AIM: To evaluate the effects of 1α, 25-dihydroxyvitamin D3 on T helper cell 17 (Th17 cells) and its related cytokines in a mouse model of corneal allograft transplantation. METHODS: C57BL/6 mice were transplanted with corneal grafts from BALB/c mice and treated intraperitoneally with 1.0 μg 1α, 25-dihydroxyvitamin D3 or soybean oil every other day after operation. The transparency of the corneal grafts was evaluated for potential rejection signs by slit lamp biomicroscopy and histopathology. The expression levels of IL-17, RORγt and IFN-γ in the spleen were measured by real-time PCR. Moreover, the protein expression of RORγt and IL-17 in the peripheral blood was analyzed by Western blotting. IL-17 and IFN-γ in peripheral blood were measured by flow cytometry. RESULTS: 1α, 25-dihydroxyvitamin D3 significantly inhibited the rejection of the corneal allograft and reduced the numbers of inflammatory infiltrates in the corneal graft. In the spleen, 1α, 25-dihydroxyvitamin D3 treatment reduced the expression levels of IL-17, RORγt and IFN-γ. In the peripheral blood, 1α, 25-dihydroxyvitamin D3 treatment downregulated the expression levels of RORγt, IL-17 and IFN-γ. CONCLUSION: The effects of 1α, 25-dihydroxyvitamin D3 on suppressing corneal transplantation-induced allograft rejection in mice are closely associated with its modulation on IL-17 and related cytokine RORγt. 相似文献

渤中25-1油田原油含蜡量高,C平台油井结蜡严重,导致清蜡作业频繁。为此,研发了一种非离子表面活性剂型水基清防蜡剂YE-60,首先通过合成反应获得YE型醇醚,合成条件:乙醇、烷基聚氧乙烯聚氧丙烯醚、酰胺基聚氧乙烯聚氧丙烯多嵌段醚摩尔比1:1.25:1.25,催化剂加入量相对单体总量的质量分数1.98%,反应温度75℃,反应时间3 h;然后将YE型醇醚与渗透剂乙二醇乙醚按照一定的质量比进行复配,得到最终产品YE-60。在100 mg/L加剂量下,YE-60对渤中25-1油田综合油样的防蜡率达到74.5%,洗蜡率达到64.1%。现场应用后,检泵和清蜡周期延长3倍以上,证明了其显著的清防蜡效果,具有较高的推广应用价值。(表6,参7) 相似文献

试验旨在探讨绞股蓝内生真菌JY25胞外多糖(JY25P)对蛋雏鸡生长性能和肠黏膜免疫功能的影响。将300羽健康的1日龄三黄蛋雏鸡,随机分为5组(每组设3个重复,每个重复20羽):Ⅰ组为灌服无菌水的空白对照组;Ⅱ组、Ⅲ组和Ⅳ组分别为灌服0.04%、0.08%和0.12%JY25P的试验组;Ⅴ组为灌服0.08%黄芪多糖的阳性对照组。结果表明:1)与Ⅰ组相比,Ⅳ组蛋雏鸡平均日增重显著提高了15.86%(P0.05),料重比极显著降低了24.13%(P0.01)。2)与Ⅰ组相比,试验各阶段,Ⅲ组、Ⅳ组、Ⅴ组均能显著提高蛋雏鸡血清中新城疫抗体效价(P0.05)。3)与Ⅰ组相比,在15日龄和20日龄,Ⅳ组可以极显著增加肠黏膜白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)含量(P0.01),而对分泌型免疫球蛋白A(SIg A)和IFN-γ/IL-4的影响不显著(P0.05)。结果提示:灌服0.12%JY25P能改善蛋雏鸡的生长性能,提高特异性新城疫抗体效价,增强非特异性肠黏膜IL-4和IFN-γ的分泌,同时维持机体的免疫平衡,因此,该多糖具有较好的应用前景。 相似文献

