用淀粉凝胶电泳及琼脂覆盖技术对山丹马的血液红细胞6磷酸葡萄糖脱氢酶和葡萄糖磷酸异构酶的电泳变异进行了测定。在6磷酸葡萄糖脱氢酶座位发现了3种表现型,即FF,FD和FS,其频率分别为0.958,0.021和0.021。在葡萄糖磷酸异构酶座位发现了两种表现型,即II和FI,其频率分别为0.771和0.229。等位基因频率直接通过表型计算出:PGDF为0.979,PGDD为0.0104,PGDS为0.0104;GPIF为0.1146,GPII为0.8854。亲子关系排除概率在6PGD和GPI座位点分别为0.1009和0.0912。两座位的个体识别概率分别为0.081和0.353。 相似文献

该研究以中国石竹2个品种的雄性不育植株带芽茎段为初始外植体进行组织培养,筛选了适合其芽诱导、继代增殖的培养基配方,并对试管苗移栽介质进行了挑选,成功地建立了2个中国石竹雄性不育植株的离体繁殖与保存体系。结果表明:中国石竹芽的最佳诱导培养基为MS+6-BA 1.0 mg/L+NAA 0.3 mg/L,最佳继代增殖培养基为MS+6-BA 0.3 mg/L+NAA 0.1 mg/L,最适宜的试管苗移栽基质是珍珠岩。同时,2个不育材料组织培养繁殖后代仍表现为雄性不育,确保了原材料的雄性不育性状的稳定性。 相似文献

The performance of I and II meiotic triploids and control oysters (Crassostrea madrasensis) reared at Tuticorin Bay was compared to determine if the improvements in the growth of edible oysters were additive to faster growth in triploids. After a grow‐out period of 12 months, both mean whole weights and shell heights were in order I meiotic triploid>II meiotic triploid>control. Mean whole weights and shell height of different oyster lines were all significantly different (P<0.05). On an average, larger morphological traits indicated that growth improvements from triploids were additive, and throughout the study triploid oysters maintained faster growth rate than their diploid siblings. Condition index and adductor muscle diameter of both triploids were higher than those of control. 相似文献

中华鲟是处在硬骨鱼类与软骨鱼类之间的中国古老的珍稀鱼类之一,但在人工繁殖和养殖过程中,容易受到不同疾病的危害,因此,需要深入研究其免疫调控,为其疾病预防提供理论依据。单个免疫球蛋白白细胞介素-1受体相关分子(SIGIRR)和肿瘤坏死因子受体相关因子6(TRAF6)是toll样受体(TLR)信号通路的2个重要的信号转导元件。本研究鉴定了中华鲟的SIGIRR和TRAF6的同源物,分别命名为ASDIGIRR (Acipenser sinensis DIGIRR)和ASTRAF6 (A. sinensis TRAF6),并研究它们在正常组织中的表达和Poly(I:C)诱导后的表达模式。结果显示,ASDIGIRR和ASTRAF6在健康鲟的10个组织中均有表达,且分别在肠道和头肾组织中表达量最高;用Poly (I:C)孵育中华鲟脾脏细胞后,ASDIGIRR在3 h时的表达量显著下调,在6 h时恢复至正常水平,后在24 h显著上调达到最高值,随后开始下调,至48 h时仍显著高于对照组,而ASTRAF6在6 h达到最高表达量,维持到24 h后再下调,48 h时下降至最低水平。研究表明,ASDIGIRR和ASTRAF6在中华鲟抵御病原入侵的免疫防御过程中起着重要作用。 相似文献

