利用Getorf、SignalP 3.0、BLASTP等生物信息学软件和PERL语言从葡萄霜霉菌基因组中预测到一条编码RXLR-EER结构域的效应蛋白候选基因,将该基因编码的蛋白命名为RXLR5。将RXLR5信号肽SP5编码序列连接到pSUC2T7M13ORI载体并转化到酵母蔗糖酶缺陷菌株YTK12后,重组菌株能成功分泌蔗糖酶,促使棉籽糖分解成单糖,因此,能在YPRAA培养基上正常生长;同时生成的单糖能与TTC反应产生红色不溶于水的氯化三苯基四氮唑。由此推测,SP5具有信号肽活性,可能在RXLR5从葡萄霜霉菌细胞内泌到胞外的过程中起重要作用。 相似文献

Comprehensive information on the role of β‐casein and plasminogen–plasmin (PG–PL) system in milk secretion of Murrah buffaloes during winter season is lacking, although effects of cold stress can be ameliorated to an extent by altering microclimate at farm level. Hence, this study was aimed to determine the changes in productivity along with PG–PL system of milk, plasma hormones and metabolites of buffaloes during winter (December–January) season under two different management systems. Average minimum temperature and wind chill index during this season were 7.02 and 12.74 °C respectively. Buffaloes were divided in two groups of six animals each: control and treatment, where treatment group animals were placed in‐house with floor bedding of paddy straw and the control group animals in loose housing system without straw bedding. Physiological responses were recorded, and milk and blood samples were collected at weekly intervals for six‐week experimental period. Under in‐house management system, buffaloes experienced better comfort by alleviating environmental stress as their physiological responses such as respiration rate and pulse rate were significantly reduced (p < 0.01) as compared to the control, which subsequently resulted higher milk yield by 9.92% (p < 0.05). Analysis of milk samples revealed higher concentration of plasminogen (10.6 vs. 8.05 μg/ml; p < 0.01) and β‐casein (p < 0.05), and lower plasmin level (0.299 vs. 0.321 μg/ml; p < 0.05) in buffaloes under treatment group. It was also found that plasma cortisol, glucose and non‐esterified fatty acids levels were higher (p < 0.01) in control group as compared to the treatment animals by 13.6%, 8.14% and 12.6% respectively. However, milk composition, growth hormone, epinephrine and norepinephrine level in plasma were similar in both the groups. Hence, it may be concluded that provision of in‐house shelter management with floor bedding of paddy straw during winter was effective to minimize environmental stress and improved milk production through manipulation of PG–PL system in buffaloes. 相似文献

本文利用由中国蔓生刺黄瓜自然突变产生的矮生型黄瓜为材料,通过多低自交选育及性状观察,初步了解该矮生黄瓜植物学性状除具有自封顶、植株矮化性状外,与蔓生型黄瓜无明显差异。在生物学方面,该矮生黄瓜具有早熟、雌花节率高、结果集中、生长期短等特性。通过对中国矮生刺黄瓜与不同品种蔓生黄瓜杂交后代的观察、分析,初步认为黄瓜植株高度是由单基因体系和多基因体系共同作用决定的。 相似文献

There is a need to develop farming systems that enable both a satisfactory level of crop production and suitable environmental conditions for natural species. Wildlife‐friendly cropping techniques, such as a reduced amount of applied herbicide or a lower crop density, might be adopted in order to maintain populations of weed species of biological interest. An alternative might be to adopt an intensive cropping system in a part of the field and spare the other part as set‐aside or field margins, available for the natural development of plant species. The objective of this study was to present a method to compare two strategies for maintaining a desirable level of abundance of a given species of interest in agricultural areas, specifically (i) a strategy based on a wildlife‐friendly cropping system in a large cultivated area and (ii) a strategy based on a more intensive cropping system in a reduced area of cultivation, i.e. with land‐sparing. The principle is to calculate the ratio of crop production obtained with strategy (i) to the production obtained with strategy (ii) for a given target density of natural species. We show that the value of this ratio, and thus the relative performance of the two strategies, depends on the density of the weed species that can be maintained in an uncultivated ecological area. The method is applied in case studies of two plant species with contrasting ecology and conservation goals. The numerical results show that the strategy based on a wildlife‐friendly cropping system is more profitable in most situations. 相似文献

Our goal was to define a breeding objective for Brangus beef cattle in Brazil. Bioeconomic models were produced and used to estimate economic values (EVs). The scenarios simulated were typical full-cycle beef production systems that are used in tropical and subtropical regions. The breeding objective contained pregnancy rate (PR), warm carcass weight (WCW), mature cow weight (MCW), number of nematode eggs per gram of faeces (EPG) and tick count (TICK). Two models were used in series to estimate the EV. A deterministic model was used to simulate effects of PR, WCW and MCW on profitability with a constant parasite load. Subsequently, stochastic models were used to estimate economic values for TICK and EPG as consequences of their environmental effects on weight gains, mortality and health costs. The EV of PR, WCW, MCW, EPG and TICK, was US$1.59, US$2.11, −US$0.24, −US$5.35 and −US$20.88, respectively. Results indicate positive emphasis should be placed on PR (12.49%) and WCW (65.07%) with negative emphasis on MCW (13.92%), EPG (2.77%) and TICK (5.75%). In comparison with the indexes usually used, these results suggest a reformulation in the selection indexes of the beef production system in tropical and subtropical regions in order to obtain greater profitability. 相似文献