AIM: To investigate the regulatory effects of Rho-associated coiled-coil-containing protein kinase 2(Rock2) on the cell cycle checkpoint cell division cycle 25A(Cdc25A). METHODS: The protein expression levels of Rock2 and Cdc25A in 51 pairs of hepatocellular carcinoma and the adjacent tissues were detected by Western blotting. shRock2 plasmids were constructed, selected and stably transfected into hepatocellular carcinoma Huh-7 and HepG2 cells. The protein expression of Cdc25A in the cells was determined by Western blotting. Based on the Rock2 interfering sequences, we designed the primers and changed the 4 indicated bases via site-specific mutagenesis. The Rock2-mutant plasmid was verified by sequencing and was transfected into stable Rock2-knockdown cells. The protein expression of Cdc25A was detected by Western blotting, and the cell proliferation was measured by MTT assay. The protein levels of checkpoint kinase(Chk)1/Chk2 were also detected in stable Rock2-knockdown cells. The interaction between Rock2 and Cdc25A was measured by co-immunoprecipitation, and the co-localization of Rock2 and Cdc25A was detected by confocal laser scanning microscopy. RESULTS: Rock2 and Cdc25A were apparently up-regulated in hepatocellular carcinoma,with a significantly positive correlation. The protein expression of Cdc25A was significantly down-regulated in stable Rock2-knockdown cells. The expression of Chk1 and Chk2 was not changed following knockdown of Rock2. The co-immunoprecipitation results verified that Rock2 bound to Cdc25A. The results of confocal laser scanning microscopy showed that Rock2 and Cdc25A were co-localized in hepatocellular carcinoma cells. CONCLUSION: Rock2 positively regulates the cell cycle checkpoint Cdc25A, which is independent of Chk1/Chk2 and this may provide a new target gene for treatment of hepatocellular carcinoma. 相似文献

为提高我国大面积应用的两系光温敏不育系培矮64S的稻瘟抗性,本研究通过杂交转育与分子标记选择相结合的途径,利用RM3330与RM262为标记,将Pi25与Pi-d(t)基因成功聚合于培矮64S中,经抗性、不育性、综合农艺性状评价,获得了5个基因型纯合且农艺性状稳定的F8代抗性改良候选不育系。抗性鉴定结果显示,抗性改良候选不育系及其抗性组合的叶瘟病情指数平均降幅为18.78%和23.24%,穗颈瘟病情指数平均降幅为18.58%和58.45%,抗性提高幅度极显著。育性观测结果表明抗性改良候选不育系不育度都在99.5%以上,柱头外露降低5.64%,包颈粒率降低8.55%。经济性状调查结果显示不育系除穗粒数下降、有效穗增加外,其余经济性状变化不明显;试配的杂交组合平均理论产量和实际产量比对照提高16.22%和6.89%,增产极显著。这些结果表明,通过Pi25与Pi-d(t)基因的导入显著提高了培矮64S稻瘟病抗性,获得了实用的光温敏不育系新材料。 相似文献

在SD大鼠成骨细胞(Osteoblast,OB)体外培养体系中添加不同浓度1α,25-二羟维生素D3(0、10^-9、10^-8、10^-7mol/L),作用24、48、72h,测定OB增殖率、碱性磷酸酶(ALP)活性,作用48h流式细胞仪测定0B周期。结果显示,10^-9mol/L 1α,25-二羟维生素D3作用24、48、72h均促进oB增殖(P〈0.05或P〈0.01),抑制ALP活性(P〈0.01);10^-8、10^-7mol/L作用24、48h,OB增殖率与对照组差异不显著(P〉0.05),但24h时ALP活性均明显升高(P〈0.05或P〈0.01),48h则抑制了ALP活性并使OB滞留在G2/M期(P〈0.05或P〈0.01);72h时10^-7mol/L组OB增殖率极显著低于其余各组(P〈0.01),并使ALP活性升高(P〈0.01)。表明低浓度1α,25-二羟维生素D3能促进OB增殖,抑制其分化;高浓度1α,25-二羟维生素D3能抑制OB增殖,促进其分化,并使细胞滞留在G2/M期。 相似文献
