为研究At4g05060基因功能,采用PCR扩增拟南芥VAMP(vesicle-associated membrane protein)家族基因At4g05060的ORF序列,定向克隆至酵母双杂交诱饵表达载体pGBKT7,经酶切鉴定并测序后将其转化至酵母Y187细胞;然后采用Western blotting技术分析At4g05060基因在酵母中的表达水平,通过缺陷型培养基培养进行自激活检测。结果表明,成功构建酵母诱饵表达载体pGBKT7-At4g05060,并在酵母细胞正确表达,表达产物对报告基因无自激活作用。 相似文献

Ozone concentrations were measured at a wind-exposed edge of a 60 year-old 15–20 m tall Norway spruce forest in south-west Sweden and simultaneously over a barley field 5 km away for 27 days, At the forest site, measurements were performed at 3 and 13 m height 15 m in front of the forest edge, at 3 m height 15 m into the forest, and at 3 and 13 m height 45 m into the forest. Measurements at 3 m were made with three replicate tubes separated by 10 m. Differences between replicates were small. At 13 m height, the concentration (24-hr-average) 45 m into the forest was 95% of that in front of the forest edge. The average concentration at 3 m height did not vary strongly with the distance into the forest, but was 86% of that at 13 m in front of the forest edge. For AOT40 (Accumulated Exposure Over Threshold 40 ppb ozone), the differences between different positions were larger. At the 13 m level the AOT40 (day and night) was 88% of that in front of the forest 45 m into the forest. The AOT40 at 3 m was 71% of that at 13 m outside the forest. At the crop site, the ozone concentration at 1.1 m (0.1 m above the canopy), was 78% of that at 9 m (06.00–22.00). The AOT40 at 1.1 m above the ground, however, was only 50% of that at 9 m, indicating that serious errors can arise if ozone monitoring data are used uncorrnected in dose-response relationships based on measurements performed at plant height. The ozone concentration for the whole period differed very little between 9 m height at the crop site and 13 m height at the forest site outside the forest during daytime conditions (06.00–22.00). Night-time (22.00–06.00) values were only 21% at the crop site of those at the forest site due to the stronger night inversion development in the agricultural environment compared to the wind exposed forest edge. The results suggest that variations in topography and vegetation are important to consider when combining ozone monitoring data with dose-response functions. 相似文献

Summary Three random synthetic leaders and three naturally-occurring leaders, the tobacco mosaic virus (TMV) coat protein, the satellite tobacco necrosis virus (STNV) and the plant chlorophyll a/b-binding protein (Cab22L), were shown to modulate the -glucuronidase reporter protein accumulation levels in transient expression experiments. The same chimeric constructs also confer differential distribution patterns of reporter protein accumulation in stably-transformed tobacco calli or regenerated transgenic plants. When the highest expression levels with a given leader are compared, the 31-nucleotide random leader stimulates translation 20- and 100-fold relative to the 9- and 4- nucleotide synthetic leaders respectively. However, this 31-nucleotide random leader is approx. 2 to 3-fold weaker than the 30-nucleotide STNV leader and even 5-fold weaker than both the 79-nucleotide TMV leader and the 66-nucleotide Cab22L leader. These results confirm the findings in transient expression experiments and stress the importance of the 5-untranslated region for the production of heterologous proteins in transgenic plants. 相似文献

The nutritional effect of vitamin E in dietsfor Litopenaeus vannamei postlarve (PL19)was investigated. Four formulated diets withdifferent combinations of -tocopherylacetate (-TA), ascorbic acid (AA) andhighly unsaturated fatty acids (HUFA) weretested, using four replicates.No significant differences in survival wereobserved among treatments after 34 days offeeding. However, shrimp fed with a dietcontaining 2% fish oil (low n-3 HUFA content),200 mg.kg–1 -TA and100 mg.kg–1 AA (diet H/E/C) showedsignificantly better growth than those fed adiet supplemented with 5% fish oil (high n-3HUFA content), 200 mg.kg–1 -TA and100 mg.kg–1 AA (diet H+/E/C). Shrimp fedwith a diet containing 5% fish oil,900 mg.kg–1 -TA and100 mg.kg–1 AA (diet H+/E+/C) showed a significantly higher tissue level of n-6 PUFAthan postlarvae fed diet H+/E/C. No definiteconclusion could be drawn about a possibleinteraction between -TA and AA, since acomparison of the diet containing 5% fish oil,200 mg.kg–1 -TA and700 mg.kg–1 AA (H+/E+/C+) and the dietH+/E/C did not show any significant differencesin any of the measured parameters. Theantioxidative status of the shrimp tissue(measured by means of the thiobarbituric acid(TBA) assay and expressed as nM malonaldehyde(MA) per gramme dry weight) was equal for alltreatments. Nevertheless, there was a slightlylower MA value with the diet H+/E/C+,indicating that AA may be an effectiveantioxidant in the aqueous phase and at thewater/lipid interface of the tissue.
