Loss of pathogenicity in the blue-stain fungus Ceratocystis polonica   总被引:1,自引:0,他引:1  
The pathogenicity and growth of six cultures of the bark-beetle-associated blue-stain fungus Ceratocystis polonica were compared. Four were subcultures, established at different times, of the same isolate (80–53/7), and at least one of these was suspected to have lost its pathogenicity to Norway spruce ( Picea abies ) during subculturing. Two other cultures (93–208/115 and 94–169/13) were pathogenic. The pathogenicity of all cultures to Norway spruce was compared in three inoculation experiments, using (i) massive inoculation (≈144 inoculations per tree), (ii) low-density inoculation (14 inoculations per tree) of larger trees and (iii) single inoculation of 2-year-old seedlings. Fungal growth rates were tested in vitro on malt agar, and on malt agar under oxygen-deficient conditions. Isolates 93–208/115 and 94–169/13 and one of the subcultures of isolate 80–53/7 were pathogenic in all three inoculation experiments. Two other subcultures were nonpathogenic, while the last subculture was intermediate. The pathogenic cultures had higher growth rates on malt agar than the nonpathogenic and intermediate cultures, and they also grew better under oxygen-deficient conditions. These results demonstrate that blue-stain fungi may lose their pathogenicity after serial vegetative transfers on artificial substrates. The results are also consistent with the hypothesis that rapid growth and the ability to grow in oxygen-poor environments are important pathogenicity factors in blue-stain fungi.  相似文献

Colletotrichum coccodes is currently being investigated as a mycoherbicide against the weed velvetleaf (Abutilon theophrasti). Two isolates ofPseudomonas spp. (Ps2 and Ps5) reduced the percentage of germ tubes and increased appressorial formation ofC. coccodes on detached leaves of velvetleaf. A study was conducted to see whether this effect could be attributed to competition for nutrients or iron betweenC. coccodes andPseudomonas spp. Ps2 and Ps5 had no effect on early spore germination, but reduced the percentage of germ tubes at 24 and 30 h, compared to the nontreated control. This reduction was diminished by the addition of nutrients but not Fe3+. Ps2 and Ps5 stimulated the formation of dark-coloured appressoria without germ tubes (AWGT), but this stimulation was diminished by the addition of nutrients or Fe3+. Germ tube branching at 30 h was also inhibited by the bacteria, but was not diminished by the addition of nutrients or iron. EDTA stimulated conidial germination at 10 h, which was reduced by the addition of Fe3+. However, EDTA did not stimulate the formation of appressoria (AWGT). These results suggest that the reduction in the percentage of germ tubes and the increase in the percentage of appressoria induced by the bacteria may be due to the competition for carbon or nitrogen. Iron competition may also be involved in the stimulation of appressorial formation, but not in the reduction in germ tube percentage and branching. Phylloplane bacteria may compete for carbon, nitrogen and iron, limiting the saprophytic phase of the pathogen on the phylloplane and accelerating the development of the parasitic phase. This may enhance the field efficacy ofC. coccodes as a biocontrol agent against velvetleaf.  相似文献

Two related iron chelates, one toxic to slugs by ingestion, the other not, were introduced into the foregut of D. reticulatum. The subsequent movement and redistribution of the metal within the slug tissues was studied by labelling the chelates with the radioactive isotope 55Fe. In slugs which survived treatment approximately half of the 55Fe was voided in faeces. The iron retained became unevenly distributed, the highest concentration occurring in the digestive gland, irrespective of the chelate used. At high doses, slugs treated with tris(1-oxo-1,2-diazabutan-2-oxido)Fe(III) were fatally poisoned while those treated with the homologue, tris(1-oxo-1,2-diazaoctan-2-oxido)Fe(III) were not. Slugs killed by the toxic chelate consistently contained proportionally less iron in the digestive gland and proportionally more in the body wall and reproductive system. Dosing slugs already killed by carbon dioxide asphyxiation gave a similar pattern, suggesting that the greater mobility of the iron from the toxic chelate was not a function of the slugs' metabolism.  相似文献

Pregnant sows were injected subcutaneously (s.c.) or intramuscularly (i.m.) with a barium selenate suspension (0.5–1.0 mg Se/kg body weight (b.w.)) and together with control animals fed a commercial diet. No response to the injection was seen either in blood selenium levels or in glutathione peroxidase (GSH-Px) activity in the sows. There was, however, a significant difference in these parameters between piglets born from treated dams and control animals. This status was maintained during the nursing period. In another experiment pigs (20 kg b.w.) on a Se-deficient diet were injected s.c. and i.m. with barium selenate (2.5 mg Se/kg b.w.). The treated groups maintained their blood levels of selenium and GSH-Px activity, although the selenium values in the group treated intramuscularly started to decline after 4 weeks. Organ samples from both groups were equal with regard to selenium at the time of slaughter while the control group showed a rapid decline both in blood selenium levels and GHS-Px activity.  相似文献

