对虾白斑综合征病毒vp28基因的表达及其与血细胞的结合

根据GenBank上WSSV囊膜蛋白基因vp28的序列,设计合成引物,PCR扩增得到vp28基因,成功构建重组表达载体pBAD/gIIIA-VP28并转化大肠杆菌E.coli.用L-阿拉伯糖在37℃诱导重组基因工程菌,表达产物经SDS-PAGE和Western-blot检测,显示有与预期大小31kDa相符合的目的蛋白.荧光显微镜方法分析显示,表达的VP28可与克氏原螯虾血细胞结合.结果表明,在合适的培养条件下,构建的重组表达载体pBAD/gIIIA-VP28不仅能够表达vp28 基因,而且表达的VP28具有很高的抗原结合活性.     

Prevalence,geographic distribution and phenotypic differences of Piscirickettsia salmonis EM‐90‐like isolates

Early reports accounted for two main genotypes of Piscirickettsia salmonis, a fish pathogen and causative agent of piscirickettsiosis, placing the single isolate EM‐90 apart from the prototypic LF‐89 and related isolates. In this study, we provide evidence that, contrary to what has been supposed, the EM‐90‐like isolates are highly prevalent and disseminated across Chilean marine farms. Molecular analysis of 507 P. salmonis field isolates derived from main rearing areas, diverse hosts and collected over 6 years, revealed that nearly 50% of the entire collection were indeed typed as EM‐90‐like. Interestingly, these isolates showed a marked host preference, being recovered exclusively from Atlantic salmon (Salmo salar) samples. Although both strains produce undistinguishable pathological outcomes, differences regarding growth kinetics and susceptibility to the antibiotics and bactericidal action of serum could be identified. In sum, our results allow to conclude that the EM‐90‐like isolates represent an epidemiologically relevant group in the current situation of piscirickettsiosis. Based on the consistency between genotype and phenotype exhibited by this strain, we point out the need for genotypic studies that may be as important for the Chilean salmon industry as the continuous surveillance of antimicrobial susceptibility patterns.     

爱媛28’香橙砧与枳砧在南平市引种表现与评价

2017年引进早熟杂柑品种‘爱媛28’香橙砧和枳砧苗分别在大棚设施和露天进行试种,栽培4年调查比较两个砧木组合的生长表现、丰产性与果实品质,结果表明：‘爱媛28’香橙砧树表现生长势强,冠幅、树高、茎粗、结果枝、秋梢等生长量指标显著大于枳砧树,3a树初产1436 kg/667㎡,4a树2523kg/667㎡,显著高于枳砧4a树初产1809kg/667㎡;大棚种植香橙砧4a树果实10月下旬TSS11-12%,12月中旬TSS可达15%,与枳砧相当,可滴定酸比枳砧略高,两者固酸比均达18以上,口感风味极佳,两者单果重、果形指数、果皮厚度、可食率无显著性差异;未密封大棚4a树体经受2020年寒冬极端低温考验无明显冻害,抗旱抗寒能力中等,易感黑点病和流胶病,大棚种植能很好预防黑点病。综合评价爱媛28枳砧和香橙砧均适合在南平市种植,香橙砧比枳砧早期丰产性稳产性好,收益更高,香橙砧‘爱媛28’设施种植较枳砧更具优势。     

Effect of root applied 28-homobrassinolide on the performance of Lycopersicon esculentum

The roots of 20 days old seedlings of tomato (Lycopersicon esculentum Mill.) at the time of their transplantation, were dipped in 10⁻⁸, 10⁻⁷, 10⁻⁶ or 10⁻⁵ M of 28-homobrassinolide (HBR) for 15, 30 or 45 min and were allowed to grow in earthen pots, in a net house. The leaves of the plants, at days 30 and 60, possessed elevated quantities of nitrate reductase (NR), carbonic anhydrase (CA) and the contents of chlorophyll. The values for all the above characteristics were significantly higher than that of the water-fed control. The fruits borned at the treated plants were more in number and possessed a lower quantity of ascorbic acid than the control. Moreover, the fruits at ripening, had higher levels of lycopene and β-carotene. Among the treatments, 15 min feeding of 10⁻⁸ M HBR proved best.     

