Abstract— Class II+ dendritic cells were widely distributed throughout normal ovine skin in two main locations: a) in or immediately adjacent to the epidermis and epidermal appendages and b) in the vicinity of the blood vessels. They are unlikely to represent a homogeneous population particularly since Langerhans cells, which previously have been found throughout the epidermal appendages, were located only in the epidermis using acetylcholinesterase staining. Following infection with orf virus, a dense mass of closely associated class II+ dendritic cells develops in the exposed necrotising dermis, adjacent to infected hair follicles and under infected degenerating epidermis. These cells interact and appear to form a barrier to invasion, a framework for immune defence and a template for subsequent epidermal repair; they seem to provide the basis of a highly integrated local dermal defence system. Résumé— Des cellules dendritiques de classe II+étaient largement réparties dans la peau ovine normale dans deux principales zones a) dans l'épiderme et les annexes épidermiques ou dans leur voisnage immédiat b) au voisinage des vaisseaux sanguins. Il est peu probable qu'elles représentant une population homogène particulièrement parce que les cellules de Langerhaps, qui ont été découvertes précédemment dans l'ensemble des annexes épidermiques, furent localisées seulement dans l'épiderme en utilisant une coloration à l'acétylcholinesterase. Après une infection par le virus de l'ecthyma, il se forme une masse dense de cellules dendritiques de classe II+étroitement associées, dans le derme nécrotique atteint, adjacente aux follicules pileux infectés et sous l'épiderme dégénératif infecté. Ces cellules interagissent et apparaissent former une barrière à l'invasion, un cadre pour les défenses immunitaires et un patron pour la réparation épidermique ultérieure; elles semblent fournir les bases d'un système de défense dermique local hautement intégré. Zusammenfassung— Klasse II+-Dendritenzellen waren in der gesamten Haut von normalen Schafen in vorwiegend zwei Bereichen verbreitet: a) in oder unmittelbar neben der Epidermis und der epidermalen Anhangsgebiete und b) in dor Nähe dar Blutgefäße. Sie stellen wahrscheinlich keine homogène Population dar, da die Langerhanszellen, die früher übarall in den epidermalen Anhangsgebilden nachgewiesen wurden, bei Acetylcholinesterase-Färbung nur in der Epidermis zu finden waren. Nach einer Orf-Virus-Infektion formiert sich eine dichte Masse aus eng verbundenen Klasse II+-Dendritenzellen in der betroffenen nekrotisierenden Dermis unmittelbar neben den infizierten Haarfollikeln und unter der infizierten, degenerierenden Epidermis. Diese Zellen stehen untereinander in Verbindung und bilden anscheinend eine Barrière gegen die Invasion, ein Gorüst für die Immunabwehr und einen Ausgangs punkt für die anschließenden Reparaturvorgänge in der Epidermis. Sie scheinen als Basis eines hochentwickelten lokalen Abwehrsystems der Haut zu fungieren. Resumen Células dendríticas de class II+ se observaron en gran cantidad en la piel de la oveja especialmente en dos localizaciones: a) en la epidermis, en la dermis muy próxima a la epidermis y en anejos cutáneos y b) en las proximidades de los vasos sanguíneos. No parece tratarse de una población homogénea de células puesto que las células de Langerhans, que previamente se habían encontrado en los anejos epidérmicos, se encontraron únicamente en la epidermis utilizando técnicas de detección del acetilcol-inesterasa. Después de la infección con el virus del ectima contagioso ovino se observó una masa de células dendríticas de clase II, dispuestas de forma muy densa, en las proximidades de la dermis necrosada y de los folículos pilosos y de la epidermis infectada. Eatas células interaccionan entre si y parecen formar una barrera contra la invasión, una red inmunitaria de defensa y participar en la reparación de la epidermis; parece ser que esta población de células dendriticas son la base de un sistema de defensa dérmico local altamente integrado.  相似文献