In order to study the effects of main technological pretreatments practised for preparing Egyptian faba bean products, i.e. decortication as well as soaking and germination followed by dehulling on the nutritional value, series of experiments were carried out. Such pretreatments had a significant effect on the changes in the chemical composition of faba beans. The proportion of the removed hulls reached generally about 14%. Data revealed also pronounced improvements on the nutritive value as a result of all studied pretreatments, especially germination being the most effective. Chemical scoring of all determined essential amino acids was >60, except methionine and cystine showed the lowest score (<20). Germinated seeds had the highest chemical score for the restricting amino acids beside the lowest GDR value [Grams consumed of product to cover the daily requirements for adult man in protein (63 g) and in energy (2900 kcal)]. All pretreatments caused a significant decrease in the antinutritional factors, especially soaking followed by dehulling, whereas decortication led to a significant increase in phytic acid content.  相似文献

试验选用360只健康1日龄艾维茵肉仔鸡(公、母混养),采用2×2×3三因子析因化设计,将360只试验雏鸡随机分成12组,每组设3个重复,每个重复10只鸡。锌的添加浓度为50、100mg/kg,铁的添加浓度为30、60mg/kg,维生素A的添加浓度为1500、3000、6000IU/kg。测定了肉仔鸡血清、肝脏谷胱甘肽过氧化物酶(GSH-Px)活性,过氧化氢酶(CAT)活性、铜锌超氧化物歧化(酶CuZn-SOD)活性,丙二醛(MDA)含量,以此来研究不同水平的锌、铁、维生素A同时添加,三者在肉仔鸡血清、肝脏抗氧化功能方面的互作效应。结果表明:锌、铁与维生素A三者互作对提高肉仔鸡血清CuZn-SOD活性影响极显(著P<0.01),对提高肉仔鸡肝脏GSH-Px活性和降低MDA含量影响极显(著P<0.01)。这说明锌、铁、维生素A三者之间在肉仔鸡抗氧化方面存在协同效应。  相似文献

为探讨VE在硒缺乏动物自由基代谢中的作用及添加VE对自由基代谢的影响,本试验采用低硒(低于10-9)、补VE(10-4)日粮饲喂小鼠,并对动物增重,血液、肝组织内丙二醛(MDA)、硒谷胱甘肽过氧化物酶(Se-GSH-Px)及自由基(FRs)水平进行了系统检测。试验发现,添加VE除可阻止缺硒所致的动物生长迟缓外,还可明显降低相关组织内MDA的含量,但它并不直接影响Se-GSH-Px的活力。试验结果表明,VE主要通过对机体脂质过氧化反应的抑制作用及其对自由基所致膜结构损伤的修复而参与机体自由基代谢,在硒缺乏状态下,它能与硒协同作用而影响病程发展和疾病转归。  相似文献

1日龄父母代罗曼(Lohmann)蛋鸡120只,随机分为3组,分别喂以低锰(基础日粮含锰13.62mg/kg)、低锰高磷(基础日粮+0.5%的磷)、补锰对照(基础日粮+锰80mg/kg)日粮。试验期8周。饲喂5~8周发病。低锰组发病率26%,低锰高磷组28%,补锰对照组无一发病。第4周时,体重分别为(267.9±8.32)、(214.2±10.42)、(265.0±12.18)g,低锰组、补锰对照组与低锰高磷组之间差异极显著(P<0.01)。肝锰含量分别为(5.40±0.74)、(2.82±0.36)、(8.48±0.96)mg/kg,补锰对照组与低锰组差异显著(P<0.05),与低锰高磷组差异极显著(P<0.01)。发病症状:精神沉郁,不愿走动,伏卧,跛行,单腿发病病腿向外侧伸展,双腿发病则内收呈“O”字形或外展呈“八”字形。剖检跗关节肿大,胫骨髁骨骨质增生、肿大,髁间沟变平坦。组织学检查,胫骨骺生长板增殖区变窄,软骨基质中粘多糖减少。超微结构变化,心肌纤维、肝细胞、胫骨骺生长板软骨细胞、睾丸精原细胞等线粒体肿胀与空泡化。  相似文献
