Expression of Anti-Lipopolysaccharide Factor Isoform 3 in Chlamydomonas reinhardtii Showing High Antimicrobial Activity

Antimicrobial peptides are a class of proteins with antibacterial functions. In this study, the anti-lipopolysaccharide factor isoform 3 gene (ALFPm3), encoding an antimicrobial peptide from Penaeus monodon with a super activity was expressed in Chlamydomonas reinhardtii, which would develop a microalga strain that can be used for the antimicrobial peptide production. To construct the expression cluster, namely pH2A-Pm3, the codon optimized ALFPm3 gene was fused with the ble reporter by 2A peptide and inserted into pH124 vector. The glass-bead method was performed to transform pH2A-Pm3 into C. reinhardtii CC-849. In addition to 8 μg/mL zeocin resistance selection, the C. reinhardtii transformants were further confirmed by genomic PCR and RT-PCR. Western blot analysis showed that the C. reinhardtii-derived ALFPm3 (cALFPm3) was successfully expressed in C. reinhardtii transformants and accounted for 0.35% of the total soluble protein (TSP). Furthermore, the results of antibacterial assay revealed that the cALFPm3 could significantly inhibit the growth of a variety of bacteria, including both Gram-negative bacteria and Gram-positive bacteria at a concentration of 0.77 μM. Especially, the inhibition could last longer than 24 h, which performed better than ampicillin. Hence, this study successfully developed a transgenic C. reinhardtii strain, which can produce the active ALFPm3 driven from P. monodon, providing a potential strategy to use C. reinhardtii as the cell factory to produce antimicrobial peptides.

赤霉素及机械处理的相互作用对破除Sandersonia aurantiaca种子休眠的研究

在预备试验的基础上，选用了21种促进种子发芽的方法，结果显示，完整的s．aurantiaca种子对任何处理都无响应，砂纸磨擦 近胚根处切除小部分可使种子发芽率提高至11％。以上处理结合300mg／L GA3溶液的使用可破除种子休眠，使种子发芽率提高到69％以上。结果还表明，S．aurantiaca种子发芽的适宜温度为20℃；25℃ 16h／d光照及30℃的条件可降低种子的发芽率。研究还对S．aurantiaca种子的吸胀能力以及所含抑制物对生菜种子发芽的影响进行了比较。初步得出，S．aurantiaca种子具有种皮限制和种胚休眠的双重休眠机制，GA3和机械处理的相互作用可破除种子休眠。

该文探讨了不用编制削度表直接建立高精度立木干曲线的方法．当采用干曲线是３次多项式时，通常根据１０分法测定树干各部位直径，用最小二乘法求出该方程式的参数．从数学角度，只要知道树干任意３个部位的直径，就可以用最小二乘法或联立方程式求解干曲线参数．本文以樟子松为例，探讨用树干哪３个部位直径拟合的干曲线最接近实际干曲线，为建立立木干曲线提供有效方法．利用９个部位半径（方法Ⅰ）和利用其中３个部位半径（方法Ⅱ，２８种组合）分别拟合干曲线，结果表明方法Ⅱ的６种半径组合的精度良好，其中的３种组合，（ｒ１．３，ｒ０．３，ｒ０．７），（ｒ１．３，ｒ０．４，ｒ０．７）和（ｒ１．３，ｒ０．４，ｒ０．８）是拟合现实干曲线的有效方法．     

立筒仓磷化氢环流熏蒸试验

文章主要探讨了生产中利用磷化氢环流熏蒸技术对立筒库群进行熏蒸的方法和杀虫效果。试验结果表明，该项技术对于立筒仓这类深粮层仓型熏蒸杀虫有很好的效果。对仓内的主要害虫有较好的杀死作用；但由于仓房气密性、环流方法和设备性能等因素的影响，操作不当就不能彻底杀死某些局部虫害或不久又再次发生害虫危害，影响整体熏蒸效果。     

Dynamics ofRhizoctonia solani (black scurf) in successive potato crops

The position of plants withRhizoctonia solani sclerotia (black scurf) on progeny tubers was mapped for an experimental field at Haren where potatoes were grown continuously and in rotation with other crops for five successive years, and for another field at Borgercompagnie with a 12 frequency of potatoes during three potato crops. Initially, the distribution of plants with black scurf on both fields was rather dense and homogeneous. In the following years the distribution became heterogeneous and patchy. The local decline ofR. solani AG 3 (the common potato pathogen) in Haren was apparently caused by an unknown factor selectively suppressingR. solani AG 3, while simultaneouslyR. solani AG 5 increased in mass. This AG 5 type proved to be an inferior competitor of AG 3 on the potato plant in a laboratory experiment. The specificR. solani antagonistVerticillium biguttatum did not play a role. A similar factor could have reduced the formation of black scurf in the experimental field at Borgercompagnie, whereV. biguttatum was also too infrequent to account for the decline.R. solani AG 5 was not present here and could not indicate the presence of a selective factor against AG 3.     

香蕉ACC氧化酶基因(MAO3)的克隆及其表达特性分析

 根据同源扩增得到香蕉(Musa acuminata) ACC氧化酶基因(MAO3) 的核心部分, 再通过3'和5'RACE扩增上、下游序列以及Genome-Walker的方法得到启动子部分, 共获得3 718 bp长度的序列。将所得结果进行聚类分析, 发现香蕉中ACC氧化酶的氨基酸序列非常保守, 各序列间的同一性高达99% ,与单子叶植物和双子叶植物中ACC氧化酶的氨基酸序列的同源性在66.7%～71.8%之间。组织原位杂交试验表明MAO3基因的表达具有组织特异性, 初步认为是在韧皮部筛管组织中特异表达。运用实时荧光定量PCR技术, 实时监测到了MAO3基因和香蕉乙烯受体基因ERS2受机械伤诱导的定量变化。     

辣椒新品种镇江3号的选育

中晚熟杂交一代辣椒镇研3号是以江苏本地羊角椒自交系Y9007为母本,由甘肃酒泉引进的甜椒自交系T9218为父本配制的一代杂交组合。镇研3号为中熟辣椒品种,果形为牛角形,纵径15cm,肩横径5.0cm、肉厚0.40cm,单果平均重42g。果面光滑,青熟果绿色、味微辣,风味佳,适宜在长江流域或南方保护地栽培种植,667m2产量约为4200～5000kg。