AIM To explore the expression and mechanisms of circular RNA hsa_circ_087631 in the patients with primary biliary cholangitis (PBC). METHODS RT-qPCR was used to detect the expression of hsa_circ_087631 in PBC patients and healthy controls. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was constructed and transfected into human acute T cell leukemia Jurkat cells, and then the expression levels of hsa_circ_087631, Bcl-6 mRNA and interleukin-21 (IL-21) mRNA were detected by RT-qPCR. Dual-luciferase reporter assay was performed to identify the interactions between hsa_circ_087631 and hsa-miR-346. RESULTS The expression of hsa_circ_087631 in the PBC patients was significantly increased compared with the healthy subjects. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was successfully constructed. The expression of hsa-miR-346 was significantly increased after the hsa-miR-346-overexpressing lentiviral vector was transfected into the Jurkat cells, while the expression levels of hsa_circ_0087631, Bcl-6 mRNA and IL-21 mRNA were significantly decreased. After wild-type or mutant hsa_circ_087631 vector and hsa-miR-346 mimics were transfected into 293T cells, the results of dual-luciferase reporter assay showed that hsa-miR-346 significantly decreased the luciferase activity of wild-type hsa_circ_087631 (P <0.01), but the regulation did not change significantly after mutation of the predicted binding site. CONCLUSION Peripheral blood hsa_circ_087631 level is elevated in the PBC patients. The hsa_circ_087631/hsa-miR-346/Bcl-6 signaling may take effect in human T cells. Hsa-miR-346 significantly reduces the expression of hsa_circ_087631, but it may not be regulated by predicted binding sites. 相似文献

Optimal conditions of 6‐dimethylaminopurine (6‐DMAP) for triploidy induction in the blacklip abalone Haliotis rubra (Leach, 1814) were investigated, targeting inhibition of second polar body (PB2) formation. Two experiments were conducted at a water temperature of 17.5–18.5°C where factorial combination of (1) four dosages (DSs) of 100, 150, 200 and 250 μM 6‐DMAP, four starting times (STs) of 15, 20, 25 and 30 min post fertilization, and two treatment durations (TDs) of 20 and 30 min and (2) three DSs of 50, 100 and 150 μM 6‐DMAP, three STs of 15, 20 and 25 min post fertilization, and three TDs of 10, 20 and 30 min, were applied respectively. Day 3 larvae were sampled for triploidy and survival. Percent triploidy was verified using flow cytometry (FCM). Results show that optimal inductions that combine both high rates of triploidy and reasonable survival were those treatments commenced 15 or 20 min post fertilization and continued for 20 or 30 min, using 100 μM 6‐DMAP. These conditions induced rates of triploidy and relative survival of 80.5–93.3% and 36.5–40.2%, respectively, in the first experiment, and corresponding rates were 79.1–93.6% and 20.7–43.0% in the second experiment. High percent triploidy were also obtained in a number of treatments using 150 μM 6‐DMAP, but with overall survival rates generally lower than those using 100 μM 6‐DMAP. 相似文献

外施10-9mol/L6-苄基腺嘌呤(6-BA)可促进大麦种子的出苗并缓解盐胁迫对大麦生长的抑制。在200mmol/LNaCl胁迫下,耐盐性较强的鉴4根尖内源异戊烯基腺苷组(iPAs)含量较高,维持高含量的时间较长;而科品7号根尖iPAs含量则迅速下降。外源6-BA使两者iPAs含量增高。盐胁迫下鉴4叶片中腐胺(Put)含量比较稳定,而科品7号叶片中Put含量却明显上升。外源6-BA、ABA和Ca(NO3)2使科品7号中Put含量明显下降,使鉴4中精胺(Spm)含量上升。 相似文献

本文研究了间苯二酚法对琼脂及琼脂糖样品中的3,6-内醚半乳糖的测定方法。实验中,根据间苯二酚试剂与3,6-内醚半乳糖和果糖的浓度在0.25μmol/mL以下时,具有近乎相同的标准比色曲线的原理,分别对测定波长、显色温度、显色时间和比色时间等检测条件进行优化。结果显示,用间苯二酚法测定3,6-内醚半乳糖的最佳检测条件为:测定波长554 nm、显色温度80°C、显色时间15 min;在该条件下,对琼脂及琼脂糖样品中的3,6-内醚半乳糖进行测定,方法的相对偏差为0.94%~1.27%,加标回收率为105.4%~111.04%。表明该方法操作简便,具有较高的准确性和重复性。 相似文献