孕酮处理对蛋鸡蛋壳钙化过程中CaBP-d28k基因表达的影响

实验旨在研究孕酮对蛋鸡CaBP-d28k基因表达的影响。对产蛋高峰期蛋鸡进行孕酮和孕酮受体拮抗剂(RU-486)处理,利用实时荧光定量PCR技术(QRT-PCR),测定在蛋壳形成过程中输卵管子宫部钙结合蛋白(CaBP-d28k)、孕酮受体(PGR)的表达量;同时对血液中钙离子和孕酮浓度变化进行测定。结果表明:孕酮处理后,血液中钙离子浓度显著降低(P<0.05),输卵管子宫部CaBP-d28k表达量也显著降低(P<0.05);孕酮受体拮抗剂处理后,血液中钙离子浓度显著升高(P<0.05),输卵管子宫部CaBP-d28k表达量极显著升高(P<0.01)。结果提示,在蛋壳钙化过程中,孕酮对CaBP-d28k表达和钙离子转运起负调控作用。     

松花菜新品种鹿松90天的选育

鹿松90天是温州市神鹿种业有限公司以自交不亲和系12号为母本、自交系BH81为父本配制而成的松花型花椰菜一代杂种,从定植到采收约90 d,每667 m2产量2 800 kg左右。在浙江省温州、杭州、金华、温岭等地试种表现高产稳产、优质、抗逆性强,同时,北京、湖北、河南、四川、甘肃等省市试种结果表明,其适合在东南沿海和黄河流域作中熟栽培。     

农业设施测控系统的研制

该文简要介绍了在农业设施中以新型单片机AT89S8252为核心的测控仪表的组成特点及其硬件与软件的优化设计方法。重点介绍了在农业设施群中，通过多机通信技术，用一台PC机集中数据采集、管理、控制各个设施中的测控仪表的主从多机工作模式，以及在这种工作模式中，单片机与PC机的串行通信接口硬件设计、通信软件设计、PC机的界面设计特点及设计方法。这些设计使得该系统在农业设施中推广成为可能。     

Berberine attenuates lipopolysaccharide-induced impairments of intestinal glutamine transport and glutaminase activity in rat

Berberine was reported to protect against the intestinal injury and improve the survival rate in sepsis, and glutamine deficiency was considered to be correlated with mortality in sepsis. We found that berberine pretreatment ameliorated lipopolysaccharide-induced direct intestinal injury and mucosal hypoplasia and attenuated impairments of intestinal glutamine transport and glutaminase activity, B⁰AT1 mRNA and protein expressions, and glutaminase protein expression. These findings showed the first time that berberine pretreatment could improve intestinal recovery and attenuate the impairment of glutamine transport and glutaminase activity in rat sepsis. This might be one of the mechanisms for the beneficial effect of berberine on sepsis.     

新型肝癌介入治疗药物90Y-P204-Lipiodol的制备及其稳定性研究

目的:研究了新型肝癌介入治疗药物90Y-P204-Lipiodol的制备方法及其稳定性,旨在为肝癌的介入性内照射放疗提供满意的放射性药物。方法:采用传统化学工艺中比较成熟的萃取技术与核医学中的放射性核素标记方法相结合,以碘油(Lipiodol)作为萃取过程中的有机相,同时碘油也是被标记核素的载体,选择萃取剂P204作为碘油标记的偶联剂,利用萃取剂的萃取性能,在一定的pH、温度、搅拌时间和搅拌频率等条件下,对钇-90(90Y)进行萃取标记,测定萃取率,然后按不同的相比加入到生理盐水、新生小牛血清以及人的血液中,于液闪仪上测定被标记90Y的损失率。结果:0.01 mol/L的P204碘化油溶液萃取率高达99.9%,符合放射性核素标记的要求。本萃取法碘油标记后形成的络合物90Y-P204-Lipiodol并不改变碘化油的结构,而且具有很好的稳定性。结论:借助现代医学影像技术尤其是实时超声显像引导精确定位技术,有望通过瘤区注射方法使90Y-P204-Lipiodol稳定积聚在肝肿瘤组织内,从而实现真正意义上的肝癌内照射放疗,未来的研究将促进其在肝癌介入性内照射放疗中的早日应用。