The purpose of this study was to evaluate oxidative stress and neurotoxic potential of organophosphorus (OP) insecticide diazinon in the sentinel freshwater fish, Oreochromis niloticus. Antioxidant and acetylcholinesterase (AChE) enzyme activities and malondialdehyde (MDA) and protein levels were measured spectrophotometrically in gill, kidney, alimentary tract, and muscle tissues of fish treated with sub-lethal diazinon concentrations for 1, 7, 15, and 30 days. Dose-dependent inhibitions of AChE were observed in all the experimental fish. On the contrary of alimentary tract, MDA levels were elevated in kidney and muscle and gill was not affected. AChE and MDA levels intercorrelated in kidney and muscle tissues. Diazinon had increased superoxide dismutase (SOD) activities in all the tissues, while kidney was the most affected tissue. Tissue-specific alterations were observed on catalase (CAT) and glutathione peroxidase (GPx) activities; however, the activities were not changed in gill and muscle tissues for GPx and in gill, muscle, and kidney tissues for CAT. Protein levels decreased in kidney, muscle, and alimentary tract, while increased in gill and alimentary tract in 15 days. With respect to these results, diazinon has oxidative and neurotoxic potentials in O. niloticus. Observed changes with diazinon treatment were generally tissue-specific and dose-dependent.  相似文献

The aim of this work was to study the pharmacokinetic behaviour and the inhibitory effect on acetylcholinesterase and butyrylcholinesterase activities of chlorpyrifos in male and female cattle after pour-on administration. Determination of cholinesterase activity in plasma and erythrocyte was carried out according to Ellman kinetic method. The mean baseline activities were 9338.39 ± 1331.61 and 13220.69 ± 2274.18 to acetylcholinesterase and 624.65 ± 39.32 and 641.68 ± 88.08 IU/L to butyrylcholinesterase in females and males, respectively. Acetylcholinesterase was the predominant form of cholinesterase analyzed, with low levels of butyrylcholinesterase. The basal acetylcholinesterase activities of the bulls were significantly greater than those of cows. The inhibitory effect of topical chlorpyrifos administration was lower on butyrylcholinesterase than on acetylcholinesterase. Chlorpyrifos peak plasma concentration (male:10.920 ± 4.18; female:12.12 ± 3.88 μg/L) were reached at 11.92 ± 9.19 and 8.17 ± 7.67 h in male and female, respectively. The values of area under curve were 185.96 ± 168.45 and 278.89 ± 270.00 μg·h/L and mean residence time were 13.95 ± 8.10 and 14.90 ± 9.80 h in male and female, respectively.  相似文献

应用连续组织切片,采用免疫荧光染色的方法,使用3种抗体:蛋白基因产物9.5(PGP9.5)、一氧化氮合成酶(NOS)和乙酰胆碱酯酶(AChE),分别对新生羔羊瘤胃内的所有神经元、氮能和胆碱能神经元进行定位分析。结果:PGP9.5、NOS和AChE阳性反应广泛分布于神经元胞体和神经纤维中。阳性神经元胞体在黏膜下未发现,但成群出现在肌间神经节内。阳性神经元胞体数目PGP9.5最多,其次是胆碱能,氮能最少。阳性神经纤维成簇分布在神经节内和节间形成神经束,并且延伸到环纵肌内。阳性神经纤维密度PGP9.5最多,其次是氮能,胆碱能最少。另外,一些阳性神经纤维经常围绕在血管周围,甚至延伸入瘤胃上皮。PGP9.5、NOS和AChE神经元和纤维在新生羔羊瘤胃内有广泛的分布,该结果提示其分布模式可能与功能的适应直接相关。  相似文献

