AIM: To explore whether angiotensin Ⅱ type 2 receptor antagonist EMA401 decreases neuropathic pain and the expression of growth-associated protein-43 (GAP-43), protein kinase C (PKC) and calmodulin (CaM) in dorsal root ganglia (DRG) during chronic constriction injury (CCI) in rats. METHODS: SD rats were used to establish CCI model and randomly divided into 4 groups. The rats in model group were given equal volume of normal saline by intragastric administration. The rats in low dose (LD) group were given 5 mg/kg EMA401 by intragastric administration. The rats in middle dose (MD) group were given 10 mg/kg EMA401 by intragastric administration. The rats high dose (HD) group were given 20 mg/kg EMA401 by intragastric administration. The rats in sham operation group received equal volume of normal saline by intragastric administration. Thermal withdrawal latency (TWL) and mechanical withdrawal threshold (MWT) were measured before operation and 7 d, 14 d and 28 d after CCI. After behavioral test, DRG of lumbar spinal was obtained from each group, and was used to determine Ca2+ concentration by o-cresolphthalein complexone microplating method, and the expression of GAP-43, PKC and CaM at mRNA and protein levels by Western blotting and RT-PCR. RESULTS: Compared with model group, EMA401 significantly increased the TWL and MWT (P<0.05). Meanwhile, EMA401 significantly reduced Ca2+ concentration and the expression of GAP-43, PKC and CaM at mRNA and protein levels in the DRG (P<0.05). CONCLUSION: EMA401 may attenuate neuropathic pain of CCI by inhibiting Ca2+ concentration and the expression of GAP-43, PKC and CaM.  相似文献

利用农杆菌介导法,将半夏凝集素(pta)基因和高赖氨酸蛋白(sb401)基因导入水稻中,经潮霉素抗性筛选,得到70株独立再生植株.通过PCR检测,从中筛选出18株转基因植株.对这些转基因植株当代进行Southern blotting分析表明:pta 基因和sb401基因已经共转化并整和到受体基因组中.对转基因植株后代的叶片进行RT-PCR分析表明:pta基因在转录水平上得到了有效的表达.测定10株T0转基因植株成熟种子的赖氨酸和总蛋白质量分数表明,与对照相比,赖氨酸和总蛋白的提高率分别为8.48%~35.34%和4.55%~32.74%,表明sb401基因在大部分转基因植株成熟种子中的蛋白质水平上得到了比较有效的表达.  相似文献

津优401是以优良DH系J05-4和自交系21-4-1配制而成的黄瓜一代杂种。植株长势较强,叶片中等大小,叶色绿。主蔓结瓜为主,持续结瓜能力强。瓜条长35cm左右,瓜把长约为瓜长的1/8,单瓜质量200g左右,瓜条深绿色,有光泽,刺瘤中等,商品瓜率高。抗霜霉病、白粉病、枯萎病、病毒病等病害。春露地栽培每667m2产量可达5500kg以上,适合河北、河南、四川、东北等露地及秋大棚栽培。  相似文献

观察分析了中山杉401无性系(Taxodium ascendens Brongn×Taxodium mueyonatum Tenore CL'401')的染色体核型,并与亲本的染色体核型进行了比较。结果表明,中山杉401无性系的染色体数目为2n=22,核型公式为K(2n)=20m+2sm。杂种后代有两条大小、形态不同的近中部着丝点染色体,一条为第11条,一条为第17条,仅第17条具大着丝点的特征。中山杉401无性系确系种间杂种,在该无性系中未发生染色体加倍、缺失的变异。  相似文献

Summary Little work has been reported on genetic transformation with maize inbred lines, especially elite inbred lines used in breeding. In this work, 7 self-pollinated inbred lines and 4 hybrid lines have been screened. The results revealed that calli derived from immature embryos from two inbred lines X333 and X301 were compact, hyperhydric and unsuitable for transformation, but the calli induced from other inbred lines and all the hybrid lines were friable and yellow and could be used for genetic transformation. The sb401 gene isolated from potato (Solanum berthaultii) encodes a protein with a high lysine content. Maize calli from 5 self-pollinated inbred lines and 4 hybrid lines were transformed using particle-bombardment with different plasmids to simultaneously introduce the sb401 lysine rich gene and the selectable gene hpt respectively. Two hundred and sixty-eight regenerated plants were obtained from these genotypes. Co-insertion was confirmed in 29 regenerated plants by PCR and Southern blot analysis. Transgene segregation of the R1 plants was observed and one marker-free transgenic maize line was recovered. Analysis of the crude protein content in mature seeds of R1 transgenic plants also showed an increase from 36.8% to 48.2%. This study thus provides a workable system for generating transgenic maize free from selectable marker genes and generates valuable resources for obtaining marker free transgenic maize with a high-lysine protein content.  相似文献
