Low molecular mass organic acids (LMMOAs) and hydroxamate siderophores (HS) are molecules secreted by microbes and have previously been found in soil solution and in cultures. Mycorrhizal fungi are suggested to be involved in the nutrient uptake processes of trees and weathering of minerals. In this study soil samples taken from the O and E horizons of a podzol were extracted with 10 mM potassium phosphate buffer at pH 7.2. Variable parameters included addition of methanol to the extraction buffer and the use of ultrasonication or rotary shaking during extraction. LMMOAs and HS content of the soil extracts were determined. Analysis of soil extracts were carried out by liquid chromatography mass spectrometry (LC–MS) and the extraction results compared to results for soil solution samples obtained by centrifugation of the soils sampled. The extraction yields were significantly increased by addition of methanol to the extraction buffer, especially for the O horizon samples. Rotary shaking of the samples for 90 min gave slightly higher yields than ultrasonication for 15 min but the reduction in extraction time makes ultrasonication an attractive option. Of the HSs determined, ferricrocin was found in all samples. Optimal extraction conditions showed citric acid and isocitric acid to be the most abundant organic acids in the O and E horizons, respectively.  相似文献

In this study, a strain of Anabaena flos-aquae UTEX 1444 was cultivated in six different concentrations of iron (III). Cultures were extracted with organic solvents and analyzed using our dereplication strategy, based on the combined use of high-resolution tandem mass spectrometry and molecular networking. The analysis showed the presence of the siderophores’ family, named synechobactins, only in the zero iron (III) treatment culture. Seven unknown synechobactin variants were present in the extract, and their structures have been determined by a careful HRMS/MS analysis. This study unveils the capability of Anabaena flos-aquae UTEX 1444 to produce a large array of siderophores and may be a suitable model organism for a sustainable scale-up exploitation of such bioactive molecules, for the bioremediation of contaminated ecosystems, as well as in drug discovery.  相似文献

In literature, it remains little explored the soil–plant relationships within Capsicum agroecosystem. We studied how chili peppers plants contribute to influence microbial diversity. Across the bulk and rhizosphere soils of three genotypes of Capsicum annuum, the structure, the diversity and the abundance of bacteria was evaluated by means of DNA-based culture-independent approach. Furthermore, 515 bacterial strains isolated from the bulk and rhizosphere soil, were used to investigate the effect of C. annuum on four plant growth promoting bacteria (PGPB) abilities. Our results indicated that the three genotypes influence differently the physical-chemical and microbial properties of soil around the roots. Bacterial abundance resulted in increasing with different trend rhizospheres to bulk soil ratio; however, bacterial diversity was significantly higher only in the rhizosphere of one genotype. Only the indolic compounds production was stimulated in the rhizosphere of the three cultivars. Inhibition of Fusarium oxysporum was stimulated just with one genotype, where 53 of rhizosphere isolates showed more than 10% of inhibition. 165 of isolates produced siderophores and the major part belonged to the high production level. Interactions between PGPB features revealed that anti-phytopathogenic activity was not associated with the others characteristics; however, phosphate solubilization was associated with both siderophores and indolic compounds productions.  相似文献

A Pseudomonas sp. isolate MSB1 efficiently inhibited the growth of Flavobacterium psychrophilum of different serotypes on agar medium. A significant difference in the inhibition was observed between isolates of the less virulent FpT serotype compared to the Fd and Th serotypes. In broth coculture experiments, a low number of cells of MSB1 inhibited and outcompeted the F. psychrophilum cells. Also cell‐free culture supernatant of MSB1 clearly repressed the growth of F. psychrophilum. A chromoazurol S assay suggested that MSB1 produced efficient siderophores, which most probably were responsible for the iron deficiency in the supernatant. The limited growth of F. psychrophilum in the supernatant was found to be partly because of the lack of available iron, but the results also indicated that some other mechanisms were probably involved in the observed inhibition. A potential use of MSB1 as a probiotic in rainbow trout aquaculture, especially in early life stages of the fish, is suggested, but future in vivo experiments needs to be carried out to verify this suggestion. This study also indicates a low iron acquisition efficiency of F. psychrophilum, compared to other examined bacterial fish pathogens.  相似文献

Bacteria belonging to the Vibrionaceae family are widespread in the marine environment. Today, 128 species of vibrios are known. Several of them are infamous for their pathogenicity or symbiotic relationships. Despite their ability to interact with eukaryotes, the vibrios are greatly underexplored for their ability to produce bioactive secondary metabolites and studies have been limited to only a few species. Most of the compounds isolated from vibrios so far are non-ribosomal peptides or hybrids thereof, with examples of N-containing compounds produced independent of nonribosomal peptide synthetases (NRPS). Though covering a limited chemical space, vibrios produce compounds with attractive biological activities, including antibacterial, anticancer, and antivirulence activities. This review highlights some of the most interesting structures from this group of bacteria. Many compounds found in vibrios have also been isolated from other distantly related bacteria. This cosmopolitan occurrence of metabolites indicates a high incidence of horizontal gene transfer, which raises interesting questions concerning the ecological function of some of these molecules. This account underlines the pending potential for exploring new bacterial sources of bioactive compounds and the challenges related to their investigation.  相似文献

铁是所有生物正常发育所必需的微量元素之一,尤其是通过生物强化培育富含铁营养的农产品是解决人类铁营养“隐性饥饿”的重要技术途径,而螯合态铁肥由于见效快被广泛应用。因此,创制和研发新型微量元素螯合剂始终是国内外研究的竞争热点。麦根酸类植物铁载体和微生物铁载体等,能够高效螯合难溶铁并被植物高效吸收利用,是潜在的新型生物源螯合剂。该类新型铁肥在改善植物铁营养的同时不需要外源铁的投入,而是发挥菌株自身活性物质较强的螯合特性高效活化土壤中丰富的铁资源,为植物提供足够的生物有效铁。为进一步挖掘和研发新型生物源的绿色、高效且稳定的螯合剂,并为绿色智能肥料研发、实现绿色农业的可持续发展提供新途径和技术突破,本文基于近年来植物和微生物对铁营养吸收利用的分子生理机制的不断深入研究,从植物缺铁现状及诱因、改善铁营养的途径,到机理Ⅱ植物根系分泌物吸收利用铁的分子生态优势以及微生物铁载体改善植物铁营养的潜力,对生物源新型铁螯合剂研究进展及其应用进行了系统综述。期望通过进一步的研究和开发,能够更深入地了解这些新型生物源螯合剂的作用机制以提高植物的铁营养吸收效率,为实现绿色农业的可持续发展提供新的解决方案。  相似文献