The objective of this study was to assess the influence of an asymptomatic experimental infection by Babesia bigemina on cholinesterase’s as markers of the inflammatory process and biomarkers of oxidative imbalance. For this purpose, eight naive animals were used, as follows: four as controls or uninfected; and four infected with an attenuated strain of B. bigemina. Blood samples were collected on days 0, 7 and 11 post-inoculation (PI). Parasitemia was determined by blood smear evaluation, showing that the infection by B. bigemina resulted in mean 0.725 and 0.025% on day 7 and 11 PI, respectively, as well as mild anemia. The activities of acetylcholinesterase, butyrylcholinesterase and catalase were lower, while levels of thiobarbituric acid reactive substances and superoxide dismutase activity were higher in infected animals, when compared with the control group. This attenuated strain of B. bigemina induced an oxidative stress condition, as well as it reduces the cholinesterasés activity in infected and asymptomatic cattle. Therefore, this decrease of cholinesterase in infection by B. bigemina purpose is to inhibit inflammation, for thereby increasing acetylcholine levels, potent anti-inflammatory molecules.  相似文献

We investigated the effect of Z-ligustilide (LIG) on scopolamine-induced memory impairment in ICR mice. LIG (2.5–40 mg/kg) or tacrine (10 mg/kg) was orally administrated for 26 days. Behavior was examined in the Morris water maze and Y-maze after scopolamine administration (2 mg/kg, i.p.). The central acetylcholinesterase (AChE) and choline acetyltransferase (ChAT) activities were assessed spectrophotometrically. LIG significantly improved spatial long-term memory and short-term memory impairment, inhibited AChE activity and increased ChAT activity. Moreover, LIG and tacrine showed the comparable efficacy in both neurobehavioral and cholinergic evaluation. These data suggest that LIG may alleviate memory deficits probably via enhancing cholinergic function.  相似文献

A new lupene triterpenetriol was isolated from the acetone extract of the aerial parts of Salvia sclareoides and characterised as (1beta,3beta)-lup-20(29)-ene-1,3,30-triol (1). In addition, nepetidin (2), nepeticin (3), lupendiol (4), (1beta,11alpha)-dihydroxy-lup-20(29)-en-3-one (5), ursolic acid (6), sumaresinolic acid (7) and hederagenin (8), were identified in this Salvia sp. To the best of our knowledge, the compounds 2 and 7 are new constituents in Salvia spp. The acetone, ethanol, butanol and water extracts of the plant were screened for the in vitro inhibitory activity of acetylcholinesterase (AChE) and butyrilcholinesterase (BChE), enzymes which play a role in the Alzheimer disease. All extracts inhibited acetylcholinesterase activity at 10 microg/ml, a remarkable activity since the standard drug rivastigmine does not inhibit acetylcholinesterase at the same concentration. Regarding the butyrilcholinesterase, the acetone extract at 1000 microg/ml was able to inhibit completely the enzyme activity and the butanol and ethanol extracts, at this concentration, produced a potent inhibition of BchE.  相似文献

The effects of the essential oil of Eucalyptus tereticornis (EOET), especially the effects of its constituents α- and β-pinene, were studied on rat trachea in vitro. In tracheal rings, EOET, α- or β-pinene potentiated the contractions induced by acetylcholine (ACh). Contractions induced by K+ (60 mM) were also potentiated by α- and β-pinene, but were reduced by EOET. Our findings show that EOET has myorelaxant effects on rat airways, but potentiates ACh-induced contractions. Monoterpenes α- and β-pinene are involved in its potentiating actions, but are not responsible for its myorelaxant effects. A putative inhibition of the acetylcholinesterase enzyme is involved.  相似文献

对感染黄绿绿僵菌后的马铃薯瓢虫幼虫体内几种酶进行测定,结果表明:黄绿绿僵菌对于马铃薯瓢虫幼虫体内的蛋白质含量、蛋白酶、谷胱甘肽-S-转移酶和乙酰胆碱酯酶的活性,都有不同程度的影响,其中对蛋白含量和谷胱甘肽-S-转移酶的影响较大。  相似文献
