS对印度芥菜对Cu积累的影响,分析了根系中与Cu主动排出有关的P型ATPase的活性及其基因表达。结果表明:根系中Cu的积累量随外界浓度的升高而升高,但Cu在茎杆与叶片中的积累量则变化不大。随着外加EDDS浓度的增加,根、茎、叶中的Cu的积累量均出现下降的趋势。上述结果表明,根系以Cu离子为主要的吸收形式,而不是EDDS-Cu。通过对印度芥菜根系细胞膜上P型ATPase活性及基因表达分析后发现,不加EDDS时,随着外界Cu浓度的升高,ATPase活性增强,而当Cu达到16μmol L-1时出现降低的趋势。但是,ATPase活性随着外加EDDS的浓度增加而降低,这是因为EDDS降低了外界的Cu活度。RT-PCR分析结果表明,该ATPase的基因(BjHMA)在转录水平上的表达与活性变化一致。由于P型ATPase的作用是将Cu排出细胞,因此上述结果说明,其活性大小以及转录水平的变化受到外界Cu离子活度的影响,在一定的范围内可以调节植物对Cu的积累。  相似文献
